Structure-based Comparative Analysis and Prediction of N-linked Glycosylation Sites in Evolutionarily Distant Eukaryotes

The asparagine-X-serine/threonine （NXS/T） motif, where X is any amino acid except proline, is the consensus motif for N-linked glycosylation. Significant numbers of high-resolution crystal structures of glycosylated proteins allow us to carry out structural analysis of the N-linked glycosylation sites （NGS）. Our analysis shows that there is enough structural information from diverse glycoproteins to allow the development of rules which can be used to predict NGS. A Python-based tool was developed to investigate asparagines implicated in N-glycosylation in five species： Homo sapiens, Mus musculus, Drosophila melanogaster, Arabidopsis thaliana and Saccharo- myees cerevisiae. Our analysis shows that 78 % of all asparagines of NXS/T motif involved in N-gly- cosylation are localized in the loop/turn conformation in the human proteome. Similar distribution was revealed for all the other species examined. Comparative analysis of the occurrence of NXS/T motifs not known to be glycosylated and their reverse sequence （S/TXN） shows a similar distribu- tion across the secondary structural elements, indicating that the NXS/T motif in itself is not bio- logically relevant. Based on our analysis, we have defined rules to determine NGS. Using machine learning methods based on these rules we can predict with 93% accuracy if a particular site will be glycosylated. If structural information is not available the tool uses structural prediction results resulting in 74% accuracy. The tool was used to identify glycosylation sites in 108 human proteins with structures and 2247 proteins without structures that have acquired NXS/T site/s due to non-synonymous variation. The tool, Structure Feature Analysis Tool （SFAT）, is freely available to the public at http：//***/tools/sfat.

Characterization of disease models of neurodegenerative disorders requires a systematic and comprehensive phenotyping in a highly standardized manner. Therefore, automated high-resolution behavior test systems such as the homecage based LabMaster system are of particular interest. We demonstrate the power of the automated LabMaster system by discovering previously unrecognized features of a recently characterized atxn3 mutant mouse model. This model provided neurological symptoms including gait ataxia, tremor, weight loss and premature death at the age of 12 months usually detectable just 2 weeks before the mice died. Moreover, using the LabMaster system we were able to detect hypoactivity in presymptomatic mutant mice in the dark as well as light phase. Additionally, we analyzed inflammation, immunological and hematological parameters, which indicated a reduced immune defense in phenotypic mice. Here we demonstrate that a detailed characterization even of organ systems that are usually not affected in SCA3 is important for further studies of pathogenesis and required for the preclinical therapeutic studies.

Detecting Damage in Reinforced Concrete Beams Using Vibrational Characteristics

This paper evaluates two methods of diagnosing damage, Natural frequency and Stiffness-Frequency change-Based damage detection method in reinforced concrete beams under load using vibration characteristics such as natural frequency and mode shape. The research uses finite element method with crack damage instead of deleting or reducing the bearing capacity of the element like in previous studies. First, a theory of the damage diagnosis method based on the change of natural frequency and mode shape is presented. Next, the simulation results of reinforced concrete beams using ANSYS will be compared with the experiment. Particularly, the investigated damage cases are cracks in reinforced concrete beams under loads. Finally, we will evaluate the accuracy of the damage diagnosis methods and suggest the location of the vibration data and specify the failure threshold of the methods.

现代电压源型换流器(VSC)能够产生准正弦波形,但对基于PWM的电平数较少的VSC,谐波含量仍然较大,需要配置性能良好的滤波器。从交直流两侧看入,换流器有一个等效谐波阻抗。估算此谐波阻抗对滤波器的设计和谐波谐振分析有着重要作用。基于开关函数的定量分析,利用VSC两侧的谐波的相互作用来计算等效谐波阻抗。同时分析了开关函数各开关分量之间的交叉调制给谐波阻抗计算带来的影响。此外,还进行了基于电压源换流器的高压直流输电的交直流两侧的谐振分析。从VSC直流侧看入的谐波阻抗虽然很大,但由于直流分压电容和直流电缆的寄生电容的影响,仍存在谐振的可能性。在交流侧,谐振可能出现在换流器的输出端口,且可能存在谐波放大现象。

谐波问题的分析与控制是保证高压直流输电系统正常运行的重要内容。为了寻找最适当的测量方法,需要对电压谐波测量进行深入研究。直流电压互感器(DCVT)是测量直流输电系统电压的重要设备。本文介绍与分析了阻容式直流电压互感器的主要组件的特性,如同轴电缆、A/D与D/A转换器、光耦合器,详细分析了分压器的对地杂散电容对电压互感器频率响应的影响,非理想状态下元器件对测量误差的影响。理论分析指出选择一个合适的并联电阻的电容可以减小分压器对地杂散电容的影响,并且可选择精密电阻和电容来减少测量误差。实验结果表明,阻容式直流电压互感器适合直流谐波电压测量,而且测量误差小于0.2%,可达到10k Hz的频率带宽。

近年来,随着互联网、移动设备、云计算和社交网络等技术的巨大进步,大量的数据在网络上存储和传输。数据的拥有者们只需要很少的代价,就可以将他们的数据委托存储在云端,并享受着方便的数据管理服务。而在这大量的委托存储数据中,具有隐私信息的图片占据了很大一部分。正因如此,这类具有个人隐私信息或者商业信息的图片的安全性要求迫在眉睫。在这类问题中,加密技术是一种最常见的解决方法,但是加密后的图片非常不便于分享和管理。为了满足管理上的需求,诸如拥有者、存储位置和时间戳等,辅助信息常常伴随着加密后的图片一起存储,而这些辅助信息往往对原始内容并没有什么影响。基于以上的观察,本论文将研究在加密图像中嵌入秘密信息的方法。在加密图像中嵌入秘密信息需要满足以下的要求:安全性、可逆性、可分离性和容量等。大多数已存在的方法并不能很好的满足以上提到的几个特性。为了解决这个问题,本文提出了一种新颖的加密域图像隐写方法。基于加密前预留空间框架,一幅原始图像先被分成两个部分,然后各自进行XOR和AES加密,从而提供安全性保证。在加密和预留空间的处理过程中使用差分扩展和奇偶转换来满足加密域可逆隐写的几个要求。同时,在加密图像中将会使用一种新颖的嵌入方法来嵌入辅助信息,该方法基于通过使用秘密像素点最低位隐藏信息的LSB技术。我们可以从嵌有秘密信息的加密图像中准确提取出秘密信息或者无失真地还原图像,信息提取和图像加密两个过程是可分离的。换句话说,只要拥有解密密钥和提取密钥,接收者可以先提取后解密或者先解密后提取。新方法的一个特别的特性就是只要拥有解密密钥,就可以直接将加密隐写后的图像还原,不需要提取出秘密信息(跟以前的其他算法不一样的)。我们还通过实验证明了本文提出的加密域可逆隐写方法拥有很大的嵌入容量。在Windows系统上,我们用C#语言实现了提出的方法,并在两个常见的图像数据库上进行了大量的实验。实验结果表明了所提出方法的有效性。同时,嵌入容量可以达到0.3 bpp或者更高,高于其他相关工作。另外,对应的PSNR值在可接受范围内(对于Lena图像,当嵌入率为0.2 bpp时,第一种方法的PSNR值为33.09 dB,第二种方法的PSNR值为58.12 dB)。最后,通过理论分析验证了所提出的方法的安全性、可逆性和可分离性。

Patterns of hepatitis B virus exposure and associated predictors in Vietnam: A crosssectional study

Objective:To examine the magnitude of isolated anti-HBc and other HBV serological patterns and associated predictors among adults seeking general health check-up at a large health center in ***:All 564 outpatients seeking general health checkup between January 2016 and December 2016 were asked to undertake HBV surface antigen,surface antibody,IgG and IgM core antibody(anti-HBc total),platelet counts,and liver function *** administered questionnaire was used to collect information regarding demography,in-house sources of infection,lifestyle,health condition and treatment,and HBV ***:Male gender(P=0.043),age(P=0.000),living in urban areas(P=0.040),HBV vaccination status(P=0.033),and ALT(P=0.040) were associated with isolated *** infection was associated with HBV vaccination status(P=0.001),ALT levels(P=0.010),AST levels(P=0.020),and platelet counts(P=0.007).Past/resolved HBV infection was associated with AST levels(P=0.005),ALT levels(P=0.014),and age(P=0.000).Conclusions:Isolated anti-HBc is quite *** of isolated anti-HBc include male gender,living in rural areas,and HBV *** prevalence of isolated anti-HBc also increases with *** timely detect occult HBV infection and prevent transmission,anti-HBc testing should be included in the health check-up for high risk individuals and screening program where HBV nucleic acid test is not *** prevent transmission,clinicians need to pay more attention on those who are at risk of having isolated anti-HBc and closely follow-up patients with isolated anti-HBc and educate them about the prevention of HBV infection.

Inferences on the Difference of Two Proportions: A Bayesian Approach

Let π=π1-π2 be the difference of two independent proportions related to two populations. We study the test H0:π≥0 against different alternatives, in the Bayesian context. The various Bayesian approaches use standard beta distributions, and are simple to derive and compute. But the more general test H0:π≥η, with η>0, requires more advanced mathematical tools to carry out the computations. These tools, which include the density of the difference of two general beta variables, are presented in the article, with numerical examples for illustrations to facilitate comprehension of results.

Estimation of Waste Generation and Recycling Potential from Traditional Market: A Case Study in Hue City, Vietnam

This study was conducted to provide a detailed description of waste generation and characteristics from a traditional market in Hue city, located in central Vietnam. The authors conducted a waste generation survey and a waste composition survey for 309 stalls/vendors in five markets by 17 business categories for 10 consecutive days. The waste generation rates by stall/vendor and by floor area were assessed in three waste categories: general waste, recyclable, and food residues. The general waste that would be sent to a landfill site was classified into 10 physical categories and 77 sub-categories. For general waste, food waste accounted for the largest part, followed by plastic and grass. By multiplying the waste generation rate by stall/vendor by the total number of stall/vendors in 23 markets, the authors estimated the total amounts of general waste, recyclable, food residue and total waste by business category. The total waste generated from market was 17.0 tons/day, of which 4.6 tons (27.1%) were collected by pig farmers for feeding livestock and 0.6 tons (3.6%) were sold to the recycling market. The composting potential accounted for 55.2% of total waste generation from the traditional market in Hue. The recycling potential accounted for 5.1%. The total disposal amount sent to the landfill site would be reduced from 69.2% to 8.8% of the total. The 95% confidence interval (CI) of total waste amount from 23 markets was also estimated using Monte Carlo simulation based on the mean and standard error of the waste generation rate. The range of 95% CI was 14.9 - 18.9 tons/day.

With the continuous development of technology,traditional manual work has been becoming more and more *** large or medium-sized companies have applied Enterprise Resource Planning(ERP)software into their business and production ***,since many small firms cannot afford ERP because of its expensive cost,they often still employ manual work for the same tasks this software resolves,especially for *** paper aims to provide a possible solution for small businesses to try automated scheduling and discover whether it can help *** are two main ways to make this determination:a mathematical model and a heuristic model,which are suitable for assessing low-and medium-sized workloads,*** case study was carried out in a small domestic interior furniture company,particularly in scheduling for their customized products in two-stage flow ***,they produce according to the sequence of customers’***,when we applied these supportive tools with batch-processing machines,they experienced enhanced production performance due to diminishing setup time for distinctive items and a more streamlined arrangement of job *** changes were implemented for some small companies that do not use many production stages and have a suitable number of jobs and *** this method were applied to larger demands,it would need further improvement and development to become a complete tool that can perform like a part of an ERP system.
