Fluorescence of Chlorophyll a for Discovering Inhibitors of Photosynthesis in Plant Extracts

Aims: Twelve crude extracts were prepared from different parts of Pluchea sagittalis, Cecropia palmata and Brachiaria brizantha, and their effect on the light reaction of photosynthesis. Study Design: Use of fluorescence of ChL a (Chlorophyll a) to detect inhibitors of photosynthesis in plant extracts. Place and Duration of Study: Instituto de Ciências Ambientais, Químicas e Farmacêuticas, Departamento de Ciências Exatas e da Terra, Universidade Federal de Sao paulo (UNIFESP), between July 2015 and January 2016. Methodology: The samples had their effect on the light reaction of photosynthesis studied by Chlorophyll a (Chl a) fluorescence transient by the use of chloroplasts isolated from spinach leaves. As negative controls we used the same amounts of DMSO that have been employed to solubilize the samples, and as positive control we have used 10 μM of the commercial herbicide DCMU (Diuron). Results: The extracts inhibited the photosynthesis, and their inhibition sites were located in the range of electron flow from OEC complex and between P680 to QA of PS II, and inhibited the photosystem II (PS II) by inducing the appearance of the transient bands K and J. Conclusion: Our results suggest an indicative that the photochemical apparatus probably is failing at the donor and acceptor sides of PSII, causing damage to electron transport during the photosynthesis phenomenon.

Cytomegalovirus (CMV) and Pneumocystis jirovecii fungus are the main opportunistic microorganisms that affect transplanted individuals. Immunosuppressive drugs administered to prevent organ rejection leave the immune system vulnerable to these infections. The present report is about a kidney transplanted patient using immunosuppressants who was diagnosed with cytomegalovirus and pneumocystosis requiring admission to the intensive care unit (ICU). Female patient, 57 years old, a kidney transplanted three years ago, with comorbidities, such as systemic arterial hypertension, hypertriglyceridemia and type 2 diabetes mellitus. She was admitted to the hospital in January 2020 with a history of diarrhea, cough, malaise and weight loss of seven kg in a month. She made continuous use of the immunosuppressants tacrolimus® and mycophenolate sodium (MFS). After five days of hospitalization, she was transferred to the ICU due to refractory diarrhea, worsening renal function and respiratory pattern, requiring mechanical ventilation. Chest tomography showed changes that led to the diagnostic hypothesis of CMV pneumonia or Pneumocystis jirovecii. Treatment with Ganciclovir® and Bactrim® was started. The bronchial lavage polymerase chain reaction test confirmed the infectious condition for CMV and Pneumocystis jirovecii. Despite the drug therapy instituted, there was no improvement in the infectious condition. The patient started to present a general and progressive worsening of the clinical picture with loss of renal graft function, respiratory failure, metabolic acidosis, hemodynamic instability and severe distributive shock, evolving to death. In the present report, it was observed that after late kidney transplantation the fragility of the immune system caused by the use of immunosuppressants contributed to the development of a severe infection with CMV and Pneumocystis jirovecii. In the present paper the main results of the pre-operational studies related to the energization maneuver of a Half-Wavelength Transmission Line (AC-Link), are presented. The simulations were performed in PSCAD with actual Brazilian system data. Specifically the following studies were performed: electromagnetic transient studies of energization maneuver considering no fault and the occurrence of fault along the trunk;dynamic analysis of generator units that will energize the AC-Link;TRV on the circuit-breaker that will energize the trunk. The main conclusion is that the AC-Link energization experiment can be implemented without jeopardizing or causing time-life reduction of the equipment involved in the experiment, which comprises the generator unit that will energize the trunk, the step-up transformer, the arresters that will be kept during the experiments, as well as the circuit-breaker that will energize and then trip the AC-Link.

Objective： The study aimed to evaluate the genotypic profiles of C. albicans （Candida albicans） sequentially isolated throughout the course of HIV infections, and to determine its MIC （minimal inhibitory concentrations） to AMB （amphotericin B）, FLC （fluconazole）, KTC （ketoconazole）, and ITC （itraconazole）. Design： samples were collected from the oral cavity of HIV-positive individuals during 4 years, with a sterilized swab. MIC was performed by using the microdilution method AFST/EUCAST. The genetic similarities within and between sequential clones of C. albicans were assessed by DNA fingerprinting using the random amplification ofpolymorphic DNA technique. Results： A total of 142 oral samples were isolated from 59 HIV-infected individuals who attempted up to five visits each, with or without symptoms of oropharyngeal candidiasis. Profile analysis revealed that yeasts isolated over sequential visits from symptomatic or asymptomatic individuals showed 78% or 87% relatedness, respectively. The degree of similarity among C. albicans was higher for isolates from colonization than for those from infection. Genetically identical C. albicans samples also formed connected subelusters in sequential visits. In regard to susceptibility profile, all isolates were susceptible to AMB, FLC, KTC, and ITC and maintained this pattern all along, no differences in MICs of any given antifungal compound were observed for sequential C. albicans isolates. Conclusions： These data suggest that genotype and susceptibility to antifimgal drugs were maintained over time in sequentially isolates of C. albicans colonization and a diverse evolutionary genetic trend in C. albicans sequentially isolated from the oral eandidiasis of HIV infected individuals.

Indiscriminate disposal of residues of domestic and/or industrial origin in aquatic ecosystems resulting from urban activities causes concern about their effects on the fauna. Substances absorbed by animals, such as those cited above, are metabolized by the liver, as it is the main target organ of several pollutants and also exercises basic vital functions in the organism. For this reason the liver is used as the main and most reliable biomarker in environmental studies. Thus the objective of the present study was to identify hepatic alterations resulting from exposure to water contaminated by a few commercial brands of biodegradable detergents and their effects when present together with several other contaminants found in nature. For such, a solution of 1 ppm of biodegradable detergents and water from an urban lake were tested on two widely distributed native Brazilian fish (Astyanax altiparanae and Prochilodus lineatus). Several alterations were found on the liver, including cytoplasm vacuolization, nuclear displacement, macrophage recruitment and reduced liver glycogen, showing a degenerative process in the liver of the fish, due to the action of contaminants.

In vitro cultivation is a technique that allows the production of great amount of plants. However, significant losses may occur during the acclimatization period (ex vitro stage). The objective of this study was to evaluate the effect of paclobutrazol, in the Murashige and Skoog culture medium, on growth, development, and survival of Zygopetalum crinitum plants. The experimental design (both in vitro and ex vitro) was completely randomized with four treatments: three paclobutrazol concentrations (0.5;1.0;and 1.5 mg L-1 active ingredient) and the control (without PBZ). Morphological changes promoted by the paclobutrazol application were observed in the seedlings;however, it did not result in higher seedling survival rates of the Zygopetalum crinitum orchid.

Photoluminescent Properties of CoMoO_(4)Nanorods Quickly Synthesized and Annealed in a Domestic Microwave Oven

A simple way to prepare α- and β-CoMoO4 nanorods is reported in this paper. CoMoO4xH2O nanorod precursors were obtained using the microwave-assisted hydrothermal (MAH) method. By annealing the as-prepared CoMoO44xH2O precursor at 600℃ for 10 min in a domestic microwave oven, α- and β-CoMoO4 nanorods were prepared. These powders were analyzed by X-ray diffraction (XRD), field emission scanning electron microscopy (FE-SEM), Fourier transform Raman microscopy and ultraviolet visible absorption spectroscopy (UV-vis spectra) as well as photoluminescence (PL) measurements. Based on the results, these materials revealed nanorod morphology. PL spectra obtained at room temperature for α- and β-CoMoO4 particles exhibited maximum components around the blue light emission. The results show that the domestic microwave oven has been successfully employed to obtain α- and β-CoMoO4 nanoparticles.

Complete dentures with Nobilo's sliding plates:A condylar position and electromyograph analysis

Objective: This study was performed with the purpose a report clinical cases involving condylar position and Nóbilo’s sliding plates. Background: Patients may present temporomandibular disorder (TMD), which is characterized by pain in masticatory muscles, temporomandibular joints and limited mandible movements. Treatment involving oral rehabilitation with the use of complete dentures with Nóbilo’s sliding plates has been an alternative for the treatment of TMD. Materials and Methods: Nine patients with a history of TMD signs and symptoms (RDC/TMD)—in particular, muscular and articular pain—received pain evaluations, mandibular movement analysis, transcranial radiography and an electromyographic analysis of the masseter and temporalis muscles before and after the use of complete dentures with Nóbilo’s sliding plates. Results: After treatment, the patients had reduced painful symptoms, an increase in mandibular movement and adequate electromyographic activity. The normalized EMG data were ta-bulated and analyzed statistically using SPSS version 17.0 for Windows (SPSS Inc., Chicago, IL, USA), and the values were compared by umpaired independent Student’s t-test. Conclusion: The use of Nóbilo’s sliding plates promoted improvements in electromyographic activity, muscular pain and man-dibular movement.
