Unbalanced biotransformation metabolism and oxidative stress status: implications for deficient fatty acid oxidation

The concept of accumulating xenobiotics within the human body as a health risk is well known. However, these compounds can also be endo-genous, as in the case of inborn errors of me-tabolism, and lead to some of the same symp-toms as seen in xenobiotic intoxication. Bio-transformation of both exogenous and endo-genous toxic compounds is an important function of the liver, and the critical balance between these systems is of fundamental importance for cellular health. We propose a novel model, to describe the critical balance between Phase I and Phase II biotransformation and how a disturbance in this balance will increase the oxidative stress status, with resulting pathological consequences. We further used deficient fatty acid oxidation to verify the proposed model, as deficient fatty acid oxidation is associated with the accumulation of characteristic metabolites. These accumulating metabolites undergo both Phase I and Phase II biotransformation reactions, with resulting depletion of biotransformation substrates and co-factors. Depletion of these important biomolecules is capable of disturbing the balance between Phase I and Phase II reactions, and disturbance of this balance will increase oxidative stress status. The value of the proposed model is illustrated by its application to a clinical case investigated in our laboratory. In this case the possibility of deficient fatty acid oxidation only became evident once the critical balance between Phase I and Phase II biotransformation was restored with oral replenishment of biotransformation substrates. In addition to bio-chemical improvement, there was also significant clinical improvement. The significance of this model lies within the treatment possibilities, as the assessment of biotransformation metabolism and oxidative stress status can lead to the development of nutritional treatment strategies to correct imbalances. This in turn may reduce the chances of, or delay the onset of certain disease states.

Recent advances in thermochemical storage technology have introduced a novel closed-loop thermal energy storage(TES)system,known as the heat battery(HB),which is believed to have great potential for aiding the energy transition in the built environment because of its higher energy density and neglectable storage loss compared to conventional TES *** order to investigate the potential use cases of the HB and provide practical feedback for its further development,this research employs a simulation-based approach to analyze its influence on building performance in various use cases within Dutch residential *** including the homeowner,distribution system operator,and district heating system operator are identified,and a preliminary list of use cases is defined based on relevant literature and input from the HB *** simulation approach is conducted to predict key performance indicators for each *** Kruskal-Wallis test was employed to sort and scrutinize the simulation outcomes and discern the significance of each use case *** findings demonstrated that the HB holds the potential to diminish both the operational energy cost by up to 30%for the homeowners and the peak heating load transmitted from the building to the district heating system.

Improvement on the Mechanical Performance and Resistance Towards Hydrolysis of Poly(glycolic acid)via Solid-state Drawing

Poly(glycolic acid)is a biocompatible as well as biocomposable polymer with superior mechanical and barrier properties and,consequently,has found important applications in both medical and packaging ***,the high hydrolysis rate in a high humidity environment restricts its *** this work,a solid-state drawing process after melt extrusion is applied in order to produce fibrous PGA with enhanced mechanical properties and a much better resistance towards *** crystal structure of PGA gradually transformed from spherulites into oriented fibrous crystals in the stretching direction upon solid-state ***,both the length of microfibril and the size of lamellae increased initially with the drawing ratio(DR),while the chain-folded lamellae transformed into extended-chain fibrils at high(er)*** oriented structures lead to an overall improvement of the mechanical properties of PGA,e.g.,the tensile strength increased from 62.0±1.4 MPa to 910±54 MPa and the elongation at break increased from around 7%to 50%.Meanwhile,the heat capacity of totally mobile amorphous PGA(∆C_(p)^(0)=0.64 j.g^(−1)·℃^(−1))was reported for the first time,which was used to analyze the content of mobile amorphous fraction(XMAF)and rigid amorphous fraction(XRAF).Both the oriented chain-folded lamellae crystals and the tightly arranged RAF are beneficial to prevent water molecules from penetrating the matrix,thus improving the resistance towards *** a consequence,the fibrous PGA with a DR of 5 showed a tensile strength retention rate of 17.3%higher in comparison with the undrawn sample after 7-days accelerated ***,this work provides a feasible method to improve the mechanical and resistance towards hydrolysis performance of PGA,which may broaden its application and prolong the shelf-life of PGA products.

The availability of probiotics and donor human milk is associated with improved survival in very preterm infants

Background To determine whether the introduction of pasteurized donor human milk and probiotics for infants born<32 weeks gestational age or<1500 g birthweight is associated with a reduction in mortality and the incidence of necrotising enterocolitis (NEC) and sepsis. Methods We performed a retrospective analysis of two cohorts: before and after the introduction of probiotics and pasteurised donor human milk. Univariate analysis of primary and secondary outcomes was performed;variables impacting outcomes were assessed using multivariate logistic regression. Results There were 1791 infants: 1334 in the pre-donor milk/probiotic cohort and 457 in the post-donor milk/probiotic cohort. On univariate analysis, mortality (7.6 vs. 2.4%, P<0.001) and incidence of sepsis (6.2 vs. 3.5%, P=0.028) were statistically significantly lower in the post-donor milk/probiotic group. NEC (2.8 vs. 1.5%, P=0.14) and non-NEC associ-ated gastrointestinal perforation (1.6 vs. 0.4%, P=0.052) were lower in the post-donor milk/probiotics cohort, but these were not statistically significant. The difference in mortality remained statistically significant on multivariate analysis in the post-donor milk/probiotic cohort compared to those in the pre-donor milk/probiotic cohort (odds ratio 0.31, 95% confidence interval 0.16–0.61). The decrease in the incidence of NEC was consistent with previous observational studies but the differ-ence was not statistically significant. Conclusion The availability of probiotics and pasteurised donor human milk is associated with a reduction in mortality in very preterm infants.

BACKGROUND The alpha-defensin lateral flow(ADLF)test is a new diagnostic tool for periprosthetic j.int infection(Pj.).Test accuracy for combined cohorts of hip and knee Pj. has been reported to be *** To assess the accuracy of the ADLF test for hip Pj.,and to compare three different diagnostic criteria for *** A cohort of 52 patients was identified,with a painful or poor-functioning total hip-or hemi-arthroplasty,that underwent aspiration and a subsequent ADLF *** was diagnosed with Musculoskeletal Infection Society(MSIS)criteria,and sensitivity,specificity,overall accuracy,positive predictive value and negative predictive value were ***,test specifics were compared with the European Bone and j.int Infection Society(EBj.S)and 2018 International Consensus Meeting(ICM)criteria for *** Using MSIS criteria,sensitivity was 100%(CI:54%-100%)and specificity was 89%(CI:76%-96%).Six true positives and 5 false positives were found,including one case of *** EBj.S criteria,more Pj.s were found(11 vs 6),sensitivity was lower(71%,CI:42%-92%)and specificity was higher(97%,CI:86%-100%),with 4 false negatives and one false positive *** 2018 ICM criteria,sensitivity was 91%(62%-100%)and specificity 100%(91%-100%).The results in this cohort are comparable to previous *** Overall test accuracy of the ADLF test was good in this cohort,with a sensitivity of 100%and specificity of 89%.Using different Pj. definition criteria,sensitivity and specificity changed slightly but overall accuracy remained around 90%.Using the ADLF test in metallosis cases can result in false positive results and should be performed with caution.

Dual Use and Biosecurity: the Case of the Avian Flu H5N1

The classical dual use problem—the potential for harmful as well as beneficial application of scientific findings—has become more immediate in biotechnology than in most other fields of science. Terrorist misuse of the information on the development of pathogenic organisms can lead to catastrophic outcomes. Therefore, particular in biosciences researchers are faced with the dilemma to find a proper balance between the right to know and the dangers of knowing. In this paper this dilemma is illustrated by the research on the influenza A virus subtype H5N1, commonly known as “bird flu”. The pros and cons of the full publication on the development of a dangerous airborne type are discussed.
