尽管近年来欧洲心血管病的死亡率有所下降,但每年约47%的死亡仍由心血管病引起。因此,心血管病的一级和二级预防十分重要。发生心血管事件的首要危险因素包括年龄、性别、吸烟、动脉高血压、血脂异常、糖尿病、肥胖及缺乏锻炼。

Peanut testa extracts enhance anticancer effect of cisplatin against human cholangiocarcinoma cells via modulation of histone deacetylase inhibitory activity

Objective:To investigate the effect of combination treatments of cisplatin and KK4 and ICG15042 peanut testa extracts against cholangiocarcinoma cells in ***:The growth inhibition,cell cycle arrest and apoptosis of cholangiocarcinoma cells were determined by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay and flow cytometry analysis,*** levels of proteins involved in apoptosis were assessed using Western blotting *** caspase activity was assessed using a colorimetric caspase activity ***:Cisplatin and peanut(KK4 and ICG15042)testa extracts inhibited the growth of cholangiocarcinoma cell lines(KKUM214 and KKU-100 cells)in a dose-and time-dependent *** combination treatments reduced cell viability and induced apoptosis of cholangiocarcinoma cells more efficiently than singledrug *** cell death synergistically mediated by cisplatin and peanut testa extracts was observed in KKU-M214 cells(combination index1.0).The combination treatments also increased the subG1 population and caused KKU-M214 cell cycle arrest at S and G2/M phases,which were the combined effects of cisplatin(S phase arrest)and peanut testa extracts(G2/M phase arrest).In addition,p ERK1/2,Ac-H3,Bcl-2 and proteins related to apoptosis,including Bax and caspases 3,8,9,exhibited enhanced expression in KKUM214 *** combination treatments caused down-regulation of p53,whereas the expression of p21 was fairly constant when compared with cisplatin single drug ***:Peanut testa extracts in combination with cisplatin synergistically reduce cell viability and induce apoptosis through stimulation of caspases 3,8 and 9 in KKU-M214 cells.

Disruption of bone homeostasis caused by metastatic osteolytic breast cancer cells increases inflammatory osteolysis and decreases bone formation,thereby predisposing patients to pathological fracture and cancer *** of osteoblast function induces skeletal diseases due to the disruption of bone *** observed increased activation of perk1/2 in osteolytic breast cancer cells and osteoblasts in human pathological specimens with aggressive osteolytic breast cancer *** confirmed that osteolytic breast cancers with high expression of perk1/2 disrupt bone homeostasis via osteoblastic ERK1/2 activation at the bone-breast cancer *** process of inflammatory osteolysis modulates ERK1/2 activation in osteoblasts and breast cancer cells through dominant-negative MEK1 expression and constitutively active MEK1 expression to promote cancer growth within ***,an FDA-approved MEK inhibitor,not only reduced breast cancer-induced bone destruction but also dramatically reduced cancer growth in bone by inhibiting the inflammatory skeletal *** together,these findings suggest that ERK1/2 activation in both breast cancer cells and osteoblasts is required for osteolytic breast cancer-induced inflammatory osteolysis and that ERK1/2 pathway inhibitors may represent a promising adjuvant therapy for patients with aggressive osteolytic breast cancers by altering the shared cancer and bone microenvironment.
