Treadmill exercise in combination with acousto-optic and olfactory stimulation improves cognitive function in APP/PS1 mice through the brain-derived neurotrophic factor-and Cygb-associated signaling pathways

A reduction in adult neurogenesis is associated with behavioral abnormalities in patients with Alzheimer's ***,enhancing adult neurogenesis represents a promising therapeutic approach for mitigating disease symptoms and ***,nonpharmacological interventions aimed at inducing adult neurogenesis are currently *** individual non-pharmacological interventions,such as aerobic exercise,acousto-optic stimulation,and olfactory stimulation,have shown limited capacity to improve neurogenesis and cognitive function in patients with Alzheimer's disease,the therapeutic effect of a strategy that combines these interventions has not been fully *** this study,we observed an age-dependent decrease in adult neurogenesis and a concurrent increase in amyloid-beta accumulation in the hippocampus of amyloid precursor protein/presenilin 1 mice aged 2-8 *** deposition became evident at 4 months,while neurogenesis declined by 6 months,further deteriorating as the disease ***,following a 4-week multifactor stimulation protocol,which encompassed treadmill running(46 min/d,10 m/min,6 days per week),40 Hz acousto-optic stimulation(1 hour/day,6 days/week),and olfactory stimulation(1 hour/day,6 days/week),we found a significant increase in the number of newborn cells(5'-bromo-2'-deoxyuridine-positive cells),immature neurons(doublecortin-positive cells),newborn immature neurons(5'-bromo-2'-deoxyuridine-positive/doublecortin-positive cells),and newborn astrocytes(5'-bromo-2'-deoxyuridine-positive/glial fibrillary acidic protein-positive cells).Additionally,the amyloid-beta load in the hippocampus *** findings suggest that multifactor stimulation can enhance adult hippocampal neurogenesis and mitigate amyloid-beta neuropathology in amyloid precursor protein/presenilin 1 ***,cognitive abilities were improved,and depressive symptoms were alleviated in amyloid precursor protein/presenilin 1 mice

Rapid synthesis of Ti-MCM-41 by microwave-assisted hydrothermal method towards photocatalytic degradation of oxytetracycline

This study employed microwave-assisted hydrothermal method to synthesize Ti-MCM-41,which are mesoporous materials with a high surface area and excellent photocatalytic ability. Fourier transform infrared spectroscopy（FTIR）, X-ray diffraction（XRD）, X-ray photoelectron spectroscopy（XPS）, transmission electron microscopy（TEM）, and ultraviolet–visible spectroscopy（UV–Vis） were employed. The XRD findings showed that Ti-MCM-41 exhibited a peak at 2θ of 2.2°, which was attributed to the hexagonal MCM-41 structure. The BET（Brunauer–Emmett–Teller） results agreed with the TEM findings that Ti-MCM-41 has a pore size of about 3–5 nm and a high surface area of 883 m-2/g. FTIR results illustrated the existence of Si–O–Si and Si–O–Ti bonds in Ti-MCM-41. The appearance of Ti2 p peaks in the XPS results confirmed the FTIR findings that the Ti was successfully doped into the MCM-41 structure. Zeta（ζ）-potential results indicated that the iso-electric point（IEP） of Ti-MCM-41 was at about pH 3.02. In this study, the photocatalytic degradation of oxytetracycline（OTC） at different pH was investigated under Hg lamp irradiation（wavelength 365 nm）. The rate constant（K′obs） for OTC degradation was 0.012 min-1at pH 3. Furthermore, TOC（total organic carbon） and high resolution LC–MS（liquid chromatography–mass spectrometry） analyses were conducted to elucidate the possible intermediate products and degradation pathway for OTC. The TOC removal efficiency of OTC degradation was 87.0%, 74.4% and 50.9% at pH 3, 7 and 10, respectively. LC–MS analysis results showed that the degradation products from OTC resulted from the removal of functional groups from the OTC ring.

Probing Oxidant Effects on Superoxide Dismutase 1 OligomericStates in Live Cells Using Single-Molecule Fluorescence Anisotropy

The protein Cu/Zn superoxide dismutase (SOD1) isknown to function as a dimer, but its concentration in cells (∼50μM) and the dimerization constant (Kd of 500 μM) results suggestthat it exists in a monomer-dimer equilibrium. It is unclear how theoligomeric state of SOD1 changes when cells are initially exposed tohigh levels of extracellular oxidative stress. To address this problem,we introduced the single-molecule ffuorescence anisotropy (smFA)assay to explore SOD1 oligomeric states in live COS7 cells. smFAspeciffcally probes the ffuorescence polarization changes caused bymolecular rotations where the fast-rotating molecules (either due tosmaller hydrodynamic volume or less viscous environments)deteriorate the emission polarization and thus lower the anisotropy. After validating that smFA is effective in distinguishingmonomeric and dimeric ffuorescence proteins, we overexpressed SOD1 in live COS7 cells and investigated how its oligomeric statechanges under basal, 2 h, and 24 h 100 μM H_(2)O_(2) treatments. We found that treating cells with H_(2)O_(2) promotes SOD1 dimerizationand decreases cellular viscosity in 2 h. Interestingly, prolonged H_(2)O_(2) treatments show similar results as the basal conditions,indicating that cells return to a steady state similar to the basal state after 24 h, despite the presence of H_(2)O_(2). Our resultsdemonstrate that SOD1 changes its oligomeric state equilibrium in response to extracellular oxidative stresses. smFA will open newopportunities to explore the relationship between the SOD1 oligomer state and its H_(2)O_(2)-based signaling and transcription regulationroles.

Drought Response to Air Temperature Change over China on the Centennial Scale

Climate data from the Climatic Research Unit （CRU） for the period 1901-2013 are used to investigate the drought response to air temperature change over China on the centennial scale. Drought is observed to have increased evidently across China, except for some regions in eastern China. This increase is much stronger in northern China compared to southern China, especially in Northwest and North China. These change characteris- tics of drought are closely associated with air temperature change, with the severe droughts in the major drought episodes of the last century generally coinciding with higher temperatures. The significantly increasing trend of drought in China based on observations only appears when considering the effects of air temperature change, which can explain -49% of droughts in observations and 30%-65% of droughts in Coupled Model Intereomparison Project Phase 5 （CMIP5） model simulations. Furthermore, the response of drought to air temperature change generally increases as the drought time scale increases. Furthermore, drought shows relatively high sensitivity in spring and early summer in China on the centennial scale.

Sensitivity of Climate Changes to CO_2 Emissions in China

In this study, the authors demonstrate that the Coupled Model Intercomparison Project Phase 5 （CMIP5） models project a robust response in changes of mean and climate extremes to warming in China. Under a scenario of a 1% CO2 increase per year, surface temperature in China is projected to increase more rapidly than the global average, and the model ensemble projects more precipitation （2.2%/℃）. Responses in changes of climate extremes are generally much stronger than that of climate means. The majority of models project a consistent re- sponse, with more warm events but fewer cold events in China due to CO2 warming. For example, the ensemble mean indicates a high positive sensitivity for increasing summer days （12.4%/℃） and tropical nights （26.0%/℃）, but a negative sensitivity for decreasing frost days （-4.7%/℃） and ice days （-7.0%/℃）. Further analyses indicate that precipitation in China is likely to become more extreme, featuring a high positive sensitivity. The sensitivity is high （2.4%/℃） for heavy precipitation days （〉 10 mm d l） and increases dramatically （5.3%/℃） for very heavy precipitation days （〉 20 mm d-1）, as well as for precipitation amounts on very wet days （10.8%/℃） and extremely wet days （22.0%/℃）. Thus, it is concluded that the more extreme precipitation events generally show higher sensitivity to CO2 warming. Additionally, southern China is projected to experience an increased risk of drought and flood occurrence, while an increased risk of flood but a decreased risk of drought is likely in other regions of China.

Robustness of Precipitation Projections in China:Comparison between CMIP5 and CMIP3 Models

Three sources of uncertainty in model projections of precipitation change in China for the 21st century were separated and quantified: internal variability,inter-model variability,and scenario *** from models involved in the third phase and the fifth phase of the Coupled Model Intercomparison Project(CMIP3 and CMIP5) were compared to identify improvements in the robustness of projections from the latest generation of *** significant differences were found between CMIP3 and CMIP5 in terms of future precipitation projections over China,with the two datasets both showing future *** uncertainty can be attributed firstly to internal variability,and then to both inter-model and internal *** analysis revealed that the uncertainty in CMIP5 models has increased by about 10%–60% with respect to CMIP3,despite significant improvements in the latest generation of *** increase is mainly due to the increase of internal variability in the initial decades,and then mainly due to the increase of inter-model variability thereafter,especially by the end of this *** change in scenario uncertainty shows no major role,but makes a negative contribution to begin with,and then an increase later.

How Large Precipitation Changes over Global Monsoon Regions by CMIP5 Models?

Future changes in precipitation over global monsoon domains and their adjacent dry regions are investigated using present-day climate simulations(1986–2005)and future climate simulations under the Representative Concentration Pathways(RCP4.5)scenario by the Coupled Model Intercomparison Project Phase 5(CMIP5)*** the present-day climate simulations,high reproducibility of the extents of global monsoon domains and dry regions is observed from the multi-model ensemble(MME)result;the associated local summer precipitation variation and its interannual variability are also successfully *** the future,the global monsoon domains are projected to be expanded,while the dry regions are expected to initially increase and then *** summer precipitation and its variability show significant increases over most global monsoon domains and obvious decreases over their adjacent dry *** results indicate that currently wet regions will become wetter and dry areas will be dryer under global warming *** analysis indicates that changes in summer precipitation over global monsoon and dry regions can be interpreted as moisture convergence changes associated with changes in horizontal moisture transport.

The moving particle semi-implicit（MPS）method has demonstrated its usefulness in practical engineering *** it has wide applicability,it is still hard to predict the pressure precisely using the MPS method.A pressure-convection particle method based on the MPS method is proposed to overcome this *** improved performance of this new method is validated with computational and measured *** approach is also applied to compute the problem of sloshing associated with floating body motion in *** pressure-convection MPS method demonstrated its capability to improve the prediction of pressure.
