Dietary advanced glycation end-products aggravate non-alcoholic fatty liver disease

AIM To determine if manipulation of dietary advanced glycation end product(AGE), intake affects nonalcoholic fatty liver disease(NAFLD) progression and whether these effects are mediated via RAGE. METHODS Male C57bl6 mice were fed a high fat, high fructose, high cholesterol(HFHC) diet for 33 wk and compared with animals on normal chow. A third group were given a HFHC diet that was high in AGEs. Another group was given a HFHC diet that was marinated in vinegar to prevent the formation of AGEs. In a second experiment, RAGE KO animals were fed a HFHC diet or a high AGE HFHC diet and compared with wildtype controls. Hepatic biochemistry, histology, picrosirius red morphometry and hepatic mR NA were determined. RESULTS Long-term consumption of the HFHC diet generated significant steatohepatitis and fibrosis after 33 wk. In this model, hepatic 4-hydroxynonenal content(a marker of chronic oxidative stress), hepatocyte ballooning, picrosirius red staining, α-smooth muscle actin and collagen type 1A gene expression were all significantly increased. Increasing the AGE content of the HFHC diet by baking further increased these markers of liver damage, but this was abrogated by pre-marination in acetic acid. In response to the HFHC diet, RAGE-/-animals developed NASH of similar severity to RAGE+/+ animals but were protected from the additional harmful effects of the high AGE containing diet. Studies in isolated Kupffer cells showed that AGEs increase cell proliferation and oxidative stress, providing a likely mechanism through which these compounds contribute to liver injury. CONCLUSION In the HFHC model of NAFLD, manipulation of dietary AGEs modulates liver injury, inflammation, and liver fibrosis via a RAGE dependent pathway. This suggests that pharmacological and dietary strategies targeting the AGE/RAGE pathway could slow the progression of NAFLD.

HbeAg negative variants and their role in the natural history of chronic hepatitis b virus infection

Molecular virology methods including polymerase chain reaction, cloning and sequencing have revolutionised our understanding of viral genome variation. In the case of hepatitis b virus (HbV), sequencing studies have identified a number of virus variants normally found during the natural course of chronic infection. The appearance of the precore stop codon (with G-for-A substitution at position 1896) and basal core promoter (bCP) (with A-for-T and G-for-A, at positions 1762 and 1764, respectively) variants which reduce or abrogate hepatitis b e antigen (HbeAg) production, heralds the initiation of the seroconversion phase from HbeAg to anti-Hbe positivity. The gradual removal of the tolerogenic effect of HbeAg leads to the awakening of the immune response (immune clearance phase). Most patients after HbeAg seroconversion become “inactive HbsAg carriers”. However during the course of infection precore and/or bCP variants may emerge and be selected leading to HbeAg negative chronic hepatitis b (CHb) with high viremia levels (reactivation phase). The prevalence of HbeAg negative CHb has been increasing over the last few decades and has become the commonest type of HbV infection in many countries of the world. This probably reflects the aging of existing HbV carriers and the effective prevention measures restricting new HbV infections. Frequent acute exacerbations accompanied by high viral replication, elevated alanine aminotransferase levels and histological activity are a common feature of HbeAg negative CHb leading to cirrhosis much faster than in HbeAg positive CHb patients.

The fundamental idea that responses to environmental factors or treatments is to be found in our individual differences, the underlying concept of “genomic medicine”, is rooted in antiquity and based on millennia of simple observation. Simply put, the objective of genomic medicine is to determine the genetic bases of those differences in response to environmental agents, including medications, and differences that may predispose to the development of common and potentially personally devastating and societally expensive disorders, and to use them in populations to thwart adverse response, increase the frequency of beneficial response, and intervene to prevent or delay onset of disease.

Making the link between ADF and 4D STEM:Resolution,transfer and coherence

Steve Pennycook is a pioneer in the application of high-resolution scanning transmission electron microscopy(STEM)and in particular the use of annular dark-field(ADF)*** we show how a general framework for 4D STEM allows clear links to be made between ADF imaging and the emerging methods for reconstructing images from 4D STEM data *** show that both ADF imaging and ptychographical reconstruction can be thought of in terms of integrating over the overlap regions of diffracted discs in the detector *** approach allows the similarities in parts of their transfer functions to be understood,though we note that the transfer functions for ptychographic imaging cannot be used as a measure of information *** also show that conditions of partial spatial and temporal coherence affect ADF imaging and ptychography similarly,showing that achromatic interference can always contribute to the image in both cases,leading to a robustness to partial temporal coherence that has enabled high-resolution imaging.

Recovery of baikiaea forest of southwestern Zambia from shifting cultivation and its implications for sustainable management

We studied the dominant species by age category of re-growth, and how subsistence forest use through shifting cultivation impacted on the floristic composition of baikiaea forest. For re-growth stands, variable sampling method was adopted in data collection while in mature stands, a sample plot size of 50 m 9 50 m was used at each of three study sites. Distribution of species by age categories showed that shrub, herb, and grass species were abundant in young re-growth stands while woody vegetation became common with increasing age of the re-growth stand. Hierarchical clustering for common woody species showed several species associations with age categories and also with relatively undisturbed woodland. Common regeneration mechanisms of key woody species were mainly by seed(48%), coppicing(40%) and root suckers(12%). The young re-growth stands of 6–10 and 11–15 years were characterized by b. petersiana, ***, Combretum zeyheri, and Guibourtia coleosperma. Older stands(16–20 years after abandonment) were characterized by b. plurijuga, Pterocarpus antunesii, and baphia massiensis, thus closely mimicking the relatively undisturbed forest whose dominant key woody species were b. plurijuga, P. antunesii, and G. coloesperma. There were many stems in younger stands(2–5 years) and fewer stems in older re-growth stands of 16–20 years after abandonment following shifting cultivation. Older regrowth stands(16–20 years) were similar to undisturbed stands(P \ 0.005). The similarity between re-growth stands of 16–20 years and control stands demonstrates the recovery of baikiaea forest over time after cessation of shifting agriculture. baikiaea forest requires some disturbance to perpetuate the dominant species and begins to show signs of recovery from shifting cultivation within16–20 years after abandonment. This is contrary to assertions that this ecosystem could take up to about 100 years for abandoned fields to begin showing signs of
