Time-lapse videography reveals different morphokinetic profiles of human embryos displaying direct or reverse cleavage at different stages of development: A retrospective sibling embryo study

Objective:To investigate morphokinetic characteristics of embryos displaying either reverse cleavage or direct cleavage during the first,second or third cleavage ***:A total of 167 in vitro fertilization and/or intracytoplasmic sperm injection treatment cycles undertaken by 167 women[aged(35.0±4.6)years]were included for reverse cleavage analysis,and a total of 167 in vitro fertilization and/or intracytoplasmic sperm injection treatment cycles undertaken by 167 women[aged(33.8±4.3)years]were included for direct cleavage analysis in this *** a sibling-embryo design,morphokinetic profiles(both before and after the onset of abnormal event)of embryos displaying reverse cleavage(n=241)or direct cleavage(n=244)were compared with their unaffected siblings(the controls)in the first,second and third cell cycles(n=32,n=142,n=562;n=195,n=412,n=205,respectively),at different developmental stages up to day ***:Direct cleavage embryos demonstrated significantly delayed cleavage rates prior to the event regardless of developmental stage of the occurrence,while reverse cleavage embryos showed similar cleavage rates to their unaffected *** event,direct cleavage embryos sped up cleavage rates while reverse cleavage embryos slowed ***:Altered morphokinetic profiles are displayed by direct cleavage embryos both before and after their occurrence and reverse cleavage embryos after the occurrence,which could potentially confound morphokinetic comparisons if not separated from their unaffected sibling *** study is warranted in order to fully understand the biological mechanisms of such events.

Dear Editor,Time-lapse videography of human embryos allows for the easy visualization of the embryos without removing them from the protective environment of the incubator[1],the measurement of various morphokinetic(quantitative)parameters[2],and the identification of abnormalities of growth(qualitative parameters)such as direct cleavage[3],reverse cleavage[4]and intercellular contact of blastomeres[5].

Proposed age-stratified reference intervals of FSH derived from normozoospermic men

Objective:To demonstrate that serum follicle-stimulating hormone(FSH)in men rises with age,and to explore FSH reference intervals of age-related ***:Men aged 20-50 years(n=1190)underwent semen analysis according to World Health Organization(2010)*** was frozen prior to measurement of FSH by using the Siemens ADVIA Centaur®XP immunoassay *** central 95%intervals after logarithmic transformation based on age were derived from 1037 normozoospermic *** were then applied to oligozoospermic and azoospermic *** producing azoospermic semen samples were further classified as having non-obstructive azoospermia by clinical diagnostic criteria,including genetic analysis and surgical ***:Serum FSH in normozoospermic men increased with age(Pls were determined with 10-year brackets:21-30 years[(1.0-8.2)IU/l],31-40 years[(1.4-9.5)IU/l],41-50 years[(1.9-12.0)IU/l].The proportion of oligozoospermic men with normal FSH concentrations was less than the normozoospermic men,which in turn was lower among azoospermic men(both Plevated serum FSH concentrations whereas only 6.7%(1/15)men with obstructive azoospermia had high FSH levels,and this was significantly different(Plusions:FSH concentrations increase in men between 20-50 years,and clinical interpretation of serum FSH results in men must be made by using age-based reference intervals.

Cloud-Based Optimal Control of Individual Borehole Heat Exchangers in a Geothermal Field

Integrating renewable energy sources is a crucial component in reducing CO_(2) emissions in the building sector. In particular, shallow geothermal energy is expected to play a significant role in the regenerative energy supply of buildings. An effective control strategy for the geothermal field is crucial to reduce the overall energy consumption. This paper analyzes the benefits of controlling an existing field’s individual borehole heat exchangers(BHE) using nonlinear model predictive control(NMPC) and moving horizon estimation. The considered geothermal field consists of 41 BHEs and is used for heating and cooling. Each BHE is equipped with temperature sensors for in-and outflow and has individually controllable valves, while a central hydraulic pump feeds all BHEs. The sensor measurements are accessed through a cloud platform, enabling also set point writing for the pump speed and the valve positions. To control the BHEs individually, we propose a two-stage process. In the calibration stage, a moving horizon estimator estimates the actual borehole and ground temperatures for each BHE. In the second stage, first, a nonlinear model predictive controller optimizes the number of active BHEs necessary to meet the buildings’ energy demand. With the estimated ground temperatures as a basis, it is determined which BHEs shall be(de)-activated. The active BHEs are fed with a fixed volume flow of 24 l/min to ensure turbulent heat transfer. To reduce the power usage of the pumps, an optimal control problem based on a simple hydraulic model of the geothermal field is used. The methodology is analyzed through simulations first and then validated experimentally. The results show that half or more of the BHEs could be deactivated most of the time, leading to 67% savings in electricity consumption by the hydraulic pump. Increasing numbers of amphibian species require conservation breeding programs for their survival.A major challenge is the efficient rearing of tadpoles,many of which require complex habitats and specialised *** tadpoles of the West Australian frog litoria moorei were kept at low density(1 tadpole per 1.95 litres water) in aquaria at 25℃.Fed on a staple diet of boiled lettuce and leaf litter,group of diets were supplemented with either control,Wardley Premium Spirulina Discs,SeraTMGVG-mix tropical fish food,or a combination of Wardley Premium Spirulina discs and SeraTMGVG-mix fish *** was a relatively high loss(i.e.,found dead,euthanized due to scoliosis,or not found) of tadpoles fed with the lettuce/leaf litter alone,but this was increased significantly when supplemented with Wardley Premium Spirulina discs,either alone or with SeraTMGVG-mix fish food,and SeraTMGVG fish food ***,the survived tadpoles fed on the three supplements were all heavier after three weeks and at metamorphosis than those fed with lettuce/leaf litter alone,and reached metamorphosis *** is concluded that any benefit of the food supplements in terms of increasing the rate of growth and development of the tadpoles is outweighed by greater *** is now a need for the efficient rearing of tadpoles,many from novel species that need complex *** studies of diet are required due to the current conservation crisis of amphibians.

High-throughput plant phenotyping for improved turfgrass breeding applications

Turfgrasses are used extensively throughout the world,and there is a steadfast demand to develop turfgrass varieties with improved abiotic and biotic stress tolerances that will perform well with limited management *** breeding programs incorporate advanced breeding strategies such as DNA sequencing and high-throughput phenotyping with traditional breeding strategies to identify and select germplasm and genes of *** biology methods and DNA sequencing technology have rapidly increased in recent years,and,as a result,plant phenotyping is currently a bottleneck in the process of advancing breeding *** advances in remote sensing technology have offered improved,non-destructive plant phenotyping approaches such as visible light imaging,spectral imaging,infrared thermal imaging,range sensing,and fluorescence *** mobile and time efficient platforms are being developed,coupling remote sensing with robotics and unmanned aerial systems technology for high-throughput plant phenotyping applications across large field *** turfgrass breeding programs will continue to research,develop,and implement remote sensing technologies to assess larger numbers of genotypes and identify elite *** together,these efforts will improve cultivar development efficiency and aid plant breeders in developing improved turfgrass cultivars to meet current and future demands of the turfgrass *** review provides an overview of ground-and aerial-based plant phenotyping platforms,with particular emphasis placed on applications to turfgrass breeding ***,imaging technologies that have been used in various plant breeding programs are discussed,with indications as to how those technologies could be applicable to turfgrass breeding programs.

Throughout its 40-year history,the field of gene therapy has been marked by many *** has seen great strides in combating human disease,has given hope to patients and families with limited treatment options,but has also been subject to many *** of patients with this class of investigational drugs has resulted in severe adverse effects and,even in rare cases,*** the heart of this dichotomous field are the viral-based vectors,the delivery vehicles that have allowed researchers and clinicians to develop powerful drug platforms,and have radically changed the face of *** the past 5 years,the gene therapy field has seen a wave of drugs based on viral vectors that have gained regulatory approval that come in a variety of designs and purposes,these modalities range from vector-based cancer therapies,to treating monogenic diseases with life-altering *** present,the three key vector strategies are based on adenoviruses,adeno-associated viruses,and *** have led the way in preclinical and clinical successes in the past two ***,despite these successes,many challenges still limit these approaches from attaining their full *** review the viral vector-based gene therapy landscape,we focus on these three highly regarded vector platforms and describe mechanisms of action and their roles in treating human disease.
