Background:Skeletal muscle energetics decline with age,and physical activity(PA)has been shown to offset these declines in older ***,many studies reporting these effects were based on self-reported PA or structured exercise ***,we examined the associations of accelerometry-measured and self-reported PA and sedentary behavior(SB)with skeletal muscle energetics and explored the extent to which PA and sedentary behavior would attenuate the associations of age with muscle ***:As part of the Study of muscle,mobility and Aging,enrolled older adults(n=879),810(age=76.4±5.0 years old,mean±SD;58%women)had maximal muscle oxidative capacity measured ex vivo via high-resolution re spirometry of permeabilized myofibers(maximal oxidative phosphorylation(maxOXPHOS))and in vivo by ^(31)phosphorus magnetic resonance spectroscopy(maximal adenosine triphosphate(ATP_(max))).Accelerometry-measured sedentary behavior,light activity,and moderate-to-vigorous PA(mVPA)were assessed using a wrist-worn ActiGraph GT9X over 7 ***-reported sedentary behavior,mVPA,and all PA were assessed with the Community Healthy Activities model Program for Seniors(CHAmPS)*** regression models with progressive covariate adjustments evaluated the associations of sedentary behavior and PA with muscle energetics,as well as the attenuation of the age/muscle energetics association by mVP A and sedentary *** a sensitivity analysis,we also examined activPAL-measured daily step count and time spent in sedentary behavior and their associations with muscle ***:Every 30 min/day more of ActiGraph-measured mVPA was associated with 0.65 pmol/(s×mg)higher maxOXPHOS and 0.012 mm/s higher ATP_(max)after adjusting for age,site/technician,and sex(pmaxOXPHOS or ATP_(max).meanwhile,every 30 min/day spent in ActiGraph-measured sedentary behavior was associated with 0.39 pmol/s×mg lower maxOXPHOS and0.

Human mesenchymal stromal cells(hmSCs)are mechanically sensitive undergoing phenotypic alterations when subjected to shear stress,cell aggregation,and substrate changes encountered in 3D dynamic bioreactor ***,little is known about how bioreactor microenvironment affects the secretion and cargo profiles of hmSC-derived extracellular vesicles(EVs)including the subset,“exosomes”,which contain therapeutic proteins,nucleic acids,and lipids from the parent *** this study,bone marrow-derived hmSCs were expanded on 3D Synthemax II microcarriers in the PBS mini 0.1L Vertical-Wheel bioreactor system under variable shear stress levels at 25,40,and 64 RPm(0.1-0.3 dyn/cm^(2)).The bioreactor system promotes EV secretion from hmSCs by 2.5-fold and upregulates the expression of EV biogenesis markers and glycolysis genes compared to the static 2D *** microRNA cargo was also altered in the EVs from bioreactor culture including the upregulation of miR-10,19a,19b,21,132,and *** protein cargo was characterized by proteomics analysis,showing upregulation of metabolic,autophagy and ROS-related proteins comparing with 2D cultured *** addition,the scalability of the Vertical-Wheel bioreactor system was demonstrated in a 0.5L bioreactor,showing similar or better hmSC-EV secretion and cargo content compared to the 0.1L *** study advances our understanding of bio-manufacturing of stem cell-derived EVs for applications in cell-free therapy towards treating neurological disorders such as ischemic stroke,Alzheimer’s disease,and multiple sclerosis.

In this work, the influence of phenol-enriched olive mill wastewater (OmWW) extract on hair growth was investigated m>in vitrom> on human follicle dermal papilla cells. OmWW has already shown great potential for use in skincare products, and its high polyphenol content is predestined to have a positive effect on hair growth. The studies included caffeine, a positive modulator of hair growth, and dihydrotestosterone, which causes hair loss m>in vivom>, as controls. The impact of the investigated compounds on hair growth was evaluated by studies on cell viability and proliferation, the release of growth factors (insulin-like growth factor-1 and vascular endothelial growth factor), and the reduction of reactive oxygen species formation. OmWW showed a positive influence on the proliferation of the human follicle dermal papilla cells. moreover, the extract leads to a significantly increased secretion of insulin-like growth factor-1, and a considerable reduction in reactive oxygen species formation was observed. Overall, our results show that the investigated phenol-enriched OmWW extract is a promising ingredient for hair care to improve hair growth, prevent hair loss due to oxidative stress and maintain a healthy scalp.

Electrospun Food-Grade Ultrafine Fibers from Pectin and Pullulan Blends

Electrospinning was used to produce ultrafine fibers and fibrous mats from aqueous solutions containing two edible polysaccharides: pectin (PEC) and pullulan (PUL). The process excluded the use of a synthetic carrier polymer or non-aqueous solvents thus maintaining the food-grade status of the components. The inclusion of PUL reduced the surface tension and electric conductivity of pectin solution, and promoted molecular entanglement between PEC and PUL as confirmed by rheological analysis. The spinnability of either polysaccharide was promoted by inclusion of the other. Cross-linking PEC networks were obtained by soaking the fibrous mats in successive ca2+ solution. The fibrous mats can be used to carry bioactive compounds as demonstrated using probiotic bacteria Lactobacillus rhamnosus GG (LGG) as a model bioactive compound. The research is significant for the development of food products with unique textures and functionalities.

measuring the antioxidative activities of Queso Fresco after post-packaging high-pressure processing

Some milk-associated proteins are known to be nutritionally valuable and form bioactive peptides that exhibit activity against hypertension and oxidative stress. Consumption of cheeses, such as the popular Hispanic-style cheese Queso Fresco (QF), may be a vehicle for delivery of these milk-associated peptides. This paper describes the effects of high-pressure processing (HPP) on the antioxidative activity (ORAC- FL value) of water-soluble proteins extracted from QF samples. QFs were manufactured according to a commercial-make procedure using pasteurized, homogenized milk, without added starter cultures. The cheese was cut into 45 × 45 × 150 mm3 blocks, double packaged in vacuum bags, and received the following HPP treatments: 200, 400, or 600 mPa for either 0, 5, 10, or 20 min, with warming to an internal temperature of either 22℃ or 40℃ prior to HPP treatment. Results show that the core temperature of the cheese during HPP directly affects the ORAC-FL value. The activities of the lower temperature cheeses are independent of time and pressure, and have a median ORAC-FL value of 27 trolox equivalents (TE). The higher temperature cheeses have higher ORAC-FL values ranging from 21.5 to 96.0 TE;the highest activity corresponded to the cheese held at 400 mPa for the longest time under pressure (20 min). The 600 mPa cheeses increase in activity with increasing time under pressure, but are less active than the control cheese. These results indicate that processing temperature and pressure are important factors in the antioxidative activity of these QF samples and further understanding of the roles of these variables may lead to the manufacture of healthier and more nutritious cheeses and dairy products.

目的　分析乳腺浸润性导管癌的临床特点和预后因素。方法　1 544 例浸润性导管癌全部为九十年代新发病例。采用观察生存率、死亡危险比、Log rank检验和拟合多因素Cox比例风险回归模型等进行单因素和多因素分析,评估各种预后因素与乳腺癌的联系强度。结果　浸润性导管癌占女性乳腺癌的82.5%,腋淋巴结转移率44.2%。外上象限发病最多,占47.7%。肿块位中央区,腋淋巴结阳性率最高,占61.7%。Ⅰ、Ⅱ、Ⅲ、Ⅳ期病例分别占30.0%,59.4%,9.4%,1.2%;5年和10年生存率分别为94.9%,78.1%;48.1%,8.6%和87.2%,67.1%;31.2%,0.0%。单因素分析:发病年龄、肿块大小、腋淋巴结转移数、TNm分期、治疗方式与生存率有关;ER、PR、Her- 2/neu、手术类型与生存率无关。多因素分析显示:发病年龄、腋淋巴结转移数、治疗方式与生存率有关。结论　腋淋巴结状况是制定治疗方案和评估预后的最主要指标。50 岁以下发病的患者以及接受综合治疗的患者生存率较高。九十年代与八十年代相比,上海市区女性乳腺癌的诊治水平有了较大的提高。
