AIM:To evaluate whether desferrioxamine decreases ischemia and perfusion injury aggravated by cold storage(CS)in a rat liver perfusion model. METHODS:Isolated rat livers were kept in CS in University of Wisconsin Solution for 20 h at 4℃,then exposed to 25 min of warm ischemia(WI)at 37℃ followed by 2 h of warm perfusion(WP)at 37℃with oxygenated(95%oxygen and 5%carbon dioxide) Krebs-Henseleit ***(DFO),an iron chelator,was added at different stages of storage,ischemia and perfusion:in CS only,in WI only,in WP only, in WI and perfusion,or in all *** samples were collected after CS and after *** samples and bile were collected every 30 min(0,0.5,1,1.5 and 2 h)during liver *** injury was assessed by the determination of lactate dehydrogenase(LDH) and aspartate aminotransferase(AST)in the effluent and perfusate *** iron was analysed in the perfusate *** WP,the liver was collected for the determination of liver swelling(wet to dry ratio) and liver morphological examination(hematoxylin and eosin staining). Efficacy and safety of bridging thrombolysis initiated before transfer in a drip-and-ship stroke service

High-throughput screening of mouse gene knockouts identifies established and novel skeletal phenotypes

Screening gene function in vivo is a powerful approach to discover novel drug targets. We present high-throughput screening （HTS） data for 3 762 distinct global gene knockout （KO） mouse lines with viable adult homozygous mice generated using either gene-trap or homologous recombination technologies. Bone mass was determined from DEXA scans of male and female mice at 14 weeks of age and by microCT analyses of bones from male mice at 16 weeks of age. Wild-type （WT） cagemates/littermates were examined for each gene KO. Lethality was observed in an additional 850 KO lines. Since primary HTS are susceptible to false positive findings, additional cohorts of mice from KO lines with intriguing HTS bone data were examined. Aging, ovariectomy, histomorphometry and bone strength studies were performed and possible non-skeletal phenotypes were explored. Together, these screens identified multiple genes affecting bone mass： 23 previously reported genes （Calcr, Cebpb, Crtap, Dcstamp, Dkkl, Duoxa2, Enppl, Fgf23, Kissl/Kisslr, Kl （Klotho）, Lrp5, Mstn, Neol, Npr2, Ostml, Postn, Sfrp4, S1c30a5, Sic39a13, Sost, Sumf1, Src, Wnt10b）, five novel genes extensively characterized （Cldn18, Fam20c, Lrrkl, Sgpll, Wnt16）, five novel genes with preliminary characterization （Agpat2, RassfS, Slc10a7, Stc26a7, Slc30a10） and three novel undisclosed genes coding for potential osteoporosis drug targets.

Astroglia are a broad class of neural parenchymal cells primarily dedicated to homoeostasis and defence of the central nervous system(CNS).Astroglia contribute to the pathophysiology of all neurological and neuropsychiatric disorders in ways that can be either beneficial or detrimental to disorder *** changes in astroglia can be primary or secondary and can result in gain or loss of *** respond to external,non-cell autonomous signals associated with any form of CNS pathology by undergoing complex and variable changes in their structure,molecular expression,and *** addition,internally driven,cell autonomous changes of astroglial innate properties can lead to CNS *** pathophysiology is complex,with different pathophysiological cell states and cell phenotypes that are context-specific and vary with disorder,disorder-stage,comorbidities,age,and ***,we classify astroglial pathophysiology into(i)reactive astrogliosis,(ii)astroglial atrophy with loss of function,(iii)astroglial degeneration and death,and(iv)astrocytopathies characterised by aberrant forms that drive *** review astroglial pathophysiology across the spectrum of human CNS diseases and disorders,including neurotrauma,stroke,neuroinfection,autoimmune attack and epilepsy,as well as neurodevelopmental,neurodegenerative,metabolic and neuropsychiatric *** cellular and molecular mechanisms of astroglial pathophysiology represents a new frontier to identify novel therapeutic strategies.

The Schwann cell reaction to nerve injury,termed the repair program,is crucial to successful nerve *** the last decade,substantial advances have been made in elucidating the underlying molecular mechanisms in Schwann cells that lead to functional nerve ***,the field has identified situations,such as aging and chronic denervation where these mechanisms go awry,paving the way for the development of therapeutic interventions(arthur-Farraj and Coleman,2021;Cattin and Lloyd,2016;Jessen and Mirsky,2019).A recent article by Deborde et al.(2022)has demonstrated that unfortunately there is a downside to Schwann cells having such an efficient regeneration-promoting program;the promotion of tumor invasion of peripheral nerves.

Structure and magnetostructural correlation of ferrimagnetic meso-tetraphenylporphinatomanganese(Ⅲ) dimethyl- N,N′-dicyanoquinone diiminide, [MnTPP]^+[Me_2DCNQI]^(.-)

The structure [MnmTPP][Me2DCNQI] （TPP = tetraphenylporphinato; Me2DCNQI = 2,5-dimethyl-N,N＇-dicyanoquinonediimine） has been determined from X-ray powder diffraction data. The nonsolvated structure is composed of linear （I-D） chains of alternating [MnmTPP]＋ and ta-[MezDCNQI]＇- with intrachain Mn...Mn separations of 12.83 A, and a Mn-NDcNQI distance of 2.18A. The dihedral angle between the mean Mn（N4）xpp and [MezDCNQI]＇-planes, and the Mn-（N-C）ocNQ1 angle are 84.18° and 143.6°, respectively. [MnInTpP][MezDCNQI] has a Tc of 4.3 K from the 10 Hz x（T） data, 2-K coercivity of 5,600 Oe, and 6,300 emuOe/mol remnant magnetization that are reduced from that observed for related materials, and their in- clusion extends the magnetostructural correlation between the intrachain coupling and both the dihedral angle between the mean Mn（N4）xPp and [TCNE]＇-（TCNE --- tetracyanoethylene） planes and Mn-（N-C）Tcr angles. This is in accord with the in- trachain coupling arising from the overlap of the Mnm dz2-1ike singly occupied molecular orbital （SOMO） and the z component of the [TCNE]- π＊（π2＊） SOMO, which increases with decreasing dihedral angle between the mean Mn（N4）xep and [TCNE]＇- planes and Mn-（N-C）TcNE angle.

Non-Random Distribution of Human Orofacial Clefts in ghana: gene-Environment Interactions

Human orofacial clefts (OFCs) are congenital anomalies that result from the breakdown of normal mechanisms that regulate the formation of the human face. They could be syndromic or non-syndromic, with a global incidence of 1:700 per live births. Environmental and genetic factors are thought to play various roles in the aetiology of OFCs. This study seeks to establish the diversity, distribution and pattern of inheritance of OFCs as well as environmental and other risk factors associated with OFCs in a ghanaian population. A family-based, descriptive cross-sectional study that employed an interview-based survey questionnaire was used to obtain information from 467 families with history of OFCs. We employed chi-square statistics to analyse the data and used graphs to interpret the data. All previously reported subphenotypes of OFCs were observed by the present study. Clinically, about 12% of clefts in the study population were syndromic. The most common syndromic forms observed were Pierre Robin Sequence, cleft-with-club foot abnormalities and Van der Woude Syndrome. Only about 5% of clefts in the study cohort were familial. The study also established that lower level of education of parents, poverty, late antenatal care and dietary folate deficiency are major environmental factors associated with clefts in the ghanaian population. In conclusion, OFCs are non-randomly distributed in ghana and folate deficiency could likely be a source of genetic mutations and “epimutations” that cause OFCs, since folate is essential for DNA methylation, replication and repair as well as histone modification.
