Reportedly,proteins involved in lipid metabolism change significantly in the jejunal crypt cells of earlyweaned piglets,but the exact lipid profile change remains uncertain.In the present study,32 piglets weaned at 21 d of age were randomly divided into 4 groups with 8 replicates.The jejunal crypt cells of a group of piglets on the post-weaning day(PWD)1,3,7,and 14 were isolated per time point.Crypt cell lipid profiles were analyzed using ultra-high-performance liquid chromatography coupled with hybrid quadrupole time-of-flight mass spectrometry.This study showed that piglets suffered the greatest weaning stress on PWD 3 in terms of the lowest relative weight of the small intestine,the highest relative weight of the spleen,and the highest levels of malondialdehyde,cholesterol,and low-density lipoprotein cholesterol.The lipid profile of jejunal crypt cells including carnitine,sulfatide,sphingomyelin,hexosylceramide,and ceramide greatly changed after weaning,especially between PWD3 and 14(Ppid species between these 2 d were mainly involved in the glycerophospholipid metabolism pathway.In addition,potential lipid biomarkers for weaning stress in crypt cells such as phosphatidylcholine(PC)(9:0/26:1),PC(17:0/18:2),carnitine(24:0),carnitine(22:0),sphingomyelin(d14:1/22:0),PC(P-18:0/18:4),phosphatidylethanolamine(P-16:0/20:4),phosphatidylinositol(15:1/24:4),and dihexosylceramide(d14:1/26:1)were identified.The changes in lipid profile might be related to the inflammation caused by early weaning.These findings might provide new therapeutical targets for intestinal dysfunctions caused by weaning stress.

Whole-genome sequencing and analysis of the Chinese herbal plant Gelsemium elegans

Gelsemium elegans(G.elegans)(2 n=2 x=16)is genus of flowering plants belonging to the Gelsemicaeae family.Here,a high-quality genome assembly using the Oxford Nanopore Technologies(ONT)platform and high-throughput chromosome conformation capture techniques(Hi-C)were used.A total of 56.11 Gb of raw GridION X5 platform ONT reads(6.23 Gb per cell)were generated.After filtering,53.45 Gb of clean reads were obtained,giving 160 x coverage depth.The de novo genome assemblies 335.13 Mb,close to the 338 Mb estimated by k-mer analysis,was generated with contig N50 of 10.23 Mb.The vast majority(99.2%)of the G.elegans assembled sequence was anchored onto 8 pseudo-chromosomes.The genome completeness was then evaluated and 1338 of the 1440 conserved genes(92.9%)could be found in the assembly.Genome annotation revealed that 43.16%of the G.elegans genome is composed of repetitive elements and 23.9%is composed of long terminal repeat elements.We predicted 26,768 protein-coding genes,of which 84.56%were functionally annotated.The genomic sequences of G.elegans could be a valuable source for comparative genomic analysis in the Gelsemicaeae family and will be useful for understanding the phylogenetic relationships of the indole alkaloid metabolism.

An integrated strategy toward comprehensive characterization and quantification of multiple components from herbal medicine: An application study in Gelsemium elegans

Objective: To develop a powerful integrated strategy based on liquid chromatography coupled with mass spectrometry(LC-MS) systems for the comprehensive characterization and quantification of multiple components of herbal medicines.Methods: Firstly, different mobile phase additives, analysis time, and MS acquisition modes were orthogonally tested with liquid chromatography coupled with quadrupole time-of-flight mass spectrometry(LC-QTOF/MS) in order to detect as many components of Gelsemium elegans as possible with high peak intensity. Secondly, several data mining strategies, including database searching, diagnostic ion filtering and neutral loss filtering, were utilized to perform chemical profiling. Subsequently, this study focused on the quantification and validation of the performance of a liquid chromatography-triple mass spectrometry(LC-Qq Q/MS) assay based on derivative multiple reaction monitoring(De MRM).Results: A total of 147 components from G. elegans were characterized, among them 116 nontarget components were reported for the first time. A sensitive and reproducible LC-Qq Q/MS method was successfully developed and validated for the simultaneous relative quantification of 41 components of G. elegans.This LC-Qq Q/MS method was then applied to compare the contents of components in the roots, stems and leaves.Conclusion: The present integrated strategy would significantly contribute to chemical studies on herbal medicine, and its utility could be extended to other research fields, such as metabolomics, quality control,and pharmacokinetics.

分别用2-丙烯酰胺-2-甲基丙磺酸(AMPS)、二甲基二烯丙基氯化铵(DMDAAC)、N-乙烯基吡咯烷酮(NVP)为单体,亚硫酸氢钠(NaHSO_(3))、过硫酸钾(KPS)、硝酸铈铵(CAN)为复合引发剂,通过自由基聚合法制备了XG-g-AMPS、XG-g-NVP和XG-g-DMDAAC三种改性聚合物。采用FTIR、EA、^(1)H NMR、SEM和GPC对接枝产物进行结构表征,并比较了黄原胶(XG)及改性聚合物的溶解性、耐盐性、剪切稀化性和耐热性能。结果表明,改性XG聚合物的溶解度显著提高,同时耐盐性有一定提高,XG-g-NVP、XG-g-DMDAAC在30%盐溶液中黏度相较XG分别增加了9%,15%。此外,XG-g-NVP和XG-g-DMDAAC溶液的剪切性能值相较XG分别提升了11%和45%。研究结果表明通过接枝共聚,改性XG聚合物的溶解性、耐盐性和剪切稀化性能得到提升,拓宽了黄原胶产品在石油工业中的应用。

Matched glass-to-Kovar seals in N_2 and Ar atmospheres

The oxidation of Kovar alloy was investigated, the wetting and spreading behavior of hard and soft glasses on Kovar alloy were studed by using the sessile drop method, and the quality and the seal strength of glass-Kovar seals were tested. The experimental results indicated that the preoxidation of Kovar alloy for approximately 10 min at 700℃ in air resulted in excellent adherence in glass-Kovar seals. The wetting and spreading behavior of glass on preoxidized Kovar alloy were superior to that on nonoxidized Kovar alloy. The wetting ability of ASF110 glass, at 950℃ and 980℃ in Ar and N2 atmospheres, was significantly superior to that of ASF200R and ASF700 glasses. The seal quality of the glass-preoxidized Kovar seal was superior to that of the glass-nonoxidized Kovar seal. The shear strength of the ASFll0 glass-preoxidized Kovar seal, which was prepared at 980℃ for 20 min in an Ar atmosphere, was approximately 3.9 MPa.

The overuse of antibiotics in animal agriculture and medicine has caused a series of potential threats to public health. Macleaya cordata is a medicinal plant species from the Papaveraceae family, providing a safe resource for the manufacture of antimicrobial feed additive for livestock. The active constituents from M. cordata are known to include benzylisoquinoline alkaloids （BIAs） such as sanguinarine （SAN） and chelerythrine （CHE）, but their metabolic pathways have yet to be studied in this non-model plant. The active biosynthesis of SAN and CHE in M. cordata was first examined and confirmed by feeding ^13C-labeled tyrosine. To gain further insights, we de novo sequenced the whole genome of M. cordata, the first to be sequenced from the Papaveraceae family. The M. cordata genome covering 378 Mb encodes 22,328 predicted protein-coding genes with 43.5% being transposable elements. As a member of basal eudicot, M. cordata genome lacks the paleohexaploidy event that occurred in almost all eudicots. From the genomics data, a complete set of 16 metabolic genes for SAN and CHE biosynthesis was retrieved, and 14 of their biochemical activities were validated. These genomics and metabolic data show the conserved BIA metabolic pathways in M. cordata and provide the knowledge foundation for future productions of SAN and CHE by crop improvement or microbial pathway reconstruction.

Morphologies and Superhydrophobicity of Hybrid Film Surfaces Based on Silica and Fluoropolymer

Fluoropolymer and different kinds of silica particles were used for controlling surface chemistry and morphology, respectively. A superhydrophobic surface originated from strawberry-like or quincunx-shaped composite silica particles was obtained. The dual size particles are obtained by utilizing the graft of different modified silica particles with epoxy functional group and amine functional group, This makes the surface of film form a composite interface to have irregular binary structure which plays an essential role in trapping air between the substrate surface and the liquid droplets to be necessary for high contact angle and low contact angle hysteresis. The maximum contact angle for water on the hybrid film is about 174±2° and the contact angle hysteresis is less than 2°. The surface morphologies, roughness and the wettability on the surface of films containing different structural silica particles were compared. It was shown that the hierarchical irregularly structure with a low roughness factor and high air-trapped ratio is indispensable for superhydrophobic surface. Although this structural surfaces based on composite silica particles play a vital role in governing the surface wettability, it is necessary to combine with a low surface energy to make the surface superhydrophobic.

高校项目教学是一种实践导向型的教学模式,能够促进学生对知识点的综合掌握,并有效提升其实践探索能力,为其今后工作打下基础。项目教学绩效的提升路径不同于一般教学活动,有其自身的特点。基于文献研究及相关调查,将项目教学绩效划分为过程感知绩效与结果感知绩效,将其影响因素的潜变量分为4组,包括教师水平、项目质量、资源投入及学生投入,并为各组潜变量配置了适宜的观测变量,进而构建了教学绩效的影响路径模型。实证部分通过学生问卷调查获得了相应的数据,进行了基本统计检验与结构方程检验。模型中相关影响路径总体上得到了验证,其中过程感知绩效受到直接影响较为显著,而结果感知绩效受到间接影响较为显著,文章基于实证结论提出了相关政策建议。

ZNF554 Inhibits Endometrial Cancer Progression via Regulating RBM5 and Inactivating WNT/β-Catenin Signaling Pathway

Objective:Uterine corpus endometrial carcinoma(UCEC),a kind of gynecologic malignancy,poses a significant risk to women’s health.The precise mechanism underlying the development of UCEC remains elusive.Zinc finger protein 554(ZNF554),a member of the Krüppel-associated box domain zinc finger protein superfamily,was reported to be dysregulated in various illnesses,including malignant tumors.This study aimed to examine the involvement of ZNF554 in the development of UCEC.Methods:The expression of ZNF554 in UCEC tissues and cell lines were examined by qRT-PCR and Western blot assay.Cells with stably overexpressed or knocked-down ZNF554 were established through lentivirus infection.CCK-8,wound healing,and Transwell invasion assays were employed to assess cell proliferation,migration,and invasion.Propidium iodide(pi)staining combined with fluorescence-activated cell sorting(FACS)flow cytometer was utilized to detect cell cycle distribution.qRT-PCR and Western blotting were conducted to examine relative mRNA and protein levels.Chromatin immunoprecipitation assay and luciferase reporter assay were used to explore the regulatory role of ZNF554 in RNA binding motif 5(RBM5).Results:The expression of ZNF554 was found to be reduced in both UCEC samples and cell lines.Decreased expression of ZNF554 was associated with higher tumor stage,decreased overall survival,and reduced disease-free survival in UCEC.ZNF554 overexpression suppressed cell proliferation,migration,and invasion,while also inducing cell cycle arrest.In contrast,a decrease in ZNF554 expression resulted in the opposite effect.Mechanistically,ZNF554 transcriptionally regulated RBM5,leading to the deactivation of the Wingless(WNT)/β-catenin signaling pathway.Moreover,the findings from rescue studies demonstrated that the inhibition of RBM5 negated the impact of ZNF554 overexpression onβ-catenin and p-glycogen synthase kinase-3β(p-GSK-3β).Similarly,the deliberate activation of RBM5 reduced the increase inβ-catenin and p-GSK-3βcaused by the suppression of ZNF554.In v
