Plant performance and morphofunctional differentiation in response to edaphic variation in Iberian columbines: cues for range distribution?

Aims Experimental studies of the response to environmental variation of closely related taxa are needed to understand the mechanisms underly-ing phenotypic divergence,habitat segregation and range span within a radiating genus.We explored the magnitude of phenotypic differenti-ation and adaptive plasticity in relation to edaphic variation and its sig-nificance for habitat segregation and range span in Iberian columbines.Methods We performed a common garden experiment varying soil type(cal-careous vs.siliceous)and depth(rocky-shallow vs.deep)with two pairs of widespread and narrowly distributed subspecies of Iberian columbines:Aquilegia vulgaris(subspp.vulgaris and nevadensis)and Aquilegia pyrenaica(subspp.pyrenaica and cazorlensis).We com-pared tolerance to edaphic variation,trait differentiation and adaptive plasticity of 10 morpho-functional traits between species and distri-bution ranges.additionally,we obtained estimates of the competitive environment faced by each taxon in two to four populations per taxa.Important Findingsresults partially support hypotheses of higher competitive ability and tolerance to edaphic variation in widespread than in narrow endemic taxa.at the species level,the widely distributed taxon was the most tolerant to edaphic variation.Within species,no consistent pattern emerged since the population from the most widespread subspecies was the most tolerant in A.vulgaris but not in A.pyr-enaica.Columbines were differentiated in many traits at species and range level.However,the pattern of differentiation does not fully support the hypothesis of higher specialization and stress toler-ance in narrow endemics.although plasticity was generally low,the results support the hypothesis of adaptive plasticity in widespread but not in restricted taxa at least at species level.Ecological differences(adaptive plasticity and competitive ability among others)may have contributed to phenotypic divergence and edaphic niche segregation,as well as to differences in range span among columbines.

Complex patterns of environmental niche evolution in Iberian columbines (genus Aquilegia, ranunculaceae)

Aims This study explores the patterns of niche differentiation in a group of seven closely related columbines(genus Aquilegia,ranunculaceae)from the Iberian Peninsula.Populations of these columbines are subject to complex patterns of divergent selection across environ-ments,which partly explain the taxonomic structure of the group.This suggests the hypothesis that niche divergence must have occurred along the process of diversification of the group.Methods We used maxEnt to build environmental niche models of seven subspecies belonging to the three species of Aquilegia present in the Iberian Peninsula.From these models,we compared the envi-ronmental niches through two different approaches:ENmtools and multivariate methods.Important FindingsmaxEnt distributions conformed closely to the actual distribution of the study taxa.ENmtools methods failed to uncover any clear patterns of niche differentiation or conservatism in Iberian columbines.multivariate analyses indicate the existence of dif-ferentiation along altitudinal gradients and along a gradient of climatic conditions determined by the summer precipitation and temperatures.However,climatic conditions related to winter tem-perature and precipitation,as well as soil properties,were equally likely to show conservatism or divergence.The complex patterns of niche evolution we found suggest that Iberian Columbines have not been significantly constrained by forces of niche conservatism,so they could respond adaptively to the fast and profound climate changes in the Iberian Peninsula through the glacial cycles of the Pleistocene.

comLocal adaptation to distinct elevational cores contributes to current elevational divergence of two Aquilegia vulgaris subspecies

Aims Phenotypic plasticity and local adaptation of populations at their distributional limits are crucial to understand species colonization and persistence in novel or marginal environments,as well as spe-cies divergence and niche width evolution.We assess the contri-bution of these processes to shape current elevational limits and determine elevational phenotypic divergence between two subspe-cies of Aquilegia vulgaris(subsp.vulgaris and nevadensis).Methods We conducted sowing and transplant experiments considering four elevations throughout the current elevational range of A.vulgaris in southern Iberian Peninsula.Experiments were designed to explore,on the one hand,local adaptation through three components of performance(germination,survival and growth)and,on the other hand,the phenotypic differentiation and/or plasticity associated to local adaptation.Four populations per subspecies(three from the elevational core and one from the elevational boundary)were used as seed sources.Patterns of local adaptation and phenotypic differentiation are examined in the context of the‘centre-periphery’hypothesis.Important Findings Central populations of both subspecies performed better at their local elevations while marginal populations were maladapted,confirming the hypothesis and contributing to explain the current elevational segregation of these subspecies.Density of glandular pubescence and germination timing seem to be related to local adaptation,through phenotypic differentiation between subspe-cies or elevations.The widespread subsp.vulgaris showed signals of adaptive plasticity in the timing of germination while it was not the case in the endemic subsp.nevadensis.

Experimental test of a building thermal system for preventive maintenance based on thermoeconomic analysis

An analysis of the sectorial structure of energy consumption shows that residential and tertiary sector buildings are the third-highest consumers,responsible for 29.5%of a city’s final energy consumption.The Building Quality Control Laboratory of the Basque Government aims to promote quality,innovation,and sustainability in buildings.To accomplish this goal,it has constructed an experimental facility with different energy generation technologies and a very versatile control system for testing different energy systems and operation modes.In this study,we tested a facility for supplying domestic hot water and heating for a multi-family house by means of a condensing boiler and an aerothermal heat pump(together with the corresponding control).This installation could reproduce the thermal demands required to be satisfied by the generation equipment through a programmed operation of the installation based on real demands.Additionally,this installation was analyzed using thermoeconomics(TE)to solve problems unable to be solved using traditional energy analyses based on the First Law of Thermodynamics.These problems include:(1)Determining the costs of the products of the installation based on physical criteria,(2)detecting the places where losses actually occur,evaluating their costs,and proposing cost-effective improvements,and(3)diagnosing issues in the installation.As a result,this paper suggests a solution to the preventive maintenance problems confronting the technical maintenance personnel for thermal installations in buildings by applying TE knowledge and using real data collected from sensors.

Purpose:To examine the effects of a school-based karate intervention on academic achievement,psychosocial functioning,and physical fitness in children aged 7-8 years.Methods:Twenty schools in 5 different European countries(2 second-grade classrooms per school)participated in a cluster randomized controlled trial(Sport at School trial).Participants were assigned to either a control group,which continued with their habitual physical education lessons,or to an intervention group,which replaced these lessons with a 1-year karate intervention(Karate Mind and Movement program).A total of 721 children(344 girls and 377 boys,7.4±0.5 years old,mean±SD)completed the study,of which 333 and 388 were assigned to the control group and intervention group,respectively.Outcomes included academic performance(average grade),psychosocial functioning(Strengths and Difficulties Questionnaire for parents),and different markers of physical fitness(cardiorespiratory fitness,balance,and flexibility).Results:The intervention provided small but significant benefits compared to the control group for academic achievement(d=0.16;p=0.003),conduct problems(d=-0.28;p=0.003),cardiorespiratory fitness(d=0.36;p0.05).Conclusion:A 1-year school-based karate intervention was effective in improving academic achievement,conduct problems,and physical fitness in primary school children.The results support the inclusion of karate during physical education lessons.

Patterns of variation in the size of vulture foraging groups,and their ecological causes and consequences,remain litle explored despite strong links with the carrion recycling service that this key functional group provides.We documented the group size-frequency pattern of Andean con-dors Vultur gryphus gathered to feed on 42 equine carcasses experimentally placed in Bolivia,between 2012 and 2019,along an elevation range of 1,300-4,500 m asl.Based on the location(altitude and latitude)of the foraging groups,we examined the relationship between their size and species'population parameters(size and trend),habitat conditions,and livestock carcass availability and predictability.Condors utilized a high frequency(93%)of carcasses forming groups that ranged from 1 to 80 individuals(mean=25,median=18)and shaped a"lazy-j curve"typical pattern of size-frequency distribution whereby few groups(5,12%)were large(>55 individuals)and most(21,50%)were relatively small(<19 individuals).Group size related to altitude in that most larger groups formed at lower sites(below c.3,000 m asl),likely following an altitudinal gradient whereby larger groups are more likely to form around larger carcasses(i.e.,cattle),which are more likely to occur at lower elevations.Regardless of population size,group size could be an adaptive response of condors via local enhancement for improving individual scavenging efficiency.Many information gaps on this topic still exist,thus we provide a set of questions to address them,especially amidst the unrestricted impacts of human activities that condition vulture survival globally.

Nitric oxide(NO)is an essential reactive oxygen species and a signal molecule in plants.Although several studies have proposed the occurrence of oxidative NO production,only reductive routes for NO production,such as the nitrate(NO_(3)^(-))-upper-reductase pathway,have been evidenced to date in land plants.However,plants grown axenically with ammonium as the sole source of nitrogen exhibit contents of nitrite and NO3−,evidencing the existence of a metabolic pathway for oxidative production of NO.We hypothesized that oximes,such as indole-3-acetaldoxime(IAOx),a precursor to indole-3-acetic acid,are intermediate oxidation products in NO synthesis.We detected the production of NO from IAOx and other oximes catalyzed by peroxidase(POD)enzyme using both 4-amino-5-methylamino-2′,7′-difluorescein fluorescence and chemiluminescence.Flavins stimulated the reaction,while superoxide dismutase inhibited it.Interestingly,mouse NO synthase can also use IAOx to produce NO at a lower rate than POD.We provided a full mechanism for POD-dependent NO production from IAOx consistent with the experimental data and supported by density functional theory calculations.We showed that the addition of IAOx to extracts from Medicago truncatula increased the in vitro production of NO,while in vivo supplementation of IAOx and other oximes increased the number of lateral roots,as shown for NO donors,and a more than 10-fold increase in IAOx dehydratase expression.Furthermore,we found that in vivo supplementation of IAOx increased NO production in Arabidopsis thaliana wild-type plants,while prx33-34 mutant plants,defective in POD33-34,had reduced production.Our data show that the release of NO by IAOx,as well as its auxinic effect,explain the superroot phenotype.Collectively,our study reveals that plants produce NO utilizing diverse molecules such as oximes,POD,and flavins,which are widely distributed in the plant kingdom,thus introducing a long-awaited oxidative pathway to NO production in plants.This knowledge has essential implications for und

The RNA Modification N^6-methyladenosine and Its Implications in Human Disease

Impaired gene regulation lies at the heart of many disorders, including developmental diseases and cancer. Furthermore, the molecular pathways that control gene expression are often the target of cellular parasites, such as viruses. Gene expression is controlled through multiple mechanisms that are coordinated to ensure the proper and timely expression of each gene. Many of these mechanisms target the life cycle of the RNA molecule, from transcription to translation. Recently, another layer of regulation at the RNA level involving RNA modifications has gained renewed interest of the scientific community. The discovery that N6-methyladenosine （m6A）, a mod- ification present in mRNAs and long noncoding RNAs, can be removed by the activity of RNA demethylases, launched the field of epitranscriptomics; the study of how RNA function is regulated through the addition or removal of post-transcriptional modifications, similar to strategies used to regulate gene expression at the DNA and protein level. The abundance of RNA post-transcriptional modifications is determined by the activity of writer complexes （methylase） and eraser （RNA demethylase） proteins. Subsequently, the effects of RNA modifications materialize as changes in RNA structure and/or modulation of interactions between the modified RNA and RNA binding proteins or regulatory RNAs. Disruption of these pathways impairs gene expression and cellular function. This review focuses on the links between the RNA modification m6A and its implications in human diseases.
