We have applied Raster Image Correlation Spectroscopy (RICS) technique to characterize the dynamics of protein 53 (p53) in living cells before and after the treatment with DNA damaging agents. HeLa cells expressing Green Fluores-cent Protein (GFP) tagged p53 were incubated with and without DNA damaging agents, cisplatin or eptoposide, which are widely used as chemotherapeutic drugs. Then, the diffusion coefficient of GFP-p53 was determined by RICS and it was significantly reduced after the drug treatment while that of the one without drug treatment was not. It is suggested that the drugs induced the interaction of p53 with either other proteins or DNA. Together, our results demonstrated that RICS is able to detect the protein dynamics which may be associated with protein-protein or protein-DNA interactions in living cells and it may be useful for the drug screening.

Geospatial simulation models can help us understand the dynamic aspects of Digital Earth.To implement high-performance simulation models for complex geospatial problems,grid computing and cloud computing are two promising computational frameworks.This research compares the benefits and drawbacks of both in Web-based frameworks by testing a parallel Geographic Information System(GIS)simulation model(Schelling’s residential segregation model).The parallel GIS simulation model was tested on XSEDE(a representative grid computing platform)and Amazon EC2(a representative cloud computing platform).The test results demonstrate that cloud computing platforms can provide almost the same parallel computing capability as high-end grid computing frameworks.However,cloud computing resources are more accessible to individual scientists,easier to request and set up,and have more scalable software architecture for on-demand and dedicated Web services.These advantages may attract more geospatial scientists to utilize cloud computing for the development of Digital Earth simulation models in the future.

In a composite medium that contains close-to-touching inclusions, the pointwise values of the gradient of the voltage potential may blow up as the distance S between some inclusions tends to 0 and as the conductivity contrast degenerates. In a recent paper [9], we showed that the blow-up rate of the gradient is related to how the eigenvalues of the associated Neumann-Poincare operator converge to ±1/2 as δ→ 0, and on the regularity of the contact. Here, we consider two connected 2-D inclusions, at a distance 5 〉 0 from each other. When δ=0, the contact between the inclusions is of order m 〉 2. We numerically determine the asymptotic behavior of the first eigenvalue of the Neumann- Poincare operator, in terms of 5 and rn, and we check that we recover the estimates obtained in [10].

Many scholars have argued that the importance of geographic proximity in human interactions has been diminished by the use of the Internet,while others disagree with this argument.Studies have noted the distance decay effect in both cyberspace and real space,showing that interactions occur with an inverse relationship between the number of interactions and the distance between the locations of the interactors.However,these studies rarely provide strong evidence to show the influence of distance on interactions in cyberspace,nor do they quantify the differences in the amount of friction of distance between cyberspace and real space.To fill this gap,this study used massive amounts of social media data(Twitter)to compare the influence of distance decay on human interactions between cyberspace and real space in a quantitative manner.To estimate the distance decay effect in both cyberspace and real space,the distance decay function of interactions in each space was modeled.Estimating the distance decay in cyberspace in this study can help predict the degree of information flow across space through social media.Measuring how far ideas can be diffused through social media is useful for users of location-based services,policy advocates,public health officials,and political campaigners.

The spatial diffusion of information is a process governed by the flow of interpersonal communication.The emergence of the Internet and especially social media platforms has reshaped this process and previous research has studied how online social networks contribute to the diffusion of information.Understanding such processes can help devise methods to maximize or control the reach of information or even identify upcoming events and social movements.Yet activities in cyberspace are still confined to physical locations and this geographic connection tends to be overlooked.In this research,we focus on geographic regions instead of individuals and study how the underlying hierarchical structure of regions relates to their response to the information.We examined the top 30 populated cities and metropolitan areas in the U.S.and retrieved Twitter data related to two selected topics from these regions,the 2015 Nepal Earthquake and the#JesuisCharlie hashtag in response to the Paris attacks on the Charlie Hebdo offices.We analyzed the similarity among regions of their response using multiple statistical methods and three urban classifications.Our results indicate that the diffusion of information is impacted by the hierarchy of urban regions and that the Twitter responses act more similar when the populated regions are positioned at the same level in the urban hierarchy.

Endoscopic salvage therapy after failed biliary cannulation using advanced techniques:A concise review

Therapeutic endoscopic retrograde cholangiopancreatography(ERCP)begins with successful biliary cannulation.However,it is not always be successful.The failure of the initial ERCP is attributed to two main aspects:the papilla/biliary orifice is endoscopically accessible,or it is inaccessible.When the papilla/biliary orifice is accessible,bile duct cannulation failure can occur even with advanced cannulation techniques,including double guidewire techniques,transpancreatic sphincterotomy,needle-knife precut papillotomy,or fistulotomy.There is currently no consensus on the next steps of treatment in this setting.Therefore,this review aims to propose and discuss potential endoscopic options for patients who have failed ERCP due to difficult bile duct cannulation.These options include interval ERCP,percutaneous-transhepatic-endoscopic rendezvous procedures(PTE-RV),and endoscopic ultrasound-assisted rendezvous procedures(EUS-RV).The overall success rate for interval ERCP was 76.3%(68%-79% between studies),and the overall adverse event rate was 7.5%(0-15.9% between studies).The overall success rate for PTE-RV was 88.7%(80.4%-100%between studies),and the overall adverse event rate was 13.2%(4.9%-19.2% between studies).For EUS-RV,the overall success rate was 82%-86.1%,and the overall adverse event rate was 13%-15.6%.Because interval ERCP has an acceptably high success rate and lower adverse event rate and does not require additional expertise,facilities,or other specialists,it can be considered the first choice for salvage therapy.EUS-RV can also be considered if local experts are available.For patients in urgent need of biliary drainage,PTE-RV should be considered.

AIM:To compare the hemostatic efficacy between epinephrine injection alone and epinephrine injection combined with thermotherapy for delayed post-endoscopic sphincterotomy (ES) bleeding.METHODS: Cases with delayed post-ES bleeding undergoing epinephrine injection alone (epinephrine injection group, n=26) or epinephrine combined with thermotherapy (combination therapy group, n=33) in our institution between 1999 and 2007 were retrospectively investigated. The main outcome measurements wereRESULTS: The initial hemostatic efficacy was 96.2% for epinephrine injection alone and 100% for combination therapy (P=0.44). There were four patients with re-bleeding in each group (16.0% vs 12.1%, P=0.72). There was only one complication of pancreatitis from the combination therapy group. Three patients (11.5%) in the epinephrine injection group and one patient (3%) in the combination therapy group required angiographic embolization or surgery (P=0.31). The total number of blood transfusions was not significantly different between the two groups (3.5±4.6 U vs 3.5±4.5 U, P=0.94). There was no bleeding-related death in either group.CONCLUSION: Epinephrine injection alone is as effective as epinephrine injection combined with thermotherapy for the management of delayed post-ES bleeding.

AIM:To evaluate the effect of double balloon endoscope(DBE)on the endoscopic retrograde cholangio-pancreatography(ERCP)success rate in patients with a history of BillrothⅡ(BⅡ)gastrectomy.METHODS:From April 2006 to March 2007,32 patients with a BⅡgastrectomy underwent 34 ERCP attempts.In all cases,the ERCP procedures were started using a duodenoscope.If intubation of the afferent loop or reaching the papilla failed,we changed to DBE for the ERCP procedure(DBE-ERCP).We assessed the success rate of afferent loop intubation,reaching the major papilla,selective cannulation,possibility of therapeutic approaches,procedure-related complications,and the overall success rate.RESULTS:Among the 32 patients with a history of BⅡgastrectomy,the duodenoscope was successfully passed up to the papilla in 22 patients(69%),and cannulation was successfully performed in 20 patients(63%).Six patients(2 with failure in afferent loop intubation and 4 with failure in reaching the papilla)underwent DBE-ERCP.The DBE reached the papilla in all the 6 patients(100%)and selective cannulation was successful in 5 patients(83%).Four patients(67%)who had common bile duct stones were successfully treated.One patient underwent diagnostic ERCP only and the other one,in whom selective cannulation failed,was diagnosed with papilla cancer proven by biopsy.There were no complications related to the DBE.The overall ERCP success rate increased to 88%(28/32).CONCLUSION:The overall ERCP success rate increases with DBE in patients with a previous BⅡgastrectomy.

BACKGROUND Heart rate variability(HRV) and pulse-wave velocity(PWV),indicators of cardiac function,are altered in patients with spinal cord injury(SCI),suggesting that autonomic cardiac function and arterial stiffness may underlie the high risk of cardiovascular complications in these patients.No study has simultaneously investigated HRV and PWV in the same patients.AIM To evaluate cardiovascular complications in SCI patients by comparing HRV and PWV between patients with and without SCI.METHODS In this cross-sectional pilot study,patients with(n = 60) and without SCI(n = 60) were recruited from December 7,2019 to January 21,2020.Each participant received a five-minute assessment of HRV and the cardiovascular system using the Medicore HRV Analyzer SA-3000 P.Differences in HRV and PWV parameters between participants with and without SCI were statistically examined.RESULTS We observed a significant difference between participants with and without SCI with respect to the standard deviation of all normal-to-normal intervals,square root of the mean sum of squared successive risk ratio interval differences,physical stress index,total power,very-low frequency,low frequency,high frequency,and arterial elasticity.CONCLUSION Patients with SCI have weaker sympathetic and parasympathetic activity as well as lower arterial elasticity compared to those without,suggesting that SCI may increase cardiac function loading.

Understanding and detecting the intended meaning in social media is challenging because social media messages contain varieties of noise and chaos that are irrelevant to the themes of interests.For example,conventional supervised classification approaches would produce inconsistent solutions to detecting and clarifying whether any given Twitter message is really about a wildfire event.Consequently,a renovated workflow was designed and implemented.The workflow consists of four sequential procedures:(1)Apply the latent semantic analysis and cosine similarity calculation to examine the similarity between Twitter messages;(2)Apply Affinity Propagation to identify exemplars of Twitter messages;(3)Apply the cosine similarity calculation again to automatically match the exemplars to known training results,and(4)Apply accumulative exemplars to classify Twitter messages using a support vector machine approach.The overall correction ratio was over 90%when a series of ongoing and historical wildfire events were examined.
