Medulla Oblongata Mechanism of Inhibitory Effect of Thermal Stimulation to Nociceptive Colorectal Distention in Rats

Objective: To discuss mechanism of moxibustion(thermal stimulation) effect and best moxibustion stimulus ***: Experiments were performed on 48 male Sprague-Dawley rats. Unit discharges from individual single neuron were recorded extracellularly with glass-microelectrode in Subnucleus Reticularis Dorsalis(SRD). Visceral-intrusive stimulation is done by colorectal distension. Thermal stimulation with different temperature(40°C, 42°C, 44°C, 46°C, 48°C, 50°C, 52°C) and different stimulus area(diameter of circle : 1.0 cm, 1.5 cm, 2.0 cm, 2.5 cm, 3.0 cm, 3.5 cm, 4.0 cm) was applied around RN12 during nociceptive colorectal ***: SRD neurons could be activated by visceral stimulation within noxious range. Under low temperature of stimulus, especially under45°C of pain threshold to ordinary people, visceral nociceptive afferent facilitated thermal stimulus from the body surface. While after thermal stimulation reached a harmful degree, the thermal stimulus will inhibit visceral nociceptive afferent. Moreover, statistics show that the higher the temperature is, the smaller the size of stimulation area is needed, and they correlate with each other ***: Visceral nociception could be inhibited by somatic thermal stimulation with specific parameter at medulla level. According to our finding, best thermal stimulation temperature is around 48°C and the best size of stimulation area is around 3.14-7.07 cm2(with 2.0-3.0 cm diameter).

Arsenic methylation and microbial communities in paddy soils under alternating anoxic and oxic conditions

Arsenic(As)methylation in soils affects the environmental behavior of As,excessive accumulation of dimethylarsenate(DMA)in rice plants leads to straighthead disease and a serious drop in crop *** the mobility and transformation of methylated arsenic in redox-changing paddy fields is crucial for food ***,soils including unarsenic contaminated(N-As),low-arsenic(L-As),medium-arsenic(M-As),and high-arsenic(H-As)soils were incubated under continuous anoxic,continuous oxic,and consecutive anoxic/oxic treatments respectively,to profile arsenic methylating process and microbial species involved in the As *** anoxic-oxic(A-O)treatment,methylated arsenic was significantly increased once oxygen was introduced into the incubation *** methylated arsenic concentrations were up to 2-24 times higher than those in anoxic(A),oxic(O),and oxic-anoxic(O-A)treatments,under which arsenic was methylated slightly and then decreased in all four As concentration *** fact,the most plentiful arsenite S-adenosylmethionine methyltransferase genes(arsM)contributed to the increase in As ***(40.8%-62.4%),Firmicutes(3.5%-15.7%),and Desulfobacterota(5.3%-13.3%)were the major microorganisms related to this *** microbial increasedmarkedly and played more important roles after oxygen was introduced,indicating that they were potential keystone microbial groups for As methylation in the alternating anoxic(flooding)and oxic(drainage)*** novel findings provided newinsights into the reoxidation-driven arsenic methylation processes and the model could be used for further risk estimation in periodically flooded paddy fields.

Neural mechanism of acupuncture-modulated gastric motility

AIM: To investigate the acupuncture-modulated gastric motility and its underlying neural ***: Intragastric pressure and/or waves of gastric contraction in rats were recorded by intrapyloric balloon and changes of gastric motility induced by acupuncture stimulation were compared with the background activity before any stimulation. Gastro-vagal or splanchnic-sympathetic nerves were recorded or cut respectively for investigating the involvement of autonomic nerve pathways. Spinalization experiment was also ***: Acupuncture-stimulation by exciting Aδ and/or C afferent fibers, could only modulate gastric motility. Acupuncture-stimulation on fore- and hind-limbs evoked a moderate gastric motility followed by increased vagus discharges with unchanged sympathetic activity, while the same stimulus to the acupoints in abdomen resulted in reversed effects on gastric motility and autonomic nervous activities. The inhibitory gastric response was completely abolished by splanchnic denervation, but the facilitative gastric response to stimulation of acupoints in limbs was not influenced, which was opposite to the effect when vagotomy was performed. The similar depressive effects were produced by the stimulation at the acupoints homo-segmental to the gastric innervation in the animals with or without spinalization. However, the facilitation induced by the stimulation at the acupoints hetero-segmental to the gastric innervation was not observed in the spinalized ***: Facilitative effects of stimulating hetero-segmental acupoints are involved in the intact preparation of vagal nerves and spinal cord, while the inhibitory response induced by stimulating homo-segmental acupoints is involved in the intact preparation of sympathetic nerves. Only the acupuncture-stimulation with intensity over the threshold of Aδ and/or C afferent fibers can markedly modulate gastrointestinal motility.

Effective regularity in modulation on gastric motility induced by different acupoint stimulation

AIM： To investigate whether manual acupuncture at representative acupoints in different parts of the body can modulate responses of gastric motility in rats and regular effects in different acupoint stimulation. METHODS： The gastric motor activity of rats was recorded by the intrapyloric balloon. The changes of gastric motility induced by the stimulation were compared with the background activity in intragastric pressure and/or waves of gastric contraction recorded before any stimulation. Morphological study was also conducted by observing the Evans dye extravasation in the skin after mustard oil injection into the intragastric mucous membrane to certify cutaneous innervations of blue dots related to gastric segmental innervations. RESULTS： In all six rats that received mustard oil injections into intragastric mucosa, small blue dots appeared in the skin over the whole abdomen, but mainly in peri-midline upper- and middle- abdomen and middle-back, a few in thigh and groin. It may speculate that cutaneous innervations of blue dots have the same distribution as gastric segmental innervations. Acustimulation in acupoints of head-neck, four limbs, upper chest-dorsum and Iower-dorsum induced markedly augmentation of gastric motility （acupoints on headneck such as St-2： n = 16, 105.19 ± 1.36 vs 112.25 ± 2.02 and St-3： n = 14, 101.5 ± 1.75 vs 109.36 ± 1.8; acupoints on limbs such as Sp-6： n = 19, 100.74 ± 1.54 vs 110.26 ± 3.88; St-32： n = 17, 103.59 ± 1.64 vs 108.24 ± 2.41; St-36： n = 16, 104.81 ± 1.72 vs 110.81 ± 2.74 and U-11： n = 17, 106.47 ± 2.61 vs 114.77 ± 3.77, P 〈 0.05-0.001）. Catalytic production of high-energy-density spiro polycyclic jet fuel with biomass derivatives

High-energy-density（HED） fuel（*** used JP-10 and RJ-4）,featuring compact 3D polycyclic structure with high strain,is of critical importance for volume-limited military aircraft,since their high density and combustion heat can provide more propulsion *** reduce the reliance on petroleum source,it is highly desirable to develop renewable alternatives for the production of strained polycyclic HED fuel,but which remains a big challenge because of the inaccessibility caused by the high strain,We herein demonstrate a three-step catalytic route towards highly strained C17and C18spirofuel with biomass *** process includes catalytic aldol condensation of renewable cyclohexanone/cyclopentanone with benzaldehyde,catalytic spiro Diels-Alder（D-A） reaction of aldol adduct with isoprene,and catalytic *** key spiro D-A reaction is enabled by the catalysis of heterogeneous Lewis acidic ionic *** chloroaluminate IL,formed by benign urea and AICl3,exhibits good catalytic performance and reusability for this *** eventual hydrodeoxygenation（HDO）over Pd/C and H-Y produces strained tricyclic spirofuel with density>0.93 g/mL,combustion heat>41 MJ/L and freezing point <-40℃,which are better than the properties of tactical fuel ***,it is anticipated that the as-prepared renewable fuels have the potential to replace traditional petroleum-derived HED fuels.

Autophagy serves as a protective effect against inflammatory injury of oxidative stress in ARPE-19 cell

AIM:To test the effect of autophagy on inflammatory damage resulting from oxidative stress in adult retinal pigment epithelial cell line(ARPE-19).METHODS:ARPE-19 cells were pretreated with 200 and 600μmol/L hydrogen peroxide(H_(2)O_(2))at various time *** changes of cell morphology,cell viability,reactive oxygen species(ROS)level,autophagic activity,and the inflammatory cytokines(TNFα,IL-6,and TGFβ)were measured at baseline and after treatment with autophagy inducer rapamycin(Rapa)and suppressor wortmannin(Wort)or ***:The levels of ROS,cytokines(TNFα,IL-6,and TGFβ),and autophagic activity were significantly increased in ARPE-19 cells after pretreated with H2O2(all P<0.05)and IL-10 was significantly decreased(P<0.05).By upregulating autophagy,Rapa significantly reduced oxidative stress-induced secretion of pro-inflammatory factors(TNFα and IL-6)and ROS(all P<0.05),yet elevated the production of TGFβ(P<0.05).In contrast,suppression of autophagy through Wort or ATG5 knockdown reduced cell viability,increased cell apoptotic rate,and exacerbated the generation of ROS and inflammatory cytokines(TNFα,IL-6,and TGFβ;all P<0.05).CONCLUSION:Autophagy demonstrates a protective effect on ARPE-19 cell through mitigating oxidative damage and oxidative stress-induced inflammatory *** of autophagy may be a potential way for agerelated macular degeneration.
