Subcutaneous Model for the Study of Dengue Virus Infection in Immune Competent Mice

Various mouse models to study dengue have been described by different authors, some of them using immunodeficient or some using humanized mice. Our group reported previously a deadly murine model, which used the intracranial inoculum of highly virulent Dengue virus (DENV) in immune competent mouse. Here we present a model of immune competent mouse (C57BL/6), infected subcutaneously by the same highly virulent DENV (DENV3 genotype I). In this immunocompetent systemic mice model, the cytokine levels and hematological parameters such as total and differential leukocyte and platelets counts, together with weight loss, were considered important monitoring parameters, allowing a better understanding of the systemic human disease. Mice were inoculated subcutaneously and evaluated by the percentage weight variation as well as the clinical signs. Hematological parameters and cytokines levels were measured and viral titration in brain tissue or serum neutralization was performed to confirm mice infection. The subcutaneously DENV inoculated mice showed weight loss after infection, but they did not show any other clinical signs. The leukocytes and platelets decreased after subcutaneous inoculation. The cytokines TNF alpha and IFN gamma increased after infection in mice. The subcutaneous model provided scope for improved understanding of the dengue pathogenesis, as well as possible mechanism for protection to subsequent mouse infected by intracranial route in mice. This model could be used to study the vertebrate immune response and evaluation of drugs or vaccine against dengue virus.

Characterization and Comparison of Microbial Soil Diversity in Two Andean Peatlands in Different States of Conservation-Vega Tocorpuri

Cerro Tocorpuri, belongs to the II region of Chile, in San pedro de Atacama, on the border of Chile-Bolivia. The presence of a more or less constant supply of water conditions the existence of characteristic vegetation systems known as bogs (bofedales, vegas and marshes). These wetlands have a cultural, environmental and economic social importance. As a result of the exploitation of aquatic rights, peatlands began to dry up with the consequent loss of natural resources and damage to ancestral rights, and natural resources. The activities of microorganisms in wetlands play an important role in biogeochemical processes. The interaction between microbial diversity and soil, influences to the ability of the ecosystem to recover from stress (resilience). In the present work, the soil characteristics and the associated microbial biodiversity were studied, comparing samples of active and deteriorated peatland. It was seen that the loss of water causes great changes in the physical-chemical characteristics of the soil, which leads to a modification of the microbiota Proteobacteria decreased by 18% in deteriorated peatlands, which are evident more sensible to extreme conditions while Acidobacteria, Actinobacteria increased in these sample showing a better adaptation to the change of conditions. In view of the fact that high Andean Peatlands are exposed to increasing environmental impact, this preliminary comparative study of pristine and altered soil could guide the research directed to recovery of dead peatlands strategies.

Re-bleeding events in patients with obscure gastrointestinal bleeding after negative capsule endoscopy

AIM: To investigate long-term re-bleeding events after a negative capsule endoscopy in patients with obscure gastrointestinal bleeding(OGIB) and the risk factors associated with the ***: Patients referred to Hospital Egas Moniz(Lisboa, Portugal) between January 2006 and October 2012 with OGIB and a negative capsule endoscopy were retrospectively analyzed. The following study variables were included: demographic data, comorbidities, bleeding-related drug use, hemoglobin level, indication for capsule endoscopy, post procedure details, work-up and follow-up. Re-bleeding rates and associated factors were assessed using a Cox proportional hazard analysis. The Kaplan-Meier method was used to estimate the cumulative incidence of re-bleeding at 1, 3 and 5 years, and the differences between factors were ***: The study population consisted of 640 patients referred for OGIB investigation. Wireless capsule endoscopy was deemed negative in 113 patients(17.7%). A total of 64.6% of the population was female, and the median age was 69 years. The median follow-up was forty-eight months(interquartile range 24-60). Re-bleeding occurred in 27.4% of the cases. The median time to re-bleeding was fifteen months(interquartile range 2-33). In 22.6%(n = 7) of the population, small-bowel angiodysplasia was identified as the culprit lesion. A univariate analysis showed that age > 65 years old, chronic kidney disease, aortic stenosis, anticoagulant use and overt OGIB were risk factors for re-bleeding; however, on a multivariate analysis, there were no risk factors for re-bleeding. The cumulative risk of re-bleeding at 1, 3 and 5 years of follow-up was 12.9%, 25.6% and 31.5%, *** who presented with overt OGIB tended to rebleed sooner(median time for re-bleeding: 8.5 mo vs 22 mo).CONCLUSION: Patients with OGIB despite a negative capsule endoscopy have a significant re-bleeding risk; therefore, these patients require an extended follow-up strategy.

Influence of Cu9Al4 Phase on Nanomechanical Behavior of Cu-14Al-4Ni-xTi Shape Memory Alloy Rapidly Solidified

This paper aims to verify the Cu9Al4 phase influence on the nanomechanical behavior of the Cu-14Al-4Ni-xTi alloy obtained by rapid solidification with addition of different amounts of *** the Scanning Electron Microscopy(SEM),Atomic Force Microscopy(AFM),Energy Dispersion Spectroscopy(EDS)and X-Ray Diffraction(XRD),it was possible to perform the samples’microstructural *** addition,the reduction of the Cu9Al4 phase precipitation and the X-phase appearance were verified according to the increase of the titanium percentage *** nanomechanical behavior was evaluated by nanoindentation tests,which showed a significant decrease of the elastic modules and an increase of the Poisson coefficient’s according to the titanium *** research establishes that the reduction of Cu9Al4 phase implies on the increase of the capacity to dissipate ***,the high damping capacity combined with the X-phase presence increases the super elasticity and the alloy ductility.

Discrepancies between the responses to skin prick test to food and respiratory antigens in two subtypes of patients with irritable bowel syndrome

AIM:To compare the response to skin prick tests (SPTs) to food antigens (FAs) and inhalant allergens (IAs) in patients with two subtypes of irritable bowel syndrome (IBS) and healthy controls. METHODS:We compared the results of SPTs for IAs and FAs in 87 volunteers divided into three groups:diarrhea predominant IBS (D-IBS) GroupⅠ(n = 19), constipation predominant IBS (C-IBS) Group Ⅱ (n = 17), and normal controls Group Ⅲ (n = 51). RESULTS:Of the 285 tests (171 for FAs and 114 for IAs) performed in GroupⅠwe obtained 45 (26.3%) positive responses for FA and 23 (20.1%) for IA. Of the 153 tests for FA in Group Ⅱ, we obtained 66 (20.1%) positive responses, and of the 102 tests for IA, we obtained 20 (19.6%) positive responses. Of the 459 tests for FA performed in Group Ⅲ, we obtained 39 (84%) positive responses, and of the 306 for IA, we obtained 52 (16.9%) positive responses. The numbers of positive responses were not significantly different between the three groups, but in the D-IBS group, the number of SPTFA responses differed significantly from those for the other two groups (P < 0.01). CONCLUSION:Despite the small number of cases studied, the higher reactivity to FAs in GroupⅠcompared to Groups Ⅱ and Ⅲ adds new information, and suggests the presence of a possible alteration in intestinal epithelial function.

Use of a selective enzymatic debridement agent(Nexobrid■)for wound management:Learning curve

AIM To evaluate the efficacy of Nexobrid~? in the initial management of burns and lessons learned with the *** From January 27^(th) 2015 until January 25^(th) 2016, 25 patients aged between 18-94 years old with deep partial and full thickness burns were treated with Nexobrid~? covering 1%-30% of their total body surface area(TBSA). The debridement was applied in the first 96 h post-injury following the protocol suggested for Nexobrid~?. In patients with burns of more than 15% TBSA a second application of Nexobrid~? was performed. After the removal of the product-4 h post application and after a 2 h period of wet dressing-we used several products to cover the wound like Suprathel~?, Biobrane~?, Mepitel~? with wet dressing, silver sulphadiazine 1% cream, and in some cases even autografts. We treated patients with inhalation injury as well. All the procedures were done under deep sedation, regional blocks in extremities or general anaesthesia in the intensive care unit room or in the operating *** After these first 25 cases, we have observed that patients with partial thickness burns treated with Nexobrid~?, experienced great benefits in the reduction of the need for autografting compared with the standard of care. This isbecause after selective enzymatic debriding of the burn scar we can distinguish different wound beds, which can coexist in the same patient, and we also managed to associate each one to its ability to epithelize. In major burns, besides the improvement in wound healing, we observed an important improvement in their general state. This may be because SIRS significantly improved through a bloodless debridement of necrotic tissue, decreasing the requirements of vasoactive drugs and fluid resuscitation. Circumferential burns also benefited from enzymatic debridement, observing a decrease in the number of compartment syndromes and the need for escharotomies. Lead and Cadmium Distribution in Tubes of Galvanized Steel by Hot-Dip Used for Drinking Water Supply

The main objective of this work was to evaluate the presence of lead and cadmium in galvanized steel tubes by hot-dip used exclusively for drinking water *** this research,samples of galvanized tubes were removed from walls of residential buildings with 30 years of *** the process of hot galvanizing,the manufacturing standards allow or for low cost conveniences,and the use of primary zinc ingot with 1.6%metallic impurities,such as lead and cadmium,is allowed,enabling the incorporation of these metals into the zinc coating deposited on the carbon *** associated corrosion of these tubes can release these toxic metals,contaminating the drinking water used by *** samples taken from the tubes were evaluated by a square-wave anodic stripping voltammetry technique,and the results for lead and cadmium suggested randomly distributed levels,indicating that the contents of these contaminants depend mainly on the quality of the added primary zinc and the operational conditions of the immersion *** metallographic evaluations performed on the samples of the galvanized tubes showed irregularities in thickness and even a lack of the zinc layer deposited on the carbon steel *** facts suggest that galvanized tubes should not be used for drinking water.

Background: Schizophrenia is a severe psychiatric disorder with a complex genetic factor determining its disease onset. Nevertheless, it is not clear in this mental disorder. Objective: To conduct a systematic review of articles regarding the genetic markers and mutations in schizophrenia. Methods: A systematic review of articles on genetic markers and mutations in schizophrenia, published from January 1, 2011, to September 7, 2015, on SCOPUS database was carried out. Search terms were “Genetic markers”, “Mutation”, and “Schizophrenia”. Results: Of the 527 retrieved studies, 31 met the eligibility criteria. Genetic polymorphism, Immune-associated genes, TCF4 and ZNF804A association with microRNA, Neuregulin gene, Chromosome 13q32 and 11p15.4, genes involved in glutamatergic via schizophrenia and brain structure, appeared to be associated with the origin of schizophrenia. Conclusion: Some studies show genes involved in several pathways leading to the disease pathogenesis such as that one related with the dopaminergic and immune system, or rare alleles. Some genes present no involvement in the etiology of this mental disorder. These findings clarify the genetic complexity of schizophrenia and affirm that together, the genes have an overall effect greater than the sum of the individual effect of each gene.

Incorrect discard of oil used by fishing ship in the brazilian north region has become a common *** the possible hazard to the aquatic organisms,still missing scientific data about their ***,this study aimed to evaluate the stress and lethality caused by lubricant oil(FSAOLU)on fish Hyphessobrycon ***,it was used six different concentration diluted in water(0,22,24,26,28,30 and 32%of oil)and three replaces during 96 *** the end or during the experiment(with dying fish),it was collected blood samples(cutting the caudal fin)to determine physiological *** FSAOLU showed mean lethal dose(LD50-96h)of 27.36%,classified as toxic causing alterations in glucose values from the 26%.The greater dilution of FSAOLU(32%)increased 115%in glucose values when compared to the ***,lubricant oil when discarded in water,it present hazard to the aquatic organisms causing stress and mortality for fish being necessary adequate management to discard of this residue.

This paper presents a proposal for an experimental salt cavern in offshore ultra-deep water for CO2 abatement,including the instrumentation plan and well conceptual design evaluated for carbon capture and storage(CCS)*** studies are based on applied computational mechanics associated with field experimentation that has contributed to the technical feasibility of the underground potash mine at the State of Sergipe in *** knowhow allowed the stability analysis of several salt caverns for brine production at the State of Alagoas in Brazil and to the drilling through stratified thick layers of salt of the pre-salt reservoirs in santos ***,this knowledge has been applied in the design of onshore and offshore salt caverns opened by dissolution for storage of natural gas and *** geomechanical study,through the application of computational mechanics,of offshore giant salt caverns of 450 m high by 150 m in diameter,shows that one cavern can store about 4 billion Sm3 or 7.2 million tons of *** the construction of the giant cavern,which will be the first gas storage offshore in the world,it has been decided to develop an experimental one,with smaller size,to obtained field *** experimental cavern will allow the calibration of parameters to be used in the structural integrity analysis of the cavern and well for storage of natural gas which is rich in CO2 under high pressure.
