OBJECTIVES:Proteins are the major component and play a key role in nutritious and therapeutic functions of edible bird’s nest(EBN);however,limited studies have been conducted on the protein due to difficulties in extraction,isolation as well as *** study aimed to provide comprehensive information for the quality evaluation of EBN peptides,which would be a valuable reference for further study on EBN ***:Here,we developed a quality control method using high performance liquid chromatography(HPLC)peptide fingerprints deriving from EBN being digested with simulated gastric *** characteristic peptide peaks were collected and identified by LC-MS/***:The characteristic peptide peaks,corresponding to the protein fragments of acidic mammalian chitinase-like,lysyl oxidase,and Mucin-5AC-like,were identified and ***,the principal component analysis indicated that the fingerprints were able to discriminate colour of EBN(white/red)and production sites(cave/house)of White EBN on the same weight *** proposed by the model developed in this study,Muc-5AC-like and AMCaselike proteins were the markers with the highest discriminative ***:The overall findings suggest that HPLC peptide fingerprints were able to clearly demonstrate peptide profile differences between genuine and adulterated EBN samples;and classify EBN samples by its color and production *** addition,the protein identification results suggested that Muc-5AC-like protein was the major protein in EBN.

Flavonoids exhibit a wide range of biological activities including anti-inflammatory activity, while some 1,3-benzoxazine derivatives show anti-inflammatory activity. In order to explore the novel anti-inflammatory agents, based on the principles of bioisosterism and hybridization, ten novel ehromeno[8,7-e][1,3]oxazin-4-ones were syn- thesized and characterized with IR, ^1H NMR, MS and elemental analyses. The anti-inflammatory activities of the target compounds were evaluated via the croton oil-induced ear oedema test in Swiss mice. According to screened resuits, some target compounds show potent anti-inflammatory activity.

褐脉少花龙葵毛状根的诱导、培养及其澳洲茄胺的产生

利用发根农杆菌的遗传转化和液体培养技术,研究了褐脉少花龙葵(Solanum nigrum L. var. pauciflorum)毛状根的诱导和离体培养及其澳洲茄胺的产生以及液体培养过程中培养基中N源和钙的消耗变化。结果表明,发根农杆菌ATCC15834感染褐脉少花龙葵叶片外植体5 d后产生毛状根,感染25d后,约90%的叶片外植体产生毛状根。毛状根能在无外源生长调节剂的MS固体和液体培养基上自主生长。PCR扩增结果显示发根农杆菌Ri质粒的rolB和rolC基因已在少花龙葵毛状根基因组中整合并得到表达。所产生的毛状根能产生药用次生物质澳洲茄胺,其含量约为非转化植株根的1.3倍,达到582.05μg/g干重。少花龙葵毛状根液体培养0-5 d内处于生长迟滞期、5-15 d为快速生长期、15d后进入生长平台期。培养基的硝态氮和铵态氮在毛状根液体培养过程中被逐渐吸收和消耗,至培养15 d时铵态氮被消耗殆尽,而硝态氮仍剩余44.7%;培养基中钙的浓度在培养过程中虽逐渐降低,但在培养25d时仍未被完全消耗,其浓度约为起始浓度的43.5%。该结果为今后设计合适的培养基来规模培养褐脉少花龙葵毛状根生产药用次生物质澳洲茄胺提供了可能性。

Background Recent studies have demonstrated the existence of a small fraction of cells with features of primitive neural progenitor cells and tumor-initiating function in brain tumors. These cells might represent primary therapeutic target for complete eradication of the tumors. This study aimed to determine the resistant phenotype of glioblastoma stem cells （GSCs） to temozolomide （TMZ） and to explore the possible molecular mechanisms underlying TMZ resistance. Methods Freshly resected glioblastoma specimen was collected and magnetic isolation of GSCs was carried out using the Miltenyi Biotec CD133 Cell Isolation kit. The cytotoxic effect of TMZ on CD133^＋ and CD133^- glioblastoma cells was determined by using the 3-（4,5-dimethylthiazol-2-yl）-2,5-diphenyltetrazolium bromide （MTT） assay. Autophagy-related proteins （Beclin-1, LC3 and Atg5） and cleaved caspase-3 （p17） were analyzed by Western blotting. Immunofluorescent staining was used to detect Atg5, glial fibrillary acidic protein （GFAP） and CD133 expression in glioblastoma cells. Statistical analysis was carried out using SPSS 10.0 software. For all tests, the level of statistical significance was set at P 〈0.05. Results CD133^＋ glioblastoma ceils exhibited neurosphere-like growth in vitro and high expression of CD133 stem cell marker. The growth-inhibiting rate in CD133- glioblastoma cells treated with 5 or 50 pmol/L TMZ was significantly higher than that in CD133^＋ glioblastoma cells （（14.36±3.75）% vs （2.54±1.36）% or （25.95±5.25）% vs （2.72±1.84）%, respectively, P 〈0.05）. Atg5, LC3-11 and Beclin-1 levels were significantly lower in CD133^＋ glioblastoma cells than those in autologous CD133^- cells after TMZ treatment （P 〈0.05）. Caspase-3 was mildly activated only in CD133^- glioblastoma cells after exposure to TMZ （P 〈0.05）. Immunofluorescent staining revealed elevated expression of Atg5 in GFAP^＋ cells following TMZ treatment. Conclusions The GSCs display strong capability of tumor＇s resistance to TMZ. This resis

Background Asthma is a significant chronic health problem worldwide. Management aims at disease control by reducing functional impairment and exacerbations and improving quality of life (QoL). We report a multi-center study to survey asthma control and QoL in four cities in the Pearl River Delta. Methods The conjoint survey involved ten Hong Kong pediatric hospitals/units, two Shenzhen hospitals, two Macao hos-pitals, and two Guangzhou hospitals on asthma control (using Asthma Control Test) and QoL (Pediatric Allergic Disease Quality of Life Questionnaire, PADQLQ). Acceptability of a treatment is graded as very good/good/fair/poor. Results Good asthma control was only reported in 80% subjects in Hong Kong, but higher in sister cities (85–94%, P<0.001). Allergic rhinitis, 'incense burning', and 'smoker in family' were prevalent among the four cities. Logistic regres-sion showed better control of asthma was associated with better PADQLQ (B=?0.029, P < 0.001), better acceptability of bronchodilator (B=?1.488, P = 0.025), negatively with 'smoker in family' (B=?0.83, P = 0.015) and various PADQLQ domains. Conversely, worse PADQLQ was associated with allergic rhinitis severity (B=4.77, P <0.001), poor control of asthma (B=7.56, P <0.001), increased frequency of traditional Chinese medicine use (B=1.7, P < 0.05), increased fre-quency of bronchodilator usage (B=1.05, P < 0.05), 'smoker in family' (B=4.05, P < 0.05), and incense burning at home (B=3.9, P < 0.05). Conclusions There are some clinical and cultural differences among the four southern Chinese cities within the Guangdong province. This study identifies potentially modifiable environmental and treatment factors associated with poor asthma control and QoL for health-care interventions. Having a smoker in the family is independently associated with poor asthma control and QoL.

本文尝试建构一种理论社会学的综合视角,它由三个部分即基础存在论、主体主义式结构论和能动主义式特征化构成。本文的主要尝试是:(一)在结构论意义上的社会结构和象征全域与能动主义特征化方面的叙事和筹划之间构想一种理论联系,使其更适合于实证性的研究;(二)为了使叙事和筹划这两个概念在具体事例分析中更具可应用性,尝试对其在若干中国古典叙事分析上的适用性;(三)着重区分主体性和能动性两个概念,尝试使社会学分析中的"行动者"自觉于某种存在论蜕变,实现由主体性到能动性的转换。

合成了一种含4,5-二氮-9,9-螺二芴(sb)配体的三齿磷铱配合物Ir(tpit)(sb)Cl(tpitH2=亚磷酸三苯基酯),通过核磁共振氢谱和磷谱及高分辨质谱对其结构进行了确定。X射线单晶衍射分析表明,sb配体的存在扭曲了分子结构,有助于降低分子聚集及发光淬灭。与存在分子内π-π堆积的模型配合物Ir(tpit)(bpy)Cl(bpy=2,2′-联吡啶)对比进行了光电性能的研究。结果表明在聚甲基丙烯酸甲酯(质量分数1%)中配合物Ir(tpit)(sb)Cl的发光波长为512 nm,相对配合物Ir(tpit)(bpy)Cl的波长(520 nm)有了8 nm蓝移。配合物Ir(tpit)(sb)Cl的发光量子效率为30%,与配合物Ir(tpit)(bpy)Cl的94%相比有明显降低,说明了分子内π-π堆积作用在降低柔性基团非辐射跃迁率方面的重要作用。基于配合物Ir(tpit)(sb)Cl的有机电致发光器件,最大电流效率和外量子效率分别为14 cd·A-1和4.5%。而由于分子内π-π堆积作用,基于配合物Ir(tpit)(bpy)Cl器件的最大电流效率和外量子效率分别高达60 cd·A-1和18.2%。
