Epidural electrical stimulation effectively restores locomotion function in rats with complete spinal cord injury

Cell cycle and complement inhibitors may be specific for treatment of spinal cord injury in aged and young mice: transcriptomic analyses

Previous studies have reported age-specific pathological and functional outcomes in young and aged patients suffering spinal cord injury,but the mechanisms remain poorly understood. In this study, we examined mice with spinal cord injury. Gene expression profiles from the Gene Expression Omnibus database （accession number GSE93561） were used, including spinal cord samples from 3 young injured mice （2–3-months old, induced by Impactor at Th9 level） and 3 control mice （2–3-months old, no treatment）, as well as 2 aged injured mice （15–18-months old, induced by Impactor at Th9 level） and 2 control mice （15–18-months old, no treatment）. Differentially expressed genes （DEGs） in spinal cord tissue from injured and control mice were identified using the Linear Models for Microarray data method,with a threshold of adjusted P 〈 0.05 and ｜logFC（fold change）｜〉 1.5. Protein–protein interaction networks were constructed using data from the STRING database, followed by module analysis by Cytoscape software to screen crucial genes. Kyoto encyclopedia of genes and genomes pathway and Gene Ontology enrichment analyses were performed to investigate the underlying functions of DEGs using Database for Annotation, Visualization and Integrated Discovery. Consequently, 1,604 and 1,153 DEGs were identified between injured and normal control mice in spinal cord tissue of aged and young mice, respectively. furthermore, a Venn diagram showed that 960 DEGs were shared among aged and young mice, while 644 and 193 DEGs were specific to aged and young mice, respectively. functional enrichment indicates that shared DEGs are involved in osteoclast differentiation, extracellular matrix–receptor interaction, nuclear factor-kappa B signaling pathway, and focal adhesion. Unique genes for aged and young injured groups were involved in the cell cycle （upregulation of PLK1） and complement （upregulation of C3） activation, respectively. Relationship between seismic structures and the diverse rupture processes of the 2023 Türkiye earthquake doublet

Recent geodetic and seismological observations of two major earthquakes in southeastern Türkiye in February 2023 have revealed complex rupture initiation,propagation,and segmentation along the East Anatolian Fault Zone(EAFZ)and surrounding ***,the role of upper crust structures along the EAFZ in determining the diverse rupture processes of this earthquake doublet remains *** further investigate this,we employed double-difference location and seismic tomography techniques to determine high-resolution seismic velocities(V_(P),V_(S))and Poisson’s ratio(σ)structures using a multiparameter joint tomographic *** dataset includes 100,833 high-quality source-receiver travel-time pairs of P-and *** find that the unique rupture processes of this earthquake doublet were primarily influenced by contrasting crustal seismic structures and localized geological *** M_(w)7.8 mainshock was initiated within a transitional edge zone characterized by a rigid part(asperity)of the seismogenic zone with sharp contrast variations in rock strength ranging from low to high along the *** comparison,the M_(w)7.6 rupture originated in a ductile belt featuring fluid saturation with low-VP,low-VS,and high-σvalues that extended parallel to the Cardak *** pronounced contrast structures observed along the former rupture can be attributed to the oblique collision system between the weakened section of the east Anatolian plateau and the brittle Arabian platform,while the latter rupture was initiated within the ductile structure associated with fluid intrusion caused by the northward subduction of the Cyprus slab and subsequent ***,the occurrence of the first earthquake(E1)serves to alleviate shear stress on the second earthquake(E2)fault,potentially impeding the initiation of an E2 *** the contrary,this event also significantly reduces the normal stress acting on the E2 fault due to a double left-lateral strike-slip system

Neuroprotective effect of rapamycin on spinal cord injury via activation of the Wnt/β-catenin signaling pathway

The Wnt/β-catenin signaling pathway plays a crucial role in neural development, axonal guid- ance, neuropathic pain remission and neuronal survival. In this study, we initially examined the effect of rapamycin on the Wnt/β-catenin signaling pathway after spinal cord iniury, by intraperitoneally injecting spinal cord injured rats with rapamycin over 2 days. Western blot analysis and immunofluorescence staining were used to detect the expression levels of β-catenin protein, caspase-3 protein and brain-derived neurotrophic factor protein, components of the Wnt/β-catenin signaling pathway. Rapamycin increased the levels of β-catenin and brain-derived neurotrophic factor in the injured spinal cord, improved the pathological morphology at the injury site, reduced the loss of motor neurons, and promoted motor functional recovery in rats after spinal cord injury. Our experimental fndings suggest that the neuroprotective effect of rapamycin intervention is mediated through activation of the Wnt/β-catenin signaling pathway after spinal cord injury.

A 2-year follow-up survey of 523 cases with peripheral nerve injuries caused by the earthquake in Wenchuan, China

We performed a 2-year follow-up survey of 523 patients with peripheral nerve injuries caused by the earthquake in Wenchuan, Sichuan Province, China. Nerve injuries were classiifed into three types： type I injuries were nerve transection injuries, type II injuries were nerve compression injuries, and type III injuries displayed no direct neurological dysfunction due to trauma. In this study, 31 patients had type I injuries involving 41 nerves, 419 had type II injuries involving 823 nerves, and 73 had type III injuries involving 150 nerves. Twenty-two patients had open tran-section nerve injury. The restoration of peripheral nerve function after different treatments was evaluated. Surgical decompression favorably affected nerve recovery. Physiotherapy was effective for type I and type II nerve injuries, but not substantially for type III nerve injury. Pharmaco-therapy had little effect on type II or type III nerve injuries. Targeted decompression surgery and physiotherapy contributed to the effective treatment of nerve transection and compression injuries. The Louisiana State University Health Sciences Center score for nerve injury severity de-clined with increasing duration of being trapped. In the ifrst year after treatment, the Louisiana State University Health Sciences Center score for grades 3 to 5 nerve injury increased by 28.2% to 81.8%. If scores were still poor （0 or 1） after a 1-year period of treatment, further treatment was not effective.

A passive sampler for synchronously measuring inorganic and organic pollutants in sediment porewater:Configuration and field application

In situ measurement of multiple pollutants coexisting in sediment porewater is an essential step in comprehensively assessing the bioavailability and risk of pollutants,but to date,this needs to be better *** this study,a passive sampler,consisting of an“I-shaped”supporting frame and inorganic/organic sampling units,incorporating equilibrium dialysis theory and kinetic/equilibrium sorption principle,was developed for the synchronous measurement of inorganic(e.g.,phosphorus and metal(loid)s)and organic pollutants(e.g.,parent and substituted PAHs).The equilibrium time and sampling rates were explored in laboratory tests to support in situ *** of pollutants in porewater within a vertical resolution of centimeters,i.e.,1 cm and 2 cm for inorganic and organic pollutants,respectively,were obtained by field deployment of the sampler for further estimation of diffusive fluxes across the sediment-water *** results suggested that the role of sediments for a specific pollutant may change(e.g.,from“sink”to“source”)during the sampling *** study demonstrated the feasibility of synchronous measurement of inorganic and organic pollutants in sediment porewater by the passive *** addition,it provided new insight for further investigation into the combined pollution effects of various pollutants in sediments.

Establishment of a rat model of chronic thoracolumbar cord compression with a flat plastic screw

Previous studies of animal models of chronic mechanical compression of the spinal cord have mainly focused on cervical and thoracic lesions, but few studies have investigated thoracolumbar injury. The specific pathophysiological mechanism of chronic thoracolumbar cord injury has not yet been elucidated. The purpose of this study was to improve animal models of chronic thoracolumbar cord compression using the progressive screw. A custom-designed flat plastic screw was implanted in the spinal cord between thoracic vertebrae 12 and lumbar 1 of rats. The screw was tightened one complete turn（0.5 mm） every 7 days for 4 weeks to create different levels of chronic spinal cord compression. Following insertion of the screw, there was a significant decline in motor function of the hind limbs, and severe stenosis of micro-computed tomography parameters in the spinal cord. Cortical somatosensory evoked potential amplitudes were reduced remarkably, and latencies were prolonged at 30 minutes after surgery. The loss of motor neurons in the gray matter was marked. Demyelination and cavitation were observed in the white matter. An appropriate rat model of chronic thoracolumbar cord compression was successfully created using the progressive screw compression method, which simulated spinal cord compression injury.

Effects of Calcium Sulfate Combined with Platelet-rich Plasma on Restoration of Long Bone Defect in Rabbits

Background：The treatment for long bone defects has been a hot topic in the field of regenerative *** study aimed to evaluate the therapeutic effects of calcium sulfate （CS） combined with platelet-rich plasma （PRP） on long bone defect ***：A radial bone defect model was constructed through an osteotomy using New Zealand *** rabbits were randomly divided into four groups （n =10 in each group）：a CS combined with PRP （CS-PRP） group,a CS group,a PRP group,and a positive （recombinant human bone morphogenetic protein-2） control *** was prepared from autologous blood using a two-step centrifugation ***-PRP was obtained by mixing hemihydrate CS with *** and histologic micrographs were *** percentage of bone regenerated bone area in each rabbit was calculated at 10 ***-way analysis of variance was performed in this ***：The radiographs and histologic micrographs showed bone restoration in the CS-PRP and positive control groups,while nonunion was observed in the CS and PRP *** percentages of bone regenerated bone area in the CS-PRP （84.60 ± 2.87%） and positive control （52.21 ± 4.53%） groups were significantly greater than those in the CS group （12.34 ± 2.17%） and PRP group （16.52 ± 4.22%）（P 〈 0.001）.In addition,the bone strength of CS-PRP group （43.l 0 ± 4.10%） was significantly greater than that of the CS group （20.10 ± 3.70%） or PRP group （25.10 ± 2.10%）（P 〈 0.001）.Conclusion：CS-PRP functions as an effective treatment for long bone defects through stimulating bone regeneration and enhancing new bone strength.

Microwave irradiation-induced alterations in physicochemical properties and methane adsorption capability of coals:An experimental study using carbon molecular sieve

In order to comprehend the applicability of microwave irradiation for recovering coalbed methane,it is necessary to evaluate the microwave irradiation-induced alterations in coals with varying levels of *** this work,the carbon molecular sieve combined with KMnO_(4)oxidation was selected to fabricate carbon molecular sieve with diverse oxidation degrees,which can serve as model substances toward ***,the microwave irradiation dependences of pores,functional groups,and highpressure methane adsorption characteristics of model substances were *** results indicated that microwave irradiation causes rearrangement of oxygen-containing functional groups,which could block the micropores with a size of 0.40-0.60 nm in carbon molecular sieve;meanwhile,naphthalene and phenanthrene generated by macro-molecular structure pyrolysis due to microwave irradiation could block the micropores with a size of 0.70-0.90 *** alterations in micropore structure weaken the saturated methane adsorption capacity of oxidized carbon molecular sieve by 2.91%-23.28%,suggesting that microwave irradiation could promote methane ***,the increased mesopores found for oxidized carbon molecular sieve after microwave irradiation could benefit CH4 *** summary,the oxidized carbon molecular sieve can act as model substances toward coals with different ***,microwave irradiation is a promising technology to enhance coalbed methane recovery.
