Catabolism of Branched Chain Amino Acids Supports Respiration but Not Volatile Synthesis in Tomato Fruits

The branched-chain amino acid transaminases （BCATs） have a crucial role in metabolism of the branched-chain amino acids leucine, isoleucine, and valine. These enzymes catalyze the last step of synthesis and the initial step of degradation of these amino acids. Although the biosynthetic pathways of branched chain amino acids in plants have been extensively investigated and a number of genes have been characterized, their catabolism in plants is not yet completely understood. We previously characterized the branched chain amino acid transaminase gene family in tomato, revealing both the subcellular localization and kinetic properties of the enzymes encoded by six genes. Here, we examined possible functions of the enzymes during fruit development. We further characterized transgenic plants differing in the expression of branched chain amino acid transaminases I and 3, evaluating the rates of respiration in fruits deficient in BCAT1 and the levels of volatiles in lines overexpressing either BCAT1 or BCAT3. We quantitatively tested, via precursor and isotope feeding experiments, the importance of the branched chain amino acids and their corresponding keto acids in the formation of fruit volatiles. Our results not only demonstrate for the first time the importance of branched chain amino acids in fruit respiration, but also reveal that keto acids, rather than amino acids, are the likely precursors for the branched chain flavor volatiles.

Plant respiration is characterized by two pathways for electron transfer to O2, namely the cytochrome pathway （CP） that is linked to ATP production, and the alternative pathway （AP）, where electrons from ubiquinol are directly transferred to O2 via an alternative oxidase （AOX） without concomitant ATP production. This latter pathway is well suited to dispose of excess electrons in the light, leading to optimized photosynthetic performance. We have characterized T- DNA-insertion mutant lines of Arabidopsis thaliana that do not express the major isoform, AOXIA. In standard growth conditions, these plants did not show any phenotype, but restriction of electron flow through CP by antimycin A, which induces AOXIA expression in the wild-type, led to an increased expression of AOXID in leaves of the aoxla-knockout mutant. Despite the increased presence of the AOX1D isoform in the mutant, antimycin A caused inhibition of photosyn- thesis, increased ROS, and ultimately resulted in amplified membrane leakage and necrosis when compared to the wild- type, which was only marginally affected by the inhibitor. It thus appears that AOX1 D was unable to fully compensate for the loss of AOXIA when electron flow via the CP is restricted. A combination of inhibition studies, coupled to metabolite profiling and targeted expression analysis of the P-protein of glycine decarboxylase complex （GDC）, suggests that the aoxla mutants attempt to increase their capacity for photorespiration. However, given their deficiency, it is intriguing that increase in expression neither of AOX1D nor of GDC could fully compensate for the lack of AOXIA to optimize pho- tosynthesis when treated with antimycin A. We suggest that the aoxla mutants can further be used to substantiate the current models concerning the influence of mitochondrial redox on photosynthetic performance and gene expression.

Objective: Botulinum toxin type A is frequently used to treat crow's feet lines. However, the optimal dose and injection sites are still controversial. The objective of this study was to compare the efficacy of different botulinum toxin type A injection patterns for the treatment of crow's feet. Methods: This single-center, retrospective, clinical study was conducted at the National Hospital of Dermatology and Venereology from July 2020 to December 2020. Data on 60 patients with crow's feet were collected and divided into 2 groups (3- or 4-point intramuscular injection) according to the intramuscular injection technique the patients received. The treatment efficacy was assessed based on the changes in the Crow's Feet Grading Scale score at 1, 4, and 16 weeks after treatment. Repeated measures analysis of variance was used for the assessment of changes in scores over time between the two groups. Results: After treatment, the average Crow's Feet Grading Scale score was significantly decreased compared with the pretreatment score at all timepoints (1, 4, and 16 weeks) in both states (dynamic: For 3-point intramuscular injection technique, 1 week: 1.90 ± 0.71 vs. 2.97 ± 0.56; 4 weeks: 1.87 ± 0.68 vs. 2.97 ± 0.56; 16 weeks: 2.60 ± 0.67 vs. 2.97 ± 0.56. For 4-point intramuscular injection technique, 1 week: 1.73 ± 0.83 vs. 3.03 ± 0.49; 4 weeks: 1.73 ± 0.74 vs. 3.03 ± 0.49; 16 weeks: 2.57 ± 0.68 vs. 3.03 ± 0.49, all P < 0.001. and static: For 3-point intramuscular injection technique, 1 week: 1.20 ± 0.89 vs. 2.20 ± 0.85; 4 weeks: 1.20 ± 0.89 vs. 2.20 ± 0.85; 16 weeks: 1.87 ± 0.97 vs. 2.20 ± 0.85; For 4-point intramuscular injection technique, 1 week: 1.50 ± 0.86 vs. 2.30 ± 0.84; 4 weeks: 1.33 ± 0.84 vs. 2.30 ± 0.84; 16 weeks: 1.87 ± 0.97 vs. 2.30 ± 0.84. The control synthesis of the high-speed underwater vehicle faces many technical challenges due to its inherent structure and surrounding operational *** this paper,the dynamical behavior is firstly described through a bifurcation analysis to give some insights for robust control *** a novel adaptive fractional-order sliding mode controller(AFOSMC)is realized to effectively manipulate the supercavitating vehicle against payload changes,nonlinear planing force,and external *** fractional order(FO)calculus can offer more flexibility and more freedom for tuning active control synthesis than the integer-order *** addition,the adaptation law has been presented to directly handle the payload change *** stability of the controlled vehicle system is proven via Lyapunov stability ***,the dynamic performance of the proposed controller is verified through extensive simulation results,which demonstrate the control accuracy with faster responses compared with existing integer-order ***,the proposed fractional order controllers can provide higher performance than their integer order counterparts with control algorithms.

In this study,mechanical characteristics of tensile strength ratio(TSR)were evaluated by comparing it to other methods in evaluating moisture sensitivity of asphalt *** types of asphalt mixtures with and without anti-stripping additives were prepared and tested in the *** compressive strength(UCS)and indirect tensile(IDT)testing were conducted to determine the mixture’s tensile strength ratio and cohesion ratio(CR).Determining TSR alone was found to be insufficient when evaluating moisture *** is a need to consider both TSR and wet IDT strength to properly evaluate moisture behavior of asphalt ***,it was found that the cohesion and the IDT strength have a good relation in which the minimum condition of the slope,must be ***,it was observed thatβvalues change depending on material type and ***,there is a need to evaluate theseβvalues for varying material and testing *** addition,it was found that the TSR and CR results are highly correlated with an average difference of 1.5%.Furthermore,other tests such as Marshall stability and dynamic immersion were also *** was found that Marshall stability ratio(MSR)and Marshall stability to flow ratio(MSFR)values are less sensitive to moisture effect when measuring moisture resistance compared to TSR ***,the 48 h testing result of the dynamic immersion test showed better correlation with TSR values.

The nuclear mean-field potential built up during the ^(12)C+^(12)C and ^(16)O+^(16)O collisions at low energies relevant for the carbon-and oxygen-burning processes is constructed within the double-folding model(DFM) using the realistic ground-state densities of^(12)C and^(16)O, and CDM3Yn density-dependent nucleon–nucleon(NN) interaction. The rearrangement term, indicated by the Hugenholtz–van Hove theorem for the single-particle energy in nuclear matter, is properly considered in the DFM calculation. To validate the use of the density-dependent NN interaction at low energies, an adiabatic approximation was suggested for the dinuclear overlap density. The reliability of the nucleus–nucleus potential predicted through this low-energy version of the DFM was tested in the optical model(OM) analysis of the elastic^(12)C+^(12)C and ^(16)O+^(16)O scattering data at energies below 10 MeV/*** OM results provide a consistently good description of the elastic angular distributions and 90 excitation function. The dinuclear mean-field potential predicted by the DFM is further used to determine the astrophysical S factor of the ^(12)C+^(12)C and ^(16)O+^(16)O fusions in the barrier penetration model. Without any adjustment of the potential strength, our results reproduce the non-resonant behavior of the S factor of the ^(12)C+^(12)C and ^(16)O+^(16)O fusions very well over a wide range of energies.

The control synthesis of a high-speed supercavitating vehicle(HSSV)faces many difficulties such as the stability,control,and maneuvering with dynamical uncertainties due to parameter perturbations,exter-nal disturbances,unmodeled dynamics,measurement noises,and actuator *** by the HSSV dynamical analysis,this paper proposes the H∞(i.e."H-infinity")robust control synthesis to gen-erate a robust low-order controller,which is intended for real implementations to ensure active control ***,the presented control scheme includes a feedback component and an anti-windup *** anti-windup synthesis is to provide system stability under actuator ***-tensive simulations show that the designed controller provides good performances with high robustness for vertical plane manoeuver while effectively eliminating planning forces,exogenous disturbances and noises as well as overcoming cavitator saturations.
