Increased.virulence of the oral microbiome in oral squamous cell carcinoma revealed.by metatranscriptome analyses

Oral squamous cell carcinoma(OSCC) is the most prevalent and.most commonly stud.ed.oral cancer. However, there is a void.regard.ng the role that the oral microbiome may play in OSCC. Although the relationship between microbial community composition and.OSCC has been thoroughly investigated. microbial profiles of the human microbiome in cancer are und.rstud.ed. Here we performed.a small pilot stud. of community-wid. metatranscriptome analysis to profile mRNA expression in the entire oral microbiome in OSCC to reveal molecular functions associated.with this d.sease. Fusobacteria showed.a statistically significantly higher number of transcripts at tumour sites and.tumour-ad.acent sites of cancer patients compared.to the healthy controls analysed. Regard.ess of the community composition, specific metabolic signatures were consistently found.in d.sease. Activities such as iron ion transport, tryptophanase activity, peptid.se activities and.superoxid. d.smutase were over-represented.in tumour and.tumour-ad.acent samples when compared.to the healthy controls. The expression of putative virulence factors in the oral communities associated.with OSCC showed.that activities related.to capsule biosynthesis, flagellum synthesis and.assembly, chemotaxis, iron transport, haemolysins and.ad.esins were upregulated.at tumour sites. Moreover, activities associated.with protection against reactive nitrogen intermed.ates, chemotaxis, flagellar and.capsule biosynthesis were also upregulated.in non-tumour sites of cancer patients. Although they are preliminary, our results further suggest that Fusobacteria may be the lead.ng phylogenetic group responsible for the increase in expression of virulence factors in the oral microbiome of OSCC patients.

Terrain is an important environment factor that plays a vital role in human d.velopment by influencing the spatial layout of land.use patterns. The terrain niche ind.x, combined.with slope and.elevation, can comprehensively present d.tailed.information about spatial d.fferences in terrain and.is superior to single terrain factors. We applied.remote sensing and.geographical information system to terrain grad.ent, used.the non-d.mensional d.stribution ind.x to examine spatial d.stribution characteristics of various land.cape types, and.analyzed.the ecological effects of land.cape were quantitatively on terrain grad.ents. Eco-environment quality (EEQ) was evaluated.using the forestry operation area as the evaluation unit. The spatial d.stributions in various land.capes were significantly influenced.by terrain factors, especially by bod.es of water, cropland.and.resid.ntial land. The spatial d.stribution varied.greatly in d.fferent terrain grad.ents for similar land.cape types. The areas associated.with good. intermed.ate and.poor EEQ were occupied.primarily by natural land.cape, semi-natural and.largely artificial land.capes, respectively.

Boosting Ad.ptive Weighted.Broad.Learning System for Multi-Label Learning

Multi-label classification is a challenging problem that has attracted.significant attention from researchers, particularly in the d.main of image and.text attribute annotation. However, multi-label d.tasets are prone to serious intra-class and.inter-class imbalance problems, which can significantly d.grad. the classification performance. To ad.ress the above issues, we propose the multi-label weighted.broad.learning system(MLW-BLS) from the perspective of label imbalance weighting and.label correlation mining. Further, we propose the multi-label ad.ptive weighted.broad.learning system(MLAW-BLS) to ad.ptively ad.ust the specific weights and.values of labels of MLW-BLS and.construct an efficient imbalanced.classifier set. Extensive experiments are cond.cted.on various d.tasets to evaluate the effectiveness of the proposed.mod.l, and.the results d.monstrate its superiority over other ad.anced.approaches.

The increasing population in old.r age will lead.to greater numbers of them presenting with acute coronary synd.omes （ACS）. This has implications on global healthcare resources and.necessitates better management and.selection for evid.nced.based.therapies. The eld.rly are a high risk group with more significant treatment benefits than younger ACS. Nevertheless, age related.inequalities in ACS care are recognised.and.persist. This d.screpancy in care, to some extent, is explained.by the higher frequency of atypical and.d.layed.presentations in the eld.rly, and.less d.agnostic electrocard.ograms at presentation, potentiating a d.lay in ACS d.agnosis. Und.r estimation of mortality risk in the eld.rly d.e to limited.consid.ration for physiological tiailty, co-morbid.ty, cognitive/psychological impairment and.physical d.sability, less input by card.ology specialists and.lack of rand.mised. controlled.trials d.ta to guid. management in the eld.rly may further confound.the inequality of care. While these inequalities exist, there remains a substantial opportunity to improve age related.ACS outcomes. The selection of eld.rly patients for specific therapies and.med.cation regimens are unanswered. There is a growing need.for rand.mised. controlled.trial d.ta to be more representative of the population and.enroll those of ad.anced.age with co-morbid.ty. A lack of reporting of ad.erse events, such as renal impairment post coronary angiography, in the eld.rly further limit risk benefit d.cisions. Substantial improvements in care of eld.rly ACS patients are required.and.should.be ad.ocated. Ultimately, these improvements are likely to lead.to better outcomes post ACS. However, the improvement in outcome is not infinite and.will be limited.by non-mod.fiable factors of age-related.risk.

Novel biologics that red.rect cytotoxic T lymphocytes （CTLs） to kill tumor cells bearing a tumor associated.antigen hold.great promise in the clinic. However, the ability to safely and.potently target Cd. on CTL toward.tumor associated.antigens （TAA） expressed.on tumor cells remains a challenge of both technology and.biol- ogy. Herein we d.scribe the use of a Half d.d.Ig format that can red.rect CTL to kill tumor cells. Notably, Half d.d.Ig molecules that are monovalent for each speci- ficity d.monstrated.red.ced.non-specific CTL activation and.cond.tional CTL activation upon bind.ng to TAA compared.to intact tetravalent d.d.Ig molecules that are bivalent for each specificity, while maintaining good.d.ug like properties and.appropriate PK properties.

Atomic layers of hexagonal boron nitrid.(h-BN)crystal are excellent cand.d.tes for structural materials as enabling ultrathin,two-d.mensional(2d.nanoelectromechanical systems(NEMS)d.e to the outstand.ng mechanical properties and.very wid. band.ap(5.9 eV)of *** this work,we report the experimental d.monstration of h-BN 2d.nanomechanical resonators vibrating at high and.very high frequencies(from~5 to~70 MHz),and.investigations of the elastic properties of h-BN by measuring the multimod. resonant behavior of these ***,we d.monstrate a d.y-transferred.d.ubly clamped.h-BN membrane with~6.7 nm thickness,the thinnest h-BN resonator known to *** ad.ition,we fabricate circular d.umhead.h-BN resonators with thicknesses ranging from~9 to 292 nm,from which we measure up to eight resonance mod.s in the range of~18 to 35 *** measurements and.mod.ling of the rich multimod. resonances,we resolve h-BN’s elastic behavior,includ.ng the transition from membrane to d.sk regime,with built-in tension ranging from 0.02 to 2 N m−*** Young’s mod.lus of h-BN is d.termined.to be EY≈392 GPa from the measured.*** ultrasensitive measurements further reveal subtle structural characteristics and.mechanical properties of the suspend.d.h-BN d.aphragms,includ.ng anisotropic built-in tension and.bulging,thus suggesting guid.lines on how these effects can be exploited.for engineering multimod. resonant functions in 2d.NEMS transd.cers.
