BACKGROUND The development of regenerative therapy for human spinal cord injury(SCI)is dramatically restricted by two main challenges:the need for a safe source of functionally active and reproducible neural stem cells and the need of adequate animal models for preclinical *** reprogramming of somatic cells into neuronal and glial precursors might be a promising solution to the first *** use of non-human primates for preclinical studies exploring new treatment paradigms in SCI results in data with more translational relevance to human *** To investigate the safety and efficacy of intraspinal transplantation of directly reprogrammed neural precursor cells(drNPCs).METHODS Seven non-human primates with verified complete thoracic SCI were divided into two groups:drNPC group(n=4)was subjected to intraspinal transplantation of 5 million drNPCs rostral and caudal to the lesion site 2 wk post injury,and lesion control(n=3)was injected identically with the equivalent volume of *** Follow-up for 12 wk revealed that animals in the drNPC group demonstrated a significant recovery of the paralyzed hindlimb as well as recovery of somatosensory evoked potential and motor evoked potential of injured *** resonance diffusion tensor imaging data confirmed the intraspinal transplantation of drNPCs did not adversely affect the morphology of the central nervous system or cerebrospinal fluid *** immunohistochemical analysis showed that drNPCs maintained SOX2 expression characteristic of multipotency in the transplanted spinal cord for at least 12 wk,migrating to areas of axon growth *** Our data demonstrated that drNPC transplantation was safe and contributed to improvement of spinal cord function after acute SCI,based on neurological status assessment and neurophysiological recovery within 12 wk after *** functional improvement described was not associated with neuronal differentiation of th

Thermal state of ice-rich soils on the Tommot-Yakutsk Railroad right-of-way

This paper summarizes the results of investigations carried out in the northern section of the Tommot-Yakutsk Railroad in eastern Siberia, underlain by ice-rich permafrost. The ongoing permafrost and geotechnical research program in support of railroad construction includes assessment of the ground thermal state on the right-of-way and adjacent areas based on long-term field observations using the method of terrain thermal physics. These studies focus on the upper permafrost within the depth of annual temperature variation. In undisturbed areas, inter-annual variability of the ground thermal state shows little response to recent climatic variations. However, forest clearing and surface disturbance during right-of-way construction cause an increase in permafrost temperature, deepening of the active layer, thaw settlement, and water accumulation along the embankment. The active layer is thickest along the sun-exposed left berm and is thinnest along the more shaded right berm. Measures to prevent thaw-related embankment problems are recommended.

Current climate change effects on the ground thermal regime in Central Yakutia

The evolution of ground thermal state has been studied to assess impacts of current climatic warming on permafrost in Central Yakutia. The analysis of long-term data of regional weather stations has revealed one of the highest increasing trends in mean annual air temperature in northern Russia. A forecast of surface air temperature fluctuations has been made by applying a frequency analysis method. Monitoring of ground thermal conditions allows us to identify inter-annual and long-term variability among a wide range of natural conditions. Experimental research has indicated a long-term dynamics of ground thermal state evolution： ground temperatures at the depth of zero annual amplitude and seasonally thawed layer depth. Long-term variability of thaw depth shows near-zero to weak positive trends in small valleys in contrast to weak negative trends on slopes. With significant climatic warming, the thermal state of near-surface layers of permafrost demonstrates steadiness. Anthropogenic impacts on ground thermal regime in various terrain types have been qualitatively evaluated. Clear-cutting, ground cover stripping, and post-fire deforestation in inter-alas type terrains result in a significant increase of temperature and seasonal ground thaw depth, as well as adverse cryogenic processes. The dynamics of mean annual ground temperature in slash and burn sites have been evaluated in reference to stages of successive vegetation recovery.

Assessment of the thermal effect of large industrial buildings on permafrost foundation soils in Yakutsk

The results of monitoring studies of the ground thermal state beneath the Yakutsk Combined Heat and Power Plant buildings are presented in order to determine their structural stability and the possibility of further troublefree operation of the entire complex of the *** main causes for the formation of water-bearing taliks under and adjacent to the buildings are presented,and the factors influencing the existing geocryological situation are *** continuing stable state of the buildings demonstrates the possibility of using two construction principles on permafrost at one site,including preserving permafrost and accommodating permafrost thawing.

In the context of global climate change it is of practical importance to study changes in the thermal state of upper *** permafrost temperatures are significantly influenced by active-layer freezeback whose duration varies from year to *** later is complete freezing of the active layer,the shorter is the period of winter heat losses from the ground and the higher is its mean annual temperature,and vice *** have been conducted since 1996 in the watershed between the Lena and Kenkeme Rivers,30 km north-west of *** study area is situated within the zone of continuous permafrost with temperatures of-2.5 to-3.5℃.In Central Yakutia,duration of active-layer freezing primarily depends on two environmental factors:pre-winter soil moisture content and early-winter snow *** with low pre-winter soil moisture contents and thin snow covers have higher freezing rates and shorter duration of the freezeback *** such winters during the observation period,the rates of freezing and the lengths of the freezeback period were,respectively,1.0-3.8 cm/day and 60-110 days for the forest landscapes on sandy soils,1.3-3.5cm/day and 40-75 days for the forest landscapes on silty soils,and 0.8-3.7 cm/day and 70-110 days for the meadows on silty *** with high pre-winter soil moisture contents and thick snow covers are characterized by lower freezing rates and by longer duration of the freezeback *** the observation period,the rates of freezing and the lengths of the freezeback period in these winters were,respectively,0.4-2.5 cm/day and 83-145 days for the forest landscapes on sandy soils,0.5-2.1 cm/day and 80-130 days for the forest landscapes on silty soils,and 0.6-2.6cm/day and 95-155 days for the meadows on silty *** on the results of investigations at the experimental sites,empirical relationships between mean annual permafrost temperature and duration of active-layer freezing have been ***
