Gene transcription profiting of astheno-and normo-zoospermic sperm subpopulations

Spermatozoa contain a repertoire of RNAs considered to be potential functional fertility *** this study,the gene expression of human sperm subpopulations with high(F1)and low(F2)motility from healthy normozoospermic(N)and asthenozoospermic(A)individuals was evaluated using RNA microarray followed by functional genomic analysis of differentially expressed *** from A-F1 versus N-F1,A-F2 versus N-F2,N-F1 versus N-F2,and A-F1 versus A-F2 comparisons showed a considerably larger set of downregulated genes in tests versus *** ontology(GO)analysis of A-F1 versus N-F1 identified 507 overrepresented biological processes(BPs),several of which are associated with sperm *** addition,gene set enrichment analysis of the same contrast showed 110 BPs,36 cellular comp on ents,and 31 molecular functions,several of which are involved in sperm motility.A leadi ng・edge analysis of selected GO terms resulted in several down regulated genes encoding to dyn eins and kin esins,both related to sperm ***,the predicted activation state of asthenozoospermia was increased,while fertility,cell movement of sperm,and gametogenesis were ***,several downregulated genes characteristic of the canonical pathway protein ubiquitination were involved in asthenozoospermia ***,GO analysis of A-F2 versus N-F2 did not identify overrepresented BPs,although the gene set enrichment analysis detected six enriched BPs,one cellular component,and two molecular ***,the results show differences in gene transcription between sperm subpopulations from asthenozoospermic and normozoospermic semen samples and allowed the identification of gene sets relevant to sperm physiology and reproduction.

Can DNA fragmentation of neat or swim-up spermatozoa be used to predict pregnancy following ICSI of fertile oocyte donors？

This study compared the potential of assessing sperm DNA fragmentation （SDF） from neat semen and the subsequent swim-up （SU） procedure to predict pregnancy when conducting ICSI of fertile donor oocytes. Infertile females （n=81） were transferred embryos resulting from intracytoplasmic sperm injection （ICSI） of their partner＇s spermatozoa and proven donor oocytes. This model normalized the impact of female factor in putative sperm DNA repair. Semen was blindly assessed for SDF using Halosperm immediately following ejaculation （NS） and after swim-up at the time of ICSI fertilisation. There was a decrease in SDF values of the ejaculated semen sample following the swim-up protocol （P=0.000）. Interestingly, pregnancy could be equally predicted from SDF values derived from either neat or swim-up semen samples. Receiver operator curves and the derived Youden＇s indices determined SDF cutoff values for NS and SU of 24.8% and 17.5%, respectively. Prediction of pregnancy from NS SDF had a sensitivity of 75% and a specificity of 69%, whereas for SU SDF was 78% and 73%, respectively. While increased levels of SDF negatively impact reproductive outcome, we have shown that a reduction in SDF following sperm selection using ICSI with proven donor oocytes is not mandatory for achieving pregnancy. This suggests that a certain level of DNA damage that is not detectable using current technologies could be impacting on the relative success of assisted reproductive technology （ART） procedures. Consequently, we propose a modification of the so called ＇iceberg model＇ as a possible rationale for understanding the role of SDF in reproductive outcome.

Dominant Macroalgae in Bahía de Cochinos, Matanzas, Cuba

Spatial-temporal variations of macroalgae were analyzed in a study conducted in Cueva de Los Peces (CP) and Punta Perdiz (PP), two dive sites located on the eastern coast of Bahía de Cochinos. The most conspicuous species found from 3 to 20 m depths were present in the biotopes of ridges and shallow and deep terraces. Sampling was carried out in September 2014, and March and October 2016 by autonomous diving and direct methods. Qualitative visual censuses and quantitative analyzes were used to estimate the coverage (%) of the genera according to the AGRRA methodology with the use of the 10 m linear transect and quadrats as the sampling unit. The first list of macroalgae for the area is offered that includes 49 taxa of which 10 were Rhodophyta, 10 Ochrophyta (Phaeophyceae), and 29 Chlorophyta. The dominant genera during the study period were Halimeda, Dictyota, Lobophora, and Udotea. Spatial differences were found regarding the specific composition. Temporal variations were evidenced in terms of the relative abundance of the genera present, which reveals temporal changes in the qualitative structure, where some species replace others. The information obtained is pioneering and can serve as a comparative baseline for future monitoring of the area.

Recognition of a multiple antigen peptide containing sequence from mimotope of the dengue type 3 virus NS4B protein by human antibodies

Objective: To evaluate the recognition of NS4 B mimotope, as multiple antigen peptide(MAP), by dengue antibodies presents in serum samples from patients with different serotype infections. Methods: A MAP containing mimotope sequence was synthesized and used to evaluate the recognition of NS4 B mimotope as MAP by a panel of 66 human sera from dengue cases by an indirect ELISA assay. Results: The MAP differentiated between sera from dengue viruses infected patients and sera from healthy individuals and the best reactivity was shown by serum from dengue type 3 virus patients. The recognition was more intense with serum from patients with secondary infection. Conclusions: The findings suggest the potential use of NS4 B mimotope on the development of a multi-epitope diagnostic tool. These results are important for further immunogenicity studies.

A Novel Classification of Brain Invasion in Malignant Ethmoidal Tumors and Management Proposal

Object: Brain invasion is unusual in patients with paranasal sinuses cancer. Staging systems do not specify brain invasion extension who has an impact in therapeutic strategy and prognosis. Surgical series of this patient treated by endonasal endoscopic approach are scarce. In this paper, a novel classification and surgical strategy is proposed. Methods: A retrospective review of patients with ethmoidal malignant tumors with brain invasion treated at the National Institute of Oncology and Radiobiology from January 2015 to October 2017 by means endonasal endoscopic surgery was performed. All medical records were reviewed for demographics, diagnoses, presentation, endoscopic approach, reconstructive techniques and clinical outcomes. Pattern of brain invasion was classified in five grades. Chi-squared test, ANOVA test and Kaplan-Meier curves were used. Results: Forty-seven patients (26 males and 21 females) were registered. Mean age was 52.3 years ± 27.0. Only 3 patients presented with neurologic symptoms. Histologic subtypes were squamous cell carcinoma (25 patients), adenocarcinoma (16 patients), and esthesioneuroblastoma (6 patients). Five patients corresponded to Grade I, 39 Grade II and 3 Grade III. The mean surgical time was 351.9 min (±45). The mean blood loss was 704.5 ml (±206). The mean ICU stay was 1.1 (ranging from 1 to 2 ) days and the hospital stay was 5.3 (ranging from 5 to 7) days. Conclusions: Brain invasion represents a particular therapeutic challenge in patients with malignant nasosinusal tumors. Surgical treatment is possible in some cases, including endonasal endoscopic approach. A stage classification of brain invasion helps to establish an optimal surgical strategy and a prognosis.

Notwithstanding the emergence of FinTech startups in the financial services industry,most of these companies face significant difficulties in breaking even and *** study examines the main managerial, institutional, and financial drivers of FinTechprofitability and the time it takes for these firms to break even. The database includesrelevant qualitative factors, such as foundational characteristics, the technologicalprofile of the startup, and its funding structure. Using the full population of FinTechstartups operating in Spain from 2005 to 2017, we observe that most of these firms areunprofitable within three years of their inception. Combining panel data and survivalanalyses, we empirically find that large and solvent FinTech firms founded by singleentrepreneurs in an incubator or accelerator program are more likely to be profitableand prevail. FinTech firms reach their break-even points faster if they receive fundingthrough seed capital.

The Role of Human Herpesvirus 8 Molecular Characterization in the Management of HIV Infected Patients Diagnosed with Malignancies Associated with Its Infection

Despite the progress has been reached with Human herpesvirus 8 (HHV-8) research, there are gaps in the knowledge of viral induced oncogenesis. The aim of the present study was to identify possible associations between HHV-8 subtypes, HHV-8 loads and clinical manifestations of HIV infected patients diagnosed with different malignancies associated with HHV-8 infection. Forty six HIV-1 infected individuals diagnosed with different HHV-8 associated diseases were studied [37 epidemic Kaposi’s sarcoma (KS), 3 pleural effusion lymphoma (PEL);5 peripheral lymphadenopathies (PL);1 Hodgkin’s lymphoma (HL);1 non Hodgkin’s lymphoma (NHL)]. HHV-8 loads were determined by quantitative real time PCR (qRT-PCR) whilst HHV-8 subtypes were determined by open-reading frame (ORF)-K1 gen genotyping. HHV-8 subtypes B, A, C, A5 and E were exhibited by 31.8%, 23.4%, 19.1%, 17% and 8.5% of the studied patients, respectively. The median HHV-8 viral load did not differ between subtypes (p > 0.05) but HHV-8 viral loads were significantly higher in PEL than in epidemic KS lesion or lymph nodes (p = 0.04). Subtype B was detected in 60% of patients with B cell lymphoma (NHL, PEL and HL) whereas subtype E was only detected in patients with epidemic KS diagnosis. Our data suggest that HHV-8 DNA quantification instead of subtype identification could be used as a surrogate marker for monitoring its infection, not only in epidemic KS patients but also in HIV infected individuals with lymphoproliferative disorders.

Vapor-phase transformations of furfural over SBA-15 silica supported Ni catalysts under H2 in a continuous-flow reactor at atmospheric pressure and 170°C and 230°C were investigated. Two different samples having Ni loadings of 5 and 20 wt% (denoted here by SBA-5Ni and SBA-20Ni, respectively) were prepared by impregnation and characterized by atomic absorption spectroscopy, N2 sorption analysis, X-ray diffraction, X-ray photoelectron spectroscopy, and transmission electron microscopy. Furan and furfuryl alcohol were two primary products resulting, respectively, from decarbonylation and hydrogenation of furfural. Under the conditions of the study, both reactions exhibited structure sensitivity evidenced by changes in product selectivities with variable Ni loadings. Compared with SBA-20Ni, the 5 wt% Ni catalyst showed better catalytic activity, reaching a furfural conversion of 100 mol% and a selectivity to furan of 98 mol%, after 5 h of time-on-stream at 230°C.
