AIM To investigate the specific effects of immunosuppressants on the antiviral action of daclatasvir and *** The antiviral activity of daclatasvir(DCV) and asunaprevir(ASV) combined with immunosuppressants was tested using two in vitro models for hepatitis C virus(HCV) *** Tacrolimus, rapamycin and cyclosporine did not negatively affect the antiviral action of DCV or ASV. Mycophenolic acid(MPA) showed additive antiviral effects combined with these direct acting antivirals(DAAs). MPA induces interferon-stimulated genes(ISGs) and is a potent GTP synthesis inhibitor. DCV or ASV did not induce ISGs expression nor affected ISG induction by MPA. Rather, the combined antiviral effect of MPA with DCV and ASV was partly mediated via inhibition of GTP *** Immunosuppressants do not negatively affect the antiviral activity of DAAs. MPA has additive effect on the antiviral action of DCV and ASV. This combined benefit needs to be confirmed in prospective clinical trials.

AIM: To investigate the effects of mammalian target of rapamycin(mT OR) inhibition on liver regeneration and autophagy in a surgical resection ***: C57BL/6 mice were subjected to a 70% partial hepatectomy(PH) and treated intraperitoneally every 24 h with a combination of the m TOR inhibitor rapamycin(2.5 mg/kg per day) and the steroid dexamethasone(2.0 mg/kg per day) in phosphate bufferedsaline(PBS) or with PBS alone as vehicle control. In the immunosuppressant group, part of the group was treated subcutaneously 4 h prior to and 24 h after PH with a combination of human recombinant interleukin 6(IL-6; 500 μg/kg per day) and hepatocyte growth factor(HGF; 100 μg/kg per day) in PBS. Animals were sacrificed 2, 3 or 5 d after PH and liver tissue and blood were collected for further analysis. Immunohistochemical staining for 5-Bromo-2'-deoxyuridine(Brd U) was used to quantify hepatocyte proliferation. Western blotting was used to detect hepatic microtubule-associated protein 1 light chain 3(LC3)-Ⅱ protein expression as a marker for autophagy. Microbial adaptation to salinity can be achieved through synthesis of organic osmolytes,which requires high amounts of energy;however,a single addition of plant residues can only temporarily improve energy supply to soil ***,a laboratory incubation experiment was conducted to evaluate the responses of soil microbes to increasing salinity with repeated additions of plant residues using a loamy sand soil with an electrical conductivity in saturated paste extract(eC_e) of 0.6 dS m^(-1).The soil was kept non-saline or salinized by adding different amounts of NaCl to achieve eC_e of 12.5,25.0 and 50.0 dS m^(-1).The non-saline soil and the saline soils were amended with finely ground pea residues at two rates equivalent to 3.9 and 7.8 g C kg^(-1) soil on days 0,15 *** soils receiving no residues were included as a *** respiration per g C added over 2 weeks after each residue addition was always greater at 3.9 than 7.8 g C kg^(-1) soil and higher in the non-saline soil than in the saline *** the saline soils,the cumulative respiration per g C added was higher after the second and third additions than after the first addition except with3.9 g C kg^(-1) at eC_e of 50 dS m^(_1).Though with the same amount of C added(7.8 g C kg^(-1)),salinity reduced soil respiration to a lesser extent when 3.9 g C kg^(-1) was added twice compared to a single addition of 7.8 g C kg^(-1).After the third residue addition,the microbial biomass C concentration was significantly lower in the soils with eC_e of 25 and 50 dS m^(_1) than in the non-saline soil at3.9 g C kg^(-1),but only in the soil with eC_e of 50 dS m^(-1) at 7.8 g C kg^(-1).We concluded that repeated residue additions increased the adaptation of soil microbial community to salinity,which was likely due to high C availability providing microbes with the energy needed for synthesis of organic osmolytes.

Three-dimensional(3D)particle focusing in microfluidics is a fundamental capability with a wide range of applications,such as on-chip flow cytometry,where high-throughput analysis at the single-cell level is ***,3D focusing is achieved mainly in devices with complex layouts,additional sheath fluids,and complex pumping *** this work,we present a compact microfluidic device capable of 3D particle focusing at high flow rates and with a small footprint,without the requirement of external fields or lateral sheath flows,but using only a single-inlet,single-outlet microfluidic sequence of straight channels and tightly curving vertical *** device exploits inertial fluidic effects that occur in a laminar regime at sufficiently high flow rates,manipulating the particle positions by the combination of inertial lift forces and dean drag *** device is fabricated by femtosecond laser irradiation followed by chemical etching,which is a simple two-step process enabling the creation of 3D microfluidic networks in fused silica glass *** use of tightly curving three-dimensional microfluidic loops produces strong dean drag forces along the whole loop but also induces an asymmetric dean flow decay in the subsequent straight channel,thus producing rapid cross-sectional mixing flows that assist with 3D particle *** use of out-of-plane loops favors a compact parallelization of multiple focusing channels,allowing one to process large amounts of *** addition,the low fluidic resistance of the channel network is compatible with vacuum driven *** resulting device is quite interesting for high-throughput on-chip flow cytometry.

Blood-based mieroRNA （miRNA） signatures as biomarkers have been reported for various pathologies, including cancer, neurological disorders, cardiovascular diseases, and also infections. The regulatory mechanism behind respective miRNA patterns is only partially understood. Moreover, ＂preserved＂ miRNAs, i.e., miRNAs that are not dysregulated in any disease, and their biological impact have been explored to a very limited extent. We set out to systematically determine their role in regulatory networks by defining groups of highly-dysregulated miRNAs that contribute to a disease signature as opposed to preserved housekeeping miRNAs. We further determined preferential targets and pathways of both dysregulated and preserved miRNAs by computing multi-layer networks, which were compared between housekeeping and dysregulated miRNAs. Of 848 miRNAs examined across 1049 blood samples, 8 potential housekeepers showed very limited expression variations, while 20 miRNAs showed highly-dysregulated expression throughout the investigated blood samples. Our approach provides important insights into miRNAs and their role in regulatory networks. The methodology can be applied to systematically investigate the differences in target genes and pathways of arbitrary miRNA sets.

Background:With an increase in the evidence for the associations between park-based physical activity(PA)and physical environments(especially park and neighborhood environments),researchers face an important challenge in interpreting and summarizing the evidence to develop environ?mental change *** updated review is needed to better inform policymaking and environmental *** current study aimed to systematically review the research on the associations of park-based PA with park and neighborhood environmental ***:We targeted english peer-reviewed articles from 5 electronic databases using keywords related to park-based PA,park environments,and neighborhood *** the 4071 identified papers,25 studies published between 2008 and 2016 met all the eligibility criteria and were included in this ***:The characteristics of physical environment that received consistent support included paths/trails,lighting,and incivilities(e.g.,broken glasses and litter).Mixed findings were revealed for 6 park environmental factors(unspecified active facilities,playgrounds and skating areas,fit?ness stations,picnic areas,greenness,and park size)and 2 neighborhood environmental factors(park density and park proximity).Conclusion:It can be concluded that paths/trails,lighting,and incivilities are 3 key physical environmental attributes of park-based *** the inconsistent findings on park and neighborhood environmental factors,more robust designs such as prospective investigation are required.
