利福平耐药与敏感肺结核患者痰液样本微生物菌群多样性对比分析

目的:利用16S高通量测序技术,探索利福平耐药与利福平敏感肺结核患者呼吸道菌群的差异。方法:收集北京市疾病预防控制中心(70份)和湖南省胸科医院(10份)确诊的肺结核患者的痰液(共80份)进行前瞻性研究,通过痰液样本GeneXpert MTB/RIF检测、靶向rpoB高通量测序(tNGS测序)和利福平药物敏感性试验,将80例患者分为利福平耐药组(RFP-R组,41例;利福平耐药决定区突变)和利福平敏感组(RFP-S组,39例;利福平耐药决定区无突变)。对痰液微生物16S V4区域进行高通量测序,测序结果进行生物信息学分析及统计分析。结果:RFP-R组和RFP-S组共获得41950224个原始16S rRNA序列。RFP-R组的平均操作分类单元(operational taxonomic units,OTU)为344个,RFP-S组的平均OTU为342个。厚壁菌门(47.72%)、拟杆菌门(22.36%)、变形菌门(14.67%)、放线菌门(7.57%)、梭杆菌门(6.46%)是肺结核患者痰液中检出的主要细菌门,占微生物群落的98.78%。与RFP-S组相比,RFP-R组梭杆菌门(7.531%vs.5.167%;t=2.218,P=0.030)和螺旋体门(0.596%vs.0.246%;t=2.128,P=0.038)相对丰度显著增加。与RFP-S组相比,RFP-R组颤杆菌克属(0.690%vs.0.326%;t=3.112,P=0.003)、产线菌属(0.135%vs.0.030%;t=2.171,P=0.035)、Allisonella菌属(0.046%vs.0.006%;t=3.237,P=0.002)、假单胞菌属(0.205%vs.0.054%;t=2.040,P=0.046)和密螺旋体属(0.601%vs.0.242%;t=2.155,P=0.036)相对丰度显著增加;二氧化碳噬纤维菌属(0.351%vs.0.772%;t=―2.044,P=0.046)和Pauljensenia菌属(2.314%vs.3.706%;t=―2.660,P=0.010)相对丰度显著减少。结论:利福平耐药与敏感结核分枝杆菌感染不影响肺结核患者呼吸道菌群的整体结构,但利福平耐药与敏感患者在2个门(梭杆菌门、螺旋体门)和7个属(颤杆菌克属、产线菌属、Allisonella菌属、假单胞菌属、密螺旋体属、二氧化碳噬纤维菌属、Pauljensenia菌属)的丰度中存在差异,其中二氧化碳噬纤维菌属和Pauljensenia菌属在利福平敏感患者中丰度较高,其余在利福平耐药患者中丰度较高。

Soft pneumatic actuators are one of the most promising actuation for soft robots,and great achievements have been *** it remains challenging to endow sensing capabilities to pneumatic actuators,especially for the sensing ability originating directly from the actuator ***,a self-sensing pneumatic torsional actuator(SPTA)is designed based on the electromagnetic induction effect and magnetically responsive *** SPTA can generate feedback voltage and current with the deformation,in which the sensing function comes from its inherent *** investigate the mechanical and electrical characteristics,an experimental platform and a finite element model are established,*** find that the torsion angle and output torque increase in nonlinear with the actuating *** maximum torsion angle is 66.35°,which is 84.34%of that for the actuator fabricated by pure *** maximum output torque(24.9 N mm)improves by 23.19%compared with the actuator made by pure *** regards the electrical characteristics,the maximum feedback voltage and current are 2.90μV and 29.50 nA when the SPTA is actuated by a pressure of−40 *** also demonstrate that the relationship between the torsion angle and the magnetic flux change is approximately ***,the number of turns of wires,magnetic powders contents,and magnetic direction on the feedback voltage and current are *** show that the feedback voltage and current can be enhanced by increasing the number of turns and magnetic powders *** envision that the SPTA would be promising for soft robots to realize their accuracy control and intelligentization.

This paper experimentally studied the effect of CO_(2) opposing multiple jets on the thermoacoustic instability and NO_(x) emissions in a lean-premixed model *** feasibility was verified from three variables:the CO_(2) jet flow rate,hole numbers,and hole diameters of the *** indicate that the control effect of thermoacoustic instability and NO_x emissions show a reverse trend with the increase of open area ratio on the whole,and the optimal jet flow rate range is 1-4 L/min with CO_(2) opposing multiple *** this flow rate range,the amplitude and frequency of the dynamic pressure and heat release signals CH~* basically decrease as the CO_(2) flow rate increases,which avoids high-frequency and high-amplitude thermoacoustic *** amplitude-damped ratio of dynamic pressure and CH*can reach as high as 98.75% and 93.64% with an optimal open area ratio of 3.72%.NO_(x) emissions also decrease as the jet flow rate increases,and the maximum suppression ratio can reach 68.14%.Besides,the flame shape changes from a steep inverted "V" to a more flat "M",and the flame length will become shorter with CO_(2) opposing multiple *** research achieved the synchronous control of thermoacoustic instability and NO_(x) emissions,which could be a design reference for constructing a safer and cleaner combustor.

Eliminating the effects of gravity and designing nonlinear energy sinks(NESs)that suppress vibration in the vertical direction is a challenging task with numerous damping *** this paper,the dynamic design of a vertical track nonlinear energy sink(VTNES)with zero linear stiffness in the vertical direction is proposed and realized for the first *** motion differential equations of the VTNES coupled with a linear oscillator(LO)are *** the strong nonlinearity considered of the VTNES,the steady-state response of the system is analyzed with the harmonic balance method(HBM),and the accuracy of the HBM is verified *** this basis,the VTNES prototype is manufactured,and its nonlinear stiffness is *** damping effect and dynamic characteristics of the VTNES are studied theoretically and *** results show that the VTNES has better damping effects when strong modulation responses(SMRs)***,even for small-amplitude vibration,the VTNES also has a good vibration suppression *** sum up,in order to suppress the vertical vibration,an NES is designed and developed,which can suppress the vertical vibration within certain ranges of the resonance frequency and the vibration intensity.
