We developed a new plant transformation vector, pHairyRed, for enabling high throughput, non-destructive selection of Agrobacterium rhizogenes-mediated ＇hairy-root＇ transformation, pHairyRed allows easy in planta visualiza- tion of transgenic tissue with minimal disturbance to the plant. The DsRed2 reporter gene, encoding a red fluorescent protein, was cloned to yield pHairyRed （harbouring a multiple cloning site）, which was used with the highly efficient K599 A. rhizogenes strain to infect soybean （Glycine max L. Merrill） plants. DsRed2 fluorescence was easily detected in planta for the duration of a 5-week study with negligible levels of background autofluorescence. This enabled visual selection of transformed roots and subsequent excission of non-transformed roots, pHairyRed-transformed roots nodu- lated normally when inoculated with Bradyrhizobiumj.ponicum. Within the nodule, DsRed2 fluorescence was plant-spe- cific, being absent in the bacteroid-dominated nodule infected zone. To test the reliability of pHairyRed as a high-fidelity binary vector reporter system, the gene encoding the soybean Nod factor receptor, GmNFRloL, was cloned into the vector for use in a complementation study with a non-nodulating nfrloL mutant of soybean. Complementation was achieved and, without exception, DsRed2 fluorescence was detected in all hairy roots that successfully formed nodules （100%, n = 34）. We anticipate broad application of this reporter system for the further analysis of root-related events in soybean and related legumes.

AIM To demonstrate the effectiveness of a digital radiographic altering technique in concealing treatment allocation to blind outcome assessment of distal femur fracture *** Digital postoperative anteroposterior and lateral radiographs from a sample of 33 randomly-selected patients with extra-articular distal femur fractures treated by surgical fixation at a Level 1 trauma center were included. Using commercially available digital altering software, we devised a technique to blind the radiographs by overlaying black boxes over the implant hardware while preserving an exposed fracture site for assessment of fracture healing. Three fellowship-trained surgeons evaluated a set of blinded radiographs twice and a control set of unblinded radiographs once. Each set of radiographs were reviewed independently and in a randomly-assigned order. The degrees of agreement and disagreement among evaluators in identifying implant type while reviewing both blinded and unblinded radiographs were assessed using the Bang Blinding Index and j.mes Blinding Index. The degree of agreement in fracture union was assessed using kappa *** The assessment of blinded radiographs with both the Bang Blinding Index(BBI) and j.mes Blinding Index(j.I) demonstrated a low degree of evaluator success at identifying implant type(Mean BBI, far cortical locking:-0.03, SD: 0.04; Mean BBI, standard screw: 0, SD: 0; j.I: 0.98, SD: 0), suggesting near perfect blinding. The assessment of unblinded radiographs with both blinding indices demonstrated a high degree of evaluator success at identifying implant type(Mean BBI, far cortical locking: 0.89, SD: 0.19; Mean BBI, standard screw: 0.87, SD: 0.04; j.I: 0.26, SD: 0.12), as expected. There was moderate agreement with regard to assessment of fracture union among the evaluators in both the blinded(Kappa: 0.38, 95%CI: 0.25-0.52) and unblinded(Kappa: 0.35, 95%CI: 0.25-0.45) arms of the study. Some of the key mathematical results are stated without proof in order to make the underlying theory accessible to a wider audience. The book assumes a knowledge only of basic calculus, matrix algebra, and elementary statistics. The emphasis is on methods and the analysis of data sets. The logic and tools of model-building for stationary and nonstationary time series are developed in detail and numerous exercises, many of which make use of the included computer package, provide the reader with ample opportunity to develop skills in this area.;The core of the book covers stationary processes, ARMA and ARIMA processes, multivariate time series and state-space models, with an optional chapter on spectral analysis. Additional topics include harmonic regression, the Burg and Hannan-Rissanen algorithms, unit roots, regression with ARMA errors, structural models, the EM algorithm, generalized state-space models with applications to time series of count data, exponential smoothing, the Holt-Winters and ARAR forecasting algorithms, transfer function models and intervention analysis. Brief introductions are also given to cointegration and to nonlinear, continuous-time and long-memory models.;The time series package included in the back of the book is a slightly modified version of the package ITSM, published separately as ITSM for Windows, by Springer-Verlag, 1994. It does not handle such large data sets as ITSM for Windows, but like the latter, runs on IBM-PC compatible computers under either DOS or Windows (version 3.1 or later). The programs are all menu-driven so that the reader can immediately apply the techniques in the book to time series data, with a minimal investment of time in the computational and algorithmic aspects of the analysis.

The radial basis function networks were applied to bacterial classification based on the matrix-assisted laser desorption/ionization time-of-flight mass spectrometric (MALDI-TOF-MS) data. The classification of bacteria cultured at different time was discussed and the effect of the network parameters on the classification was investigated. The cross-validation method was used to test the trained networks. The correctness of the classification of different bacteria investigated changes in a wide range from 61.5% to 92.8%. Owing to the complexity of biological effects in bacterial growth, the more rigid control of bacterial culture conditions seems to be a critical factor for improving the rate of correctness for bacterial classification.

The ability to rationally engineer the growth and nanomanufacturing of one-dimensional nanowires in high volumes has the potential to enable applications of nanoscale materials in a diverse range of fields including energy conversion and storage,catalysis,sensing,medicine,and information *** review provides a roadmap for the development of large-scale nanowire *** myriad techniques exist for bench-scale nanowire synthesis,these growth strategies typically fall within two maj.r categories:1) anisotropically-catalyzed growth and 2) confined,template-based ***,comparisons between growth methods with different mass transport pathways have led to confusion in interpreting observations,in particular Gibbs-Thomson *** review mass transport in nanowire synthesis techniques to unify growth models and to allow for direct comparison of observations across different *** addition,we discuss the applicability of nanoscale,Gibbs-Thomson effects on mass transport and provide guidelines for the development of new growth *** explore the scalability of these complex processes with dimensionless numbers and consider the effects of pressure,temperature,and precursor material on nanowire growth.

青藏高原及邻区前成冰纪物质组成及其地质演化过程,在地球早期超大陆形成演化过程的研究中扮演了重要角色.本文系统梳理和总结了青藏高原及邻区前成冰纪沉积岩、古生物、岩浆和变质作用特征.根据现有资料,青藏高原及邻区前成冰纪沉积岩和古生物记录主要集中于中~新元古代,古元古代及更早的记录相对较少,最古老的地质记录为喜马拉雅和昌都地区前寒武纪变质沉积岩中的冥古宙碎屑锆石(约4.0Ga).青藏高原及邻区保存了凯诺兰超级克拉通以及哥伦比亚、罗迪尼亚和冈瓦纳超大陆形成演化过程的记录,其前成冰纪基底可划分为3种类型:塔里木型、扬子型和拉萨型.塔里木型基底与澳大利亚和印度大陆北缘具有古地理亲缘性,且不发育罗迪尼亚超大陆聚合相关的碎屑锆石年龄峰值和岩浆-变质记录(约1.3~0.9Ga);扬子型基底发育与前成冰纪最晚期全球变冷密切相关的火山活动记录,可见中~新元古代叠层石和微古植物化石,以及约1.1~1.0Ga与罗迪尼亚超大陆聚合相关的岩浆-变质作用记录;拉萨型基底发育新元古代早期大陆裂谷沉积记录(约900Ma)和晚期高压变质事件(约650Ma),具有明显的1.2~1.1Ga碎屑锆石年龄峰值,它很可能与非洲大陆具有古地理亲缘性.
