Biotinylated polyurethane-urea nanoparticles for targeted theranostics in human hepatocellular carcinoma

Over the past years, significant efforts have been devoted to explore novel drug delivery and detection strategies for simultaneous therapy and diagnostics. The development of biotinylated polyurethane-urea nanoparticles as theranostic nanocarriers for targeted drug and plasmid delivery, for fluorescence detection of human hepatocellular carcinoma cells, is described herein. These targeted nanoparticles are specifically designed to incorporate biotin into the polymeric matrix, since many tumor types overexpress receptors for biotin as a mechanism to boost uncontrolled cell growth. The obtained nanoparticles were spherical, exhibited an average diameter ranging 110-145 nm, and showed no cytotoxicity in healthy endothelial cells. Biotinylated nanoparticles are selectively incorporated into the perinuclear and nuclear area of the human hepatocellular carcinoma cell line, HepG2, in division, but not into growing, healthy, human endothelial cells. Indeed, the simultaneous incorporation of the anticancer drugs, phenoxodiol or sunitinib, together with plasmid DNA encoding green fluorescent protein, into these nanoparticles allows a targeted pharmacological antitumor effect and furthermore, selective transfection of a reporter gene, to detect these cancer cells. The combined targeted therapy and detection strategy described here could be exploited for liver cancer therapy and diagnostics, with a moderate safety profile, and may also be a potential tool for other types of cancer.

Phyllosticta capitalensis is an endophyte and weak plant pathogen with a worldwide distribution presently known from 70 plant *** study isolated *** from different host plants in northern Thailand,and determined their different life *** strains of *** were isolated as endophytes from 20 *** additional 30 strains of *** from other hosts and geographic locations were also obtained from established culture *** analysis using ITS,AcTand TEF gene data confirmed the identity of all *** tests with five strains of *** originating from different hosts were completed on their respective host *** all cases there was no infection of healthy leaves,indicating that this endophyte does not cause disease on healthy,unstressed host *** *** is often isolated as an endophyte has important implications in fungal biology and plant *** to its endophytic nature,*** commonly found associated with lesions of plants,and often incorrectly identified as a species of quarantine importance,which again has implications for trade in agricultural and forestry production.

A way to extend operators in spaces of continuous functions to spaces of continuous set_valued functions is proposed. This extension is developed through the Steiner selections of the set_valued functions. Their properties and characteristics of the convergence of sequences of operators of this class are studied. In Part Ⅱ of this series some applications to approximation theory will be shown.

High Accurate Howland current Source: Output constraints Analysis

Howland circuits have been widely used as powerful source for exciting tissue over a wide frequency range. When a Howland source is designed, the components are chosen so that the designed source has the desired characteristics. However, the operational amplifier limitations and resistor tolerances cause undesired behaviors. This work proposes to take into account the influence of the random distribution of the resistors in the modified Howland circuit over the frequency range of 10 Hz to 10 MHz. Both output current and impedance of the circuit are deduced either considering or the operational amplifiers parameters. The probability density function due to small changes in the resistors of the circuit was calculated by using the analytical modeling. Results showed that both output current and impedance are very sensitive to the resistors variations. In order to get higher output impedances, high operational amplifier gains are required. The operational amplifier open-loop gain increases as increasing the sensitivity of the output impedance. The analysis done in this work can be used as a powerful co-adjuvant tool when projecting this type of circuit in Spice simulators. This might improve the implementations of practical current sources used in electrical bioimpedance.

Quantitative morphometric analysis of the Jama River profile in a tectonically active margin (Northwestern Ecuador)

The longitudinal profile of a river channel can be described in function of mathematical expressions. The logarithmic fit is the most used method to describe the relative equilibrium of a channel elevation profile. Rivers showing zones of high channel gradient and convex-upward profiles can be evaluated in function of the offset distance with respect to the logarithmic curve. The Jama River profile has been constructed using differential GPS data for the downstream reach and 4-m-grid DEM data base for the remaining headward profile. The resulting longitudinal profile shows a prominent knickpoint of about 80 m in elevation characterized by sharp local convexity. The offset distance in vertical direction from the logarithmic plot is interpreted as the successive accumulation of multiple uplift episodes associated with the Jama Fault System activity. The horizontal offset is suggested to represent the remaining retreated distance for the upstream propagation of the Jama knickpoint. The highly resistant cretaceous rocks outcropping along the Jama knickpoint reach acts as an inflection point between two subprofiles that show well-fitted logarithmic curves, thereby representing a temporary partitioned equilibrium along the Jama River profile.

A Two-Point Boundary Value Problem by Using a Mixed Finite Element Method

This paper describes a numerical solution for a two-point boundary value problem. It includes an algorithm for discretization by mixed finite element method. The discrete scheme allows the utilization a finite element method based on piecewise linear approximating functions and we also use the barycentric quadrature rule to compute the stiffness matrix and the L2-norm.

The Major Facilitator Superfamily(MFS)is ubiquitous in living organisms and represents the largest group of secondary active membrane *** plants,significant research efforts have focused on the role of specific families within the MFS,particularly those transporting macronutrients(c,N,and P)that constitute the vast majority of the members of this *** MFS families remain less explored,although a plethora of additional substrates and physiological functions have been ***,the lack of a systematic approach to analyzing the MFS as a whole has obscured the high diversity and versatility of these ***,we present a phylogenetic analysis of all annotated MFS domaincontaining proteins encoded in the Arab/dops/s fA?a//ana genome and propose that this superfamily of transporters consists of 218 members,clustered in 22 *** reviewing the available information regarding the diversity in biological functions and substrates of Arab/dops/s MFS members,we provide arguments for intensified research on these membrane transporters to unveil the breadth of their physiological relevance,disclose the molecular mechanisms underlying their mode of action,and explore their biotechnological potential.

Modeling and Simulation of Molecular Mechanism of Action of Dietary Polyphenols on the Inhibition of Anti-Apoptotic PI3K/AKT Pathway

In recent years, the role of dietary phenolic compounds in the regulation of cellular metabolism in normal and pathological conditions has become increasingly important in cancer research. In most cases, the molecular mechanism of action related to the anticarcinogenic effect of phenolic compounds has been studied in vitro and in animal models, but these studies are still not complete. It is precisely here where in silico approaches can be an invaluable tool for complementing in vitro and in vivo research. In this paper, we adopt a tuple space-based modeling and simulation approach, and show how it can be applied to the simulation of complex interaction patterns of intracellular signaling pathways. Specifically, we are working to explore and to understand the molecular mechanism of action of dietary phenolic compounds on the inhibition of the PI3K/AKT anti-apoptotic pathway. As a first approximation, using the tuple spaces- based in silico approach, we model and simulate the anti-apoptotic PI3K/AKT pathway in the absence and presence of phenolic compounds, in order to determine the effectiveness of our platform, to employ it in future prediction of experimentally non visualized interactions between the pathway components and phenolic compounds.

Plant performance and morphofunctional differentiation in response to edaphic variation in Iberian columbines: cues for range distribution?
