Study on biological characters of SGC7901 gastric cancer cell-dendritic cell fusion vaccines

AIM： To detect the biological characters of the SGC7901 gastric cancer cell-dendritic cell fusion ***： The suspending living SGC7901 gastric cancer cells and dendritic cells were induced to be fusioned by polyethylene glycol. Pure fusion cells were obtained by selective culture with the HAT/HT culture systems. The fusion cells were counted at different time points of culture and their growth curves were drawn to reflect their proliferative activities. The fusion cells were also cultured in culture medium to investigate whether they could grow into cell clones. MTT method was used to test the stimulating abilities of the fusion cells on T lymphocytes＇ proliferations. Moreover, the fusion cells were planted into nude mice to observe whether they could grow into new planted tumors in this kind of immunodeficiency ***： The fusion cells had weaker proliferative activity and clone abilities than their parental cells. When they were cultured, the counts of cells did not increase remarkably, nor could they grow into cell clones in culture medium. The fusion cells could not grow into new planted tumors after planted into nude mice. The stimulating abilities of the fusion cells on T lymphocytes＇ proliferations were remarkably increased than their parental dendritic cells. CONCLUSION： The SGC7901 gastric cancer cell-dendritic cell fusion vaccines have much weaker proliferative abilities than their parental cells, but they keep strong abilities to irritate the T lymphocytes and have no abilities to grow into new planted tumors in immunodeficiency animals. These are the biological basis for their antitumor biotherapies.

Postnatal weight gain in very low birth weight infants in Beijing and the risk of retinopathy of prematurity

AIMTo analyze the low weight gain (WG) from birth to 4 and 6wk of life to predict the development of retinopathy of prematurity (ROP) among very low birth weight (VLBW) preterm *** hundred and three newborns with VLBW were analyzed. Body weight measurements were recorded weekly. In all patients, the proportion of the WG was defined as the preterm weight measured at the 4th and 6th weeks of life minus the birth weight (BW) divided by the BW. Other risk factors for ROP were also *** gestational age and mean BW of the whole cohort were 29.56±1.44wk and 1270.58±176.18g respectively. WG proportion at 4wk postnatal age (18.89%±13.58%) were significantly lower in infants with ROP (P=0.003). WG proportion at 6wk was not different between ROP and no ROP group (42.48%±20.36% vs 46.43%±15.65% P=0.118). When all the other risk factors signiﬁcant for ROP were included in the logistic regression poor WG did not arise as an independent risk factor. Area under the ROC curve was 0.591 (95%CI: 0.515-0.666; P=0.016). For ROP, the best discriminative cutoff of 18.06% of the proportional WG at the 4th week over the BW, sensitivity and specificity values were 67.3% and 50.0% *** WG proportion in the first 4wk of life is maybe an additional predictor of ROP in very low BW infants. Preterm babies with low BW and low WG proportion should be followed closely for ROP.

AIM: To investigate the possibilities and advantages of laparoscopic hepatic left lateral lobectomy combined with fiber choledochoscopic exploration of the common bile duct compaired with traditional open ***: Laparoscopic hepatic left lateral lobectomy combined with fiber choledochoscopic exploration of the common bile duct and traditional open operation were performed in two groups of patients who had gallstones in the left lobe of liver and in the common bile duct. The hospitalization time, hospitalization costs, operation time, operative complications and post-operative liver functions of the two groups of patients were ***: The operation time and post-operative liver functions of the two groups of patients had no significant differences, while the hospitalization time, hospitalization costs and operative complications of the laparoscopic hepatic left lateral lobectomy combined with fiber choledochoscopic exploration in the common bile duct group were significantly lower than those in the traditional open operation ***: For patients with gallstones in the left lobe of liver and in the common bile duct, laparoscopic hepatic left lateral lobectomy combined with fiber choledochoscopic exploration of the common bile duct can significantly shorten the hospitalization time, reduce the hospitalization costs and the post-operative complications,without prolonging the operation time and bringing about more liver function damages compared with traditional open operation. This kind of operation has more advantages than traditional open operation.

Perineuronal nets increase inhibitory GABAergic currents during the critical period in rats

AIM: To investigate inhibitory γ-aminobutyric acid (GABA) ergic postsynaptic currents (IPSCs) and postsynaptic currents (PSCs) in layer IV of the rat visual cortex during the critical period and when plasticity was extended through dissolution of the perineuronal nets (PNNs). METHODS: We employed 24 normal Long-Evans rats to study GABA A-PSC characteristics of neurons within layer IV of the visual cortex during development. The animals were divided into six groups of four rats according to ages at recording: PW3 (P21 -23d), PW4 (P28 -30d), PW5 (P35-37d), PW6 (P42-44d), PW7 (P49-51d), and PW8 (56-58d). An additional 24 chondroitin sulfate proteoglycan (CSPG) degradation rats (also Long-Evans) were generated by making a pattern of injections of chondroitinase ABC (chABC) into the visual cortex 1 week prior to recording at PW3, PW4, PW5, PW6, PW7, and PW8. Immunohistochemistry was used to identify the effect of chABC injection on CSPGs. PSCs were detected with whole-cell patch recordings, and GABA A receptor-mediated IPSCs were pharmacologically isolated. RESULTS: IPSC peak current showed a strong rise in the age-matched control group, peaked at PW5 and were maintained at a roughly constant value thereafter. Although there was a small increase in peak current for the chABC group with age, the peak currents continued to decrease with the delayed highest value at PW6, resulting in significantly different week-by-week com-parison with normal development. IPSC decay time continued to increase until PW7 in the control group, while those in the chABC group were maintained at astable level after an initial increase at PW4. Compared with normal rats, the decay times recorded in the chABC rats were always shorter, which differed significantly at each age. We did not observe any differences in IPSC properties between the age-matched control and penicillinase (P-ase) group. Pathfinder algorithm(PFA)is a swarm intelligent optimization algorithm inspired by the collective activity behavior of swarm animals,imitating the leader in the population to guide followers in finding the best food *** algorithm has the characteristics of a simple structure and high ***,PFA faces challenges such as insufficient population diversity and susceptibility to local optima due to its inability to effectively balance the exploration and exploitation *** paper proposes an Ameliorated Pathfinder Algorithm called APFA to solve complex engineering optimization ***,a guidance mechanism based on multiple elite individuals is presented to enhance the global search capability of the ***,to improve the exploration efficiency of the algorithm,the Logistic chaos mapping is introduced to help the algorithm find more high-quality potential solutions while avoiding the worst ***,a comprehensive following strategy is designed to avoid the algorithm falling into local optima and further improve the convergence *** three strategies achieve an effective balance between exploration and exploitation overall,thus improving the optimization performance of the *** performance evaluation,APFA is validated by the CEC2022 benchmark test set and five engineering optimization problems,and compared with the state-of-the-art metaheuristic *** numerical experimental results demonstrated the superiority of APFA.

An Efficient Multi-objective Approach Based on Golden Jackal Search for Dynamic Economic Emission Dispatch

Dynamic Economic Emission Dispatch(DEED)aims to optimize control over fuel cost and pollution emission,two conflicting objectives,by scheduling the output power of various units at specific *** many methods well-performed on the DEED problem,most of them fail to achieve expected results in practice due to a lack of effective trade-off mechanisms between the convergence and diversity of non-dominated optimal dispatching *** address this issue,a new multi-objective solver called Multi-Objective Golden Jackal Optimization(MOGJO)algorithm is proposed to cope with the DEED *** proposed algorithm first stores non-dominated optimal solutions found so far into an ***,it chooses the best dispatching solution from the archive as the leader through a selection mechanism designed based on elite selection strategy and Euclidean distance index *** mechanism can guide the algorithm to search for better dispatching solutions in the direction of reducing fuel costs and pollutant ***,the basic golden jackal optimization algorithm has the drawback of insufficient search,which hinders its ability to effectively discover more Pareto *** this end,a non-linear control parameter based on the cosine function is introduced to enhance global exploration of the dispatching space,thus improving the efficiency of finding the optimal dispatching *** proposed MOGJO is evaluated on the latest CEC benchmark test functions,and its superiority over the state-of-the-art multi-objective optimizers is highlighted by performance ***,empirical results on 5-unit,10-unit,IEEE 30-bus,and 30-unit systems show that the MOGJO can provide competitive compromise scheduling solutions compared to published DEED ***,in the analysis of the Pareto dominance relationship and the Euclidean distance index,the optimal dispatching solutions provided by MOGJO are the closest to the ideal solutions for minimizing fuel costs

Thermal and mechanical behavior of casting copper alloy wheel during wheel and belt continuous casting process

To investigate the thermal and mechanical behavior of casting wheel,a two-dimensional thermoelastic-plastic finite element model was used to predict the temperature,stress and distortion distribution of the casting wheel during the wheel and belt continuous casting *** effects of grinding thickness and casting speed on the thermal and mechanical behaviors of the center of the hot face of the casting wheel were discussed in *** each rotation,the casting wheel passes through four different spray *** results show that the temperature distribution of the casting wheel in different spray zones is similar,the temperature of the hot face is the highest and the temperature reaches the peak in the spray zoneⅢ.The stress and distortion depend on the temperature distribution,and the maximum stress and distortion of the hot face are 358.2 MPa and 1.82 mm,*** temperature at the center of the hot face decreases with increasing grinding thickness and increases with increasing casting speed.

Impacts of central-Pacific El Niño and physical drivers on eastern Pacific bigeye tuna

Bigeye tuna Thunnus obesus is an important migratory species that forages deeply,and El Niño events highly influence its distribution in the eastern Pacific *** sea surface temperature is widely recognized as the main factor affecting bigeye tuna(BET)distribution during El Niño events,the roles of different types of El Niño and subsurface oceanic signals,such as ocean heat content and mixed layer depth,remain *** conducted A spatial-temporal analysis to investigate the relationship among BET distribution,El Niño events,and the underlying oceanic signals to address this knowledge *** used monthly purse seine fisheries data of BET in the eastern tropical Pacific Ocean(ETPO)from 1994 to 2012 and extracted the central-Pacific El Niño(CPEN)indices based on Niño 3 and Niño ***,we employed Explainable Artificial Intelligence(XAI)models to identify the main patterns and feature importance of the six environmental variables and used information flow analysis to determine the causality between the selected factors and BET ***,we analyzed Argo datasets to calculate the vertical,horizontal,and zonal mean temperature differences during CPEN and normal years to clarify the oceanic thermodynamic structure differences between the two types of *** findings reveal that BET distribution during the CPEN years is mainly driven by advection feedback of subsurface warmer thermal signals and vertically warmer habitats in the CPEN domain area,especially in high-yield fishing *** high frequency of CPEN events will likely lead to the westward shift of fisheries centers.

Numerical simulation of fluid flow and free surface fluctuations during wheel and belt casting process

The unstable fluid flow and severe free surface fluctuations in the wheel and belt caster can affect the quality of the cast *** lower level height tends to entrap inclusions in the molten *** the other hand,the higher level height makes the production process more dangerous due to the overflow of high temperature fluid from the mold.A computational model of the molten metal pouring process was *** transient fluid flow and free surface fluctuations behavior were calculated using the three-dimensional large eddy simulation model and the volume of fluid *** results show that the flow velocity of the main jet gradually decreases under the influence of the low kinetic energy fluid in the *** is an obvious oscillation in the tail of the jet,while the flow field is asymmetric in *** jet is closer to the inside radius side due to the Coanda effect,and there is a recirculation zone on the inside radius and the outside radius respectively,according to the 10 s time-averaged *** with the industrial observation and simulation results,the shape of the free surface is a wave that varies with *** addition,the free surface height is lowest and the flow velocity is highest in the region near the jet.

Contrasting physical mechanisms of yellowfin tuna fluctuations between the western and eastern Indian Ocean

As an economically critical pelagic migratory species,yellowfin tuna(Thunnus albacores,YFT)is very sensible to physical and environmental conditions,such as sea surface temperature(SST),ocean heat content(OHC),and the mixed layer depth(MLD).We investigated the impact of SST,OHC,and MLD on fluctuations of YFT catch in the western/eastern Indian Ocean using the long time series of 63-year environmental and YFT *** found that the impact of SST on YFT was heavily overestimated in the past,and MLD plays a more critical role in the YFT catch *** the MLD deepens(>34.8 m),SST was more influential in predicting the catches of YFT than OHC in the western Indian Ocean,and OHC was more critical to YFT than SST in the eastern Indian ***,when the MLD shallows(<34.8 m),MLD was more vital to predict the catch per unit effort(CPUE)of YFT than SST/OHC in the *** 2000,there was an asynchronous pattern of YFT CPUE induced by higher frequency variations and ocean hiatus of SST/OHC signals in the western and eastern Indian Oceans *** impact of the subsurface hiatus may induce the decrease of YFT in the eastern Indian *** above findings clarified a non-stationary relationship between the environmental factors and catches of YFT and provided new insights into variations in YFT abundance.
