Using classification assignment rules to assess land-use change impacts on forest biodiversity at local-to-national scales

Background： Ecosystem representation is one key component in assessing the biodiversity impacts of land-use changes that will irrevocably alter natural ecosystems. We show how detailed vegetation plot data can be used to assess the potential impact of inundation by a proposed hydroelectricity dam in the Mokihinui gorge, New Zealand, on representation of natural forests. Specifically we ask： 1） How well are the types of forest represented Locally, regionally, and nationally; and 2） How does the number of distinct communities （i.e. beta diversity） in the target catchment compare with other catchments nationally？ Methods： For local and regional comparisons plant species composition was recorded on 45 objectively located 400 m2 vegetation plots established in each of three gorges, with one being the proposed inundation area of the Mokihinui lower gorge. The fuzzy classification framework of noise clustering was used to assign these plots to a specific alliance and association of a pre-existing national-scale classification. NationaLly, we examined the relationship between the number of alliances and associations in a catchment and either catchment size or the number of plots per catchment by fitting Generalised Additive Models. results： The four alliances and five associations that were observed in the Mokihinui lower gorge arepresent in the region but limited locally. One association was narrowly distributed nationally, but is the mostfrequent association in the Mokihinui lower gorge; inundation may have consequences of national importance to its long-term persistence. That the Mokihinui lower gorge area had nearly twice as many plots that could not be assigned to pre- existing alliances and associations than either the Mokihinui upper or the karamea lower gorges and proportionally more than the national dataset emphasises the compositional distinctiveness of this gorge. Estrogen receptor expression in chronic hepatitis C and hepatocellular carcinoma pathogenesis

AIM To investigate gender-specific liver estrogen receptor(Er) expression in normal subjects and patients with hepatitis C virus(HCV)-related cirrhosis and hepatocellular carcinoma(HCC).METHODS Liver tissues from normal donors and patients diagnosed with HCV-related cirrhosis and HCV-related HCC were obtained from the NIH Liver Tissue and Cell Distribution System. The expression of Er subtypes, Erα and Erβ, were evaluated by Western blotting and real-time rT-PCr. The subcellular distribution of Erα and Erβ was further determined in nuclear and cytoplasmic tissue lysates along with the expression ofinflammatory [activated NF-κB and IκB-kinase(Ikk)] and oncogenic(cyclin D1) markers by Western blotting and immunohistochemistry. The expression of Erα and Erβ was correlated with the expression of activated NF-κB, activated Ikk and cyclin D1 by Spearman's correlation. rESULTS Both Er subtypes were expressed in normal livers but male livers showed significantly higher expression of Erα than females(P rved significantly higher m rNA expression of Erα in HCV-related HCC liver tissues as compared to normals(P rβ in livers of HCV-related cirrhosis and HCV-related HCC subjects(P rotein level, there was a significantly higher expression of nuclear Erα in livers of HCV-related HCC patients and nuclear Erβ in HCV-related cirrhosis patients as compared to normals(P rthermore, we observed a significantly higher expression of phosphorylated NF-κB and cyclin D1 in diseased livers(P re was a positive correlation between the expression of nuclear Er subtypes and nuclear cyclin D1 and a negative correlation between cytoplasmic Er subtypes and cytoplasmic phosphorylated Ikk in HCV-related HCC livers. Monkey B virus(Macacine herpesvirus 1; BV) is noted for its extreme neurovirulence in humans. Since the vhs protein encoded by the UL41 gene has been shown to be a neurovirulence factor in the related human herpes simplex viruses, the role of the UL41 gene in BV neurovirulence was investigated. BV mutants were constructed that lacked the entire UL41 OrF(Δ41) or had the rNase active site mutated(Δ41A). Neither mutant shut off host protein synthesis, degraded β-actin mrNA, or prevented an IFN-β response, indicating that the vhs protein and its rNase activity are both necessary for these activities. replication of both mutants in primary mouse cells was impaired and they exhibited a prolonged disease course in mice. Whereas Δ41 infected mice were euthanized for symptoms related to central nervous system(CNS) infection, Δ41A infected mice were euthanized primarily for symptoms of autonomic nervous system dysfunction. While neuroinvasiveness was not affected, lesions in the CNS were more limited in size, anatomical distribution, and severity than for wild-type virus. These results indicate that the vhs protein affects the general replicative efficiency of BV in vivo rather than being a specific neurovirulence factor critical for invasion of or preferential replication in the CNS.

Dear Editor, Host chaperone proteinsarecritical factors in plant virus infec- tion. One ofthe most commonly identified host factors involved in plant virus infection is Hsp70, which functions in ATP hydroly- sis, protein folding, assembly of protein complexes, and translo- cation between cellular compartments. A comparative study of plant virus infection, including Potato virus X （PVX）, reported that Hsp70 is up-regulated by virus infection and possibly con- tributes to pathogenesis in both Arabidopsis and N. benthami- ana plants （Aparicio et al., 2005）. Cytosolic Hsp70 interacts with the tombusvirus and tymovirus replication complexes （Wang et al., 2009）, the potyvirus coat protein （Hafren et al., 2010）, and closteroviruses encode an HSP70 homolog that functions to enable virus intercellular transport （Agranovsky et al., 1998）.

revisiting Sphere Unfolding relationships for the Stereological Analysis of Segmented Digital Microstructure Images

Sphere unfolding relationships are revisited with a specific focus on the analysis of segmented digital images of microstructures. Since the features of such images are most easily quantified by counting pixels, the required equations are re-derived in terms of the histogram of areas (instead of diameters or radii) as inputs and it is shown that a substitution can be made that simplifies the calculation. A practical method is presented for utilizing negative number fraction bins (which sometimes arise from erroneous assumptions and/or insufficient numbers of observations) for the creation of error bars. The complete algorithm can be implemented in a spreadsheet. The derived unfolding equations are explored using both linear and logarithmic binning schemes, and the pros and cons of both binning schemes are illustrated using simulated data. The effects of the binning schemes on the stereological results are demonstrated and discussed with reference to their consequences for practical materials characterization situations, allowing for the suggestion of guidelines for proper application of this, and other, distribution-free stereological methods.

Continuously Monitored Canopy Temperature as a Proxy for Plant Water Status

Water deficits are major limiters of crop yield worldwide. The detection of water deficits can be difficult. Measurements of the aerial and soil environment are often used to infer the water status and detect water deficits. Since crop yield accumulates incrementally and cumulatively over seasonal time scales, continuous direct monitoring of the water status of the crop may provide needed insight into plant/environment interactions. Canopy temperature can be measured near continuously on seasonal scales in the field. Cotton was grown under 11 irrigation regimes in 2009 and 2010 with water deficits from 26% to 86% of crop evapotranspiration. Yield varied accordingly from ~500 kg·ha-1 to ~2600 kg·ha-1. Canopy temperature was measured on a 15-minute interval for ~65 days in each year. Yield was described by a linear function of total water (irrigation + rain) for each year with similar slopes and different intercepts. When canopy temperature was used as a surrogate for total water, yield was linearly related to daytime leaf-to-air VPD, mean seasonal canopy temperature, mean seasonal daytime canopy temperature, and cumulative seasonal daytime canopy temperature. Limiting the analysis to daytime periods improved the ability to account for yield variation. Mean daytime seasonal canopy temperature and cumulative seasonal daytime temperature were most effective in accounting for yield variation across the seasons with a single regression line for both years.

The effect of manual acupuncture and electroacupuncture on lower limb muscle strength

Objective： To determine whether six weeks＇ of acupuncture has a positive effect on plantar flexion muscle strength and whether electroacupuncture （EA） has a greater effect than manual acupuncture （MA） on plantar flexion, so that the most beneficial method of acupuncture can be established for strengthening muscles. Methods： A randomized controlled trial with parallel groups. A total of 20 participants were randomly assigned to three groups： a control/non-acupuncture group （n=6）, a manual acupuncture group （MAG）（n=8） and an electroacupuncture group （EAG）（n=6）. MAG and EAG received three 25-minute treatments per week for 6 weeks. The control group continued with their normal training activity and received no treatment. The maximal voluntary contraction （MVC） of the ankle plantar-flexors at 30~ per second was measured using Biodex system 3, results： Both MA and EA had a positive effect in increasing plantar flexion strength. The likelihood for this happening was high; 69% for MA and 92% for EA when compared with the control group. However the effect size was small for both groups. There was only a trivial difference in the effect size between the EAG and the MAG in regards to average plantar flexion torque. Conclusion： EA and MA have a beneficial effect on plantar flexion muscle strength compared with the control group in healthy population.
