While hypoxic signaling has been shown to play a role in many cellular processes,its role in metabolism-linked extracellular matrix(ECM)organization and downstream processes of cell fate after musculoskeletal injury remains to be ***(HO)is a debilitating condition where abnormal bone formation occurs within extra-skeletal *** andhypoxia-inducible factor 1α(HIF-1α)activation have been shown to promote ***,the underlying molecular mechanisms bywhich the HIF-1αpathway in mesenchymal progenitor cells(MPCs)contributes to pathologic bone formation remain to ***,we used a proven mouse injury-induced HO model to investigate the role of HIF-1αon aberrant cell ***-cell RNA sequencing(scRNA-seq)and spatial transcriptomics analyses of the HO site,we found that collagen ECM organizationis the most highly up-regulated biological process in *** mesenchymal cell-specific deletion of Hif1α(Hoxa11-CreER^(T2);Hif1a^(fl/fl))significantly mitigated HO in ***-seq analysis of these Hoxa11-CreER^(T2);Hif1a^(fl/fl)mice identified the PLOD2/LOXpathway for collagen cross-linking as downstream of the HIF-1αregulation of ***,our scRNA-seq data and mechanisticstudies further uncovered that glucose metabolism in MPCs is most highly impacted by HIF-1α*** a translational aspect,a pan-LOX inhibitor significantly decreased HO.A newly screened compound revealed that the inhibition of PLOD2 activity in MPCssignificantly decreased osteogenic differentiation and glycolytic *** suggests that the HIF-1α/PLOD2/LOX axis linked tometabolism regulates HO-forming MPC *** results suggest that the HIF-1α/PLOD2/LOX pathway represents a promisingstrategy to mitigate HO formation.

Ammonia has emerged as a promising energy carrier owing to its carbon neutral content and low expense in long-range ***,development of a specific pathway to release the energy stored in ammonia is therefore in urgent *** oxidation provides a convenient and reliable route to attain efficient utilization of ***,we report that the high entropy(Mn,Fe,Co,Ni,Cu)_(3)O_(4)oxides can achieve high electrocatalytic activity for ammonia oxidation reaction(AOR)in non-aqueous *** AOR onset overpotential of(Mn,Fe,Co,Ni,Cu)_(3)O_(4)is 0.70 V,which is nearly 0.2 V lower than that of their most active single metal cation *** mass spectroscopy study reveals that(Mn,Fe,Co,Ni,Cu)_(3)O_(4)preferentially oxidizes ammonia to environmentally friendly diatomic nitrogen with a Faradic efficiency of over 85%.The Xray photoelectron spectroscopy(XPS)result indicates that the balancing metal d-band of Mn and Cu cations helps retain a longlasting electrocatalytic ***,this work introduces a new family of earth-abundant transition metal high entropy oxide electrocatalysts for AOR,thus heralding a new paradigm of catalyst design for enabling ammonia as an energy carrier.

Electroacupuncture improves microcirculation and neuronal morphology in the spinal cord of a rat model of intervertebral disc extrusion

Most studies on spinal cord neuronal injury have focused on spinal cord tissue histology and the expression of nerve cell damage and repair-related genes. The importance of the microcirculation is often ignored in spinal cord injury and repair research. Therefore, in this study, we established a rat model of intervertebral disc extrusion by inserting a silica gel pad into the left ventral sur-face of T13. Electroacupuncture was used to stimulate the bilateralZusanlipoint （ST36） and Neiting point （ST44） for 14 days. Compared with control animals, blood lfow in the ifrst lumbar vertebra （L1） was noticeably increased in rats given electroacupuncture. Microvessel density in the T13 segment of the spinal cord was increased significantly as well. The number of normal neurons was higher in the ventral horn of the spinal cord. In addition, vacuolation in the white matter was lessened. No obvious glial cell proliferation was visible. Furthermore, hindlimb motor function was improved signiifcantly. Collectively, our results suggest that electroacupuncture can improve neuronal morphology and microcirculation, and promote the recovery of neurological functions in a rat model of intervertebral disc extrusion.

Improving the conduct of meta-analyses of observational studies

The author, who has published numerous meta-analyses of epidemiological studies, particularly on tobacco, comments on various aspects of their content. While such meta-analyses, even when well conducted, are more difficult to draw inferences from than are meta-analyses of clinical trials, they allow greater insight into an association than do simple qualitative reviews. This editorial starts with a discussion of some problems relating to hypothesis definition. These include the definition of the outcome, the exposure and the population to be considered, as well as the study inclusion and exclusion criteria. Under literature searching, the author argues against restriction to studies published in peer-reviewed journals, emphasising the fact that relevant data may be available from other sources. Problems of identifying studies and double counting are discussed, as are various issues in regard to data entry. The need to check published effect estimates is emphasised, and techniques to calculate estimates from material provided in the source publication are described. Once the data have been collected and an overall effect estimate obtained, tests for heterogeneity should be conducted in relation to different study characteristics. Though some meta-analysts recommend classifying studies by an overall index of study quality, the author prefers to separately investigate heterogeneity by those factors which contribute to the assessment of quality. Reasons why an association may not actually reflect a true causal relationship are also discussed, with the editorial describing techniques for investigating the relevance of confounding, and referring to problems resulting from misclassification of key variables. Misclassification of disease, exposure and confounding variables can all produce a spurious association, as can misclassification of the variable used to determine whether an individual can enter the study, and the author points to techniques to adjust for this. 空间分析及基础技术应用是研究集装箱运输的必要手段.大部分交通运输数据集由不同出处、关联性不强的原始数据组成,因而很难得到其空间分布特点.同时,集装箱路径优化和市场可达性在缓解集装箱运输拥堵方面的作用日益凸显.本文基于可用数据库对美国集装箱运输OD分布进行分析.以美国水陆运输理事会的公共货票数据库数据作为内陆铁路线路优化的原始输入.利用类似河流管理的方法优化集装箱运输路径.从进口集装箱的数据集看,从亚洲或者欧洲出发经由北美港口到达美国的集装箱,大部分经由太平洋海岸到达内陆市场.横渡太平洋贸易的进口集装箱枢纽港主要是洛杉矶、长滩和西雅图.横渡大西洋贸易的进口集装箱枢纽港主要是纽约、新泽西州和萨凡纳乔治亚州.太平洋海岸和密西西比河谷地区包括阿尼阿波利斯、芝加哥、堪萨斯城、孟菲斯、达拉斯和休斯顿的流量很高.从美国中西部至东部的集装箱流多经过纽约、新泽西州和诺福克最后到达芝加哥.对于集装箱运输OD分布的分析可以为高成本和多式联运相关分析提供数据基础.

Optical properties of excitons in strained Ga_x In_1-x As/GaAs quantum dot: effect of geometrical confinement on exciton g-factor

Taking into account anisotropy, nonparabolicity of the conduction band, and geometrical confinement, we discuss the heavy-hole excitonic states in a strained GaxIn1-xAs/GaAs quantum dot for various Ga alloy contents. The strained quantum dot is considered as a spherical InAs dot surrounded by a GaAs barrier material. The dependence of the effective excitonic g-factor as a function of dot radius and Ga ion content is numerically measured. Interband optical energy with and without the parabolic effect is computed using structural confinement. The interband matrix element for different Ga concentrations is also calculated. The oscillator strength of interband transitions on the dot radius is studied at different Ga concentrations in the GaxIn1-xAs/GaAs quantum dot. Heavy-hole excitonic absorption spectra are recorded for various Ga alloy contents in the GaxIn1-xAs/GaAs quantum dot. Results show that oscillator strength diminishes when dot size decreases because of the dominance of the quantum size effect. Furthermore, exchange enhancement and exchange sDlitting increase as exciton confinement inereases.
