Endothelial METRNL determines circulating METRNL level and maintains endothelial function against atherosclerosis

METRNL is a recently identified secreted protein with emerging functions.This study is to find major cellular source of circulating METRNL and to determine METRNL novel function.Here,we show METRNL is abundant in human and mouse vascular endothelium and released by endothelial cells using endoplasmic reticulum-Golgi apparatus pathway.By creating endothelial cell-specific Metrnl knockout mice,combined with bone marrow transplantation to produce bone marrow-specific deletion of Metrnl,we demonstrate that most of circulating METRNL(approximately 75%)originates from the endothelial cells.Both endothelial and circulating METRNL decrease in atherosclerosis mice and patients.By generating endothelial cell-specific Metrnl knockout in apolipoprotein E-deficient mice,combined with bone marrow-specific deletion of Metrnl in apolipoprotein E-deficient mice,we further demonstrate that endothelial METRNL deficiency accelerates atherosclerosis.Mechanically,endothelial METRNL deficiency causes vascular endothelial dysfunction including vasodilation impairment via reducing eNOS phosphorylation at Ser1177 and inflammation activation via enhancing NFκB pathway,which promotes the susceptibility of atherosclerosis.Exogenous METRNL rescues METRNL deficiency induced endothelial dysfunction.These findings reveal that METRNL is a new endothelial substance not only determining the circulating METRNL level but also regulating endothelial function for vascular health and disease.METRNL is a therapeutic target against endothelial dysfunction and atherosclerosis.

Neuroscientists have recognized the importance of astrocytes in regulating neurological function and their influence on the release of glial transmitters. Few studies, however, have focused on the effects of general anesthetic agents on neuroglia or astrocytes. Astrocytes can also be an important target of general anesthetic agents as they exert not only sedative, analgesic, and amnesic effects but also mediate general anesthetic-induced neurotoxicity and postoperative cognitive dysfunction. Here, we analyzed recent advances in understanding the mechanism of general anesthetic agents on astrocytes, and found that exposure to general anesthetic agents will destroy the morphology and proliferation of astrocytes, in addition to acting on the receptors on their surface, which not only affect Ca2+signaling, inhibit the release of brain-derived neurotrophic factor and lactate from astrocytes, but are even involved in the regulation of the pro-and anti-inflammatory processes of astrocytes. These would obviously affect the communication between astrocytes as well as between astrocytes and neighboring neurons, other neuroglia, and vascular cells. In this review, we summarize how general anesthetic agents act on neurons via astrocytes, and explore potential mechanisms of action of general anesthetic agents on the nervous system. We hope that this review will provide a new direction for mitigating the neurotoxicity of general anesthetic agents.

Biogeochemistry of sedimentary organic matter in the Yongjiang River estuary in the southern part of Hangzhou Bay,China,since the Late Pleistocene

A sediment core(YJK19-02)collected from the southern outlet of Hangzhou Bay near the Yongjiang River estuary in East China was analyzed for grain size,lignin,bulk elemental composition,stable carbon isotope,and rare earth elements(REEs)to determine the sources and diagenesis of sedimentary organic matter(OM)of the estuary and adjacent areas since the Late Pleistocene.δ^(13)C values(-24.80‰–-23.60‰),total organic carbon/total nitrogen(TOC/TN)molar ratios(8.00–12.14),and light rare earth element/heavy rare earth element ratios(LREE/HREE=8.34–8.91)revealed the predominance of terrestrial sources of OM,mainly from the Changjiang(Yangtze)River.The lignin parameters of syringyl/vanillyl(S/V=0.20–0.73)and cinnamyl/vanillyl(C/V=0.03–0.19)ratios indicate the predominance of nonwoody angiosperms,and the vanillic acid/vanillin ratios[(Ad/Al)_(V)=0.32–1.57]indicate medium to high degrees of lignin degradation.An increasing trend ofΛ(total lignin in mg/100-mg OC)values from ca.14500 a BP to ca.11000 a BP reflected the increase in temperature during the Late Pleistocene.However,a time lag effect of temperature on vegetation abundance was also revealed.The relatively higher and stableΛvalues correspond to the higher temperature during the mid-Holocene from ca.8500 a BP to ca.4500 a BP.Λvalues decreased from ca.4000 a BP to the present,corresponding to historical temperature fluctuations during this time.Our results show that the vegetation abundance in the Yongjiang River Basin since the Late Pleistocene was related to the temperature fluctuation duo to climate change.

An Extrinsic Faradaic Layer on CuSn for High-Performance Electrocatalytic CO_(2) Reduction

An intrinsic Faradaic layer on the surface of a metal electrocatalyst is usually considered an active site for CO_(2) reduction.Different strategies have been used to improve the performance of CO_(2) reduction by adjusting the intrinsic Faradaic layer.However,it is still challenging to achieve CO_(2) reduction with high activity,selectivity,and stability.In this study,for the first time,we improve the three parameters simultaneously by introducing a Zn(OH)_(x) over layer onto a CuSn electrocatalyst.We find that the intrinsic Faradaic layer of Sn(OH)_(x) on the surface of CuSn provides active sites for CO_(2) reduction,while Zn(OH)_(x) plays multiple roles as an adsorption/activation layer,a cover layer,and a protective layer.Further studies suggest that the enhanced activity comes from a Faradaic reaction of Zn(OH)_(x) during CO_(2) reduction,which can be considered as an extrinsic Faradaic layer.This new strategy of introducing an extrinsic Faradaic layer can deepen understanding of electrocatalytic process and offers guidance to design other high-performance electrocatalysts.

The neutron-induced total cross sections of natural lead have been measured in a wide energy range(0.3 eV-20 MeV)on the back-streaming white neutron beamline(Back-n)at the China Spallation Neutron Source.Neutron energy was determined by the neutron total cross-section spectrometer using the time-of-flight technique.A fast multi-cell fission chamber was used as the neutron detector,and a 10-mm-thick high-purity natural lead sample was employed for the neutron transmission measurements.The on-beam background was determined using Co,In,Ag,and Cd filters.The excitation function of ^(nat)Pb(n,tot)reaction below 20 MeV was calculated using the TALYS-1.96 nuclear-reaction modeling program.The present results were compared with previous results,the evaluated data available in the five major evaluated nuclear data libraries(i.e.,ENDF/B-VIII.0,JEFF-3.3,JENDL-5,CENDL-3.2,and BROND-3.1),and the theoretical calculation curve.Good agreement was found between the new results and those of previous experiments and with the theoretical curves in the corresponding region.This measurement obtained the neutron total cross section of natural lead with good accuracy over a wide energy range and added experimental data in the resonance energy range.This provides more reliable experimental data for nuclear engineering design and nuclear data evaluation of lead.

Background Pediatric antineutrophil cytoplasmic antibody-associated vasculitis(AAV)is a life-threatening systemic vasculitis featured by liability to renal involvement.However,there are few studies on the risk factors and predictive models for renal outcomes of AAV in children.Methods Data from 179 AAV children in multiple centers between January 2012 and March 2020 were collected retrospectively.The risk factors and predictive model of end-stage renal disease(ESRD)in AAV were explored.Results Renal involvement was the most typical manifestation(95.5%),and the crescent was the predominant pathological lesion(84.9%).The estimated glomerular filtration rate(eGFR)was evaluated in 114 patients,of whom 59.6%developed ESRD,and the median time to ESRD was 3.20 months.The eGFR[P=0.006,odds ratio(OR)=0.955,95%confidence interval(CI)=0.924–0.987]and the percentages of global glomerulosclerosis(pGGS;P=0.018,OR=1.060,95%CI=1.010–1.112)were independent risk factors for ESRD of renal biopsy.Based on the pGGS and eGFR at renal biopsy,we developed three risk grades of ESRD and one predictive model.The Kaplan‒Meier curve indicated that renal outcomes were significantly different in different risk grades(P<0.001).Compared with serum creatinine at baseline,the predictive model had higher accuracy(0.86 versus 0.58,P<0.001)and a lower coefficient of variation(0.07 versus 0.92)in external validation.Conclusions Renal involvement is the most common manifestation of pediatric AAV in China,of which more than half deteriorates into ESRD.The predictive model based on eGFR at renal biopsy and the pGGS may be stable and accurate in speculating the risk of ESRD in AAV children.

Efficient purification of Al_(30) by organic complexation method

A method was developed for preparing high purity Al30O8(OH)56(H2O)_(24)^(18)+(Al30) through elimination of impurities by complexation.Polyaluminum chloride II(PAC_(30))with Al_(c )content of 75%was adopted as the source of Al_(30).The PAC_(30)was prepared under conditions of total aluminum concentration 0.1 mol/L and OH^-/Al ratio 2.2 to obtain the highest content of Al_(30).A precipitation/metathesis method,organic solvent precipitation method and organic complexation method were examined to separate and purify Al_(30).It was found that only by the organic complexation method could high purity Al_(30)products be obtained in large yield economically.In the experiments,benzoic acid was used as the coordinating reagent to decompose the main impurity AlO_4Al_(12)(OH)_(24)(H_2O)_(12)^(7+)(Al_(13)),and the Al_(30)product could be obtained by precipitation and metathesis operations.It was noteworthy that the decomposition of impurities by benzoic acid could be completed in 2 hr.The Al_(30)product was characterized by Ferron assay,^(27)Al-NMR,SEM,XRD and TGA.The results showed that the purity of the Al_(30)product could exceed 92%.

Graphene is widely used in photodetection due to its high carrier mobility and wide spectral absorption range.However, the high dark current due to its low light absorption severely limits the performance of photodetection. Molybdenum dihalide(MoX2, X= S, Se and Te) has a high absorption coefficient, which can compensate high dark current in graphene-based photodetectors and result in outstanding photoelectronic properties in graphene/MoX2 van der Waals heterostructure(vdWH) photodetectors.In this review, we firstly review working principles, performance indicators, and structures of photodetectors. After that, the significance of graphene/MoX2 vdWH photodetectors are highlighted from the material fundamental perspective. Preparation methodologies and performance enhancement strategies of graphene/MoX2 vdWH photodetectors are correspondingly summarized. In the end, we highlight the current challenges and future directions of the graphene/MoX2 vdWH photodetectors. This review will guide the design of high-performance vdWH photodetectors.

集成方法是不平衡学习方法的重要分支,然而,现有不平衡集成方法均作用于原样本而没考虑样本的结构信息,因此其效能仍然有限.样本的结构信息包括局部和全局结构信息.为了解决上述问题,本文提出了一种基于深度样本包络网络(Deep Instance Envelope Network,DIEN)和分级结构一致性机制(Hierarchical Structure Consistency Mechanism,HSCM)的不平衡集成学习算法.该算法在考虑局部流形和全局结构信息的情况下,通过多层样本聚类,生成高质量的多层包络样本,从而实现类平衡化.首先,算法基于样本近邻拼接和模糊C均值聚类算法,设计DIEN来挖掘样本的结构信息,得到深度包络样本.然后,设计局部流形结构度量和全局结构分布度量来构建HSCM用于增强层间样本的分布一致性.接着,将DIEN和HSCM结合起来,构建出优化后的深度样本包络网络——DH(DIEN with HSCM).之后,将基分类器应用于包络样本.最后,设计bagging集成学习机制来融合基分类器的预测结果.文末组织了多组实验,采用了十多个公共数据集和有代表性的相关算法进行验证比较.实验结果表明,本文算法在AUC(Area Under Curve),F-measure等四个性能指标上显著最优.
