Upregulation of the APE1 and H2AX genes and miRNAs involved in DNA damage response and repair in gastric cancer

Gastric cancer remains one of the leading causes of cancer-related death worldwide,and most of the cases are associated with Helicobacter pylori *** bacterium promotes the production of reactive oxygen species(ROS),which cause DNA damage in gastric epithelial *** this study,we evaluated the expression of important genes involved in the recognition of DNA damage(ATM,ATR,and H2AX)and ROS-induced damage repair(APE1)and the expression of some miRNAs(miR-15a,miR-21,miR-24,miR-421 and miR-605)that target genes involved in the DNA damage response(DDR)in 31 fresh tissues of gastric *** v3.1.1 was used to construct the postulated miRNA:mRNA interaction *** performed by real-time quantitative PCR exhibited significantly increased levels of the APE1(RQ=2.55,p<0.0001)and H2AX(RQ=2.88,p=0.0002)genes beyond the miR-421 and miR-605 in the gastric cancer *** addition,significantly elevated levels of miR-21,miR-24 and miR-421 were observed in diffuse-type gastric *** analysis reinforced some of the gene:gene(ATM/ATR/H2AX)and miRNA:mRNA relationships obtained also with the interaction ***,our findings show that tumor cells from gastric cancer presents deregulation of genes and miRNAs that participate in the recognition and repair of DNA damage,which could confer an advantage to cell survival and proliferation in the tumor microenvironment.

Factors Associated with Physical-Activity Performance by Older Individuals in a Medium-Sized City in S^o paulo State, Brazil

Physical activity has been scientifically discussed as fundamental in the process of healthy ageing. Hence, this study aimed at determining the factors that influence older people to perform physical activities. The complete IPAQ （International Physical Activity Questionnaire） was applied to a population-based sample consisting of 364 elderly persons in the city of Botucatu, SAo paulo, Brazil. Days of physical activity performed by the older people were considered by taking into account household and leisure activities. Models for count data were fitted by including socio-demographic variables as well as those related to life satisfaction. It was shown that housework physical-activity performance is associated with female, who predominantly showed to be more active in all levels. Male seemed to be more predisposed to perform lighter recreation, sports and leisure-time physical activities, such as walking. Additionally, poor schooling showed to he decisive for not performing physical activities both at home and during leisure.

Hepatic ischemic preconditioning increases portal vein flow in experimental liver ischemia reperfusion injury

BACKGROUND: Ischemic preconditioning(IPC) has been shown to decrease liver injury and to increase hepatic microvascular perfusion after liver ischemia reperfusion. This study aimed to evaluate the effects of IPC on hemodynamics of the portal venous system. METHODS: Thirty-two rats were randomized into two groups: IPC group and control group. The rats of the IPC group underwent IPC by 10 minutes of liver ischemia followed by 10 minutes of reperfusion before liver ischemia, and the rats of the control group were subjected to 60 minutes of partial liver ischemia. Non-ischemic lobes were resected immediately after reperfusion. The animals were studied at 4 hours and 12 hours after reperfusion. Mean arterial pressure, heart rate, portal vein flow and pressure were analyzed. Blood was collected for the determination of the levels of aspartate aminotransferase, alanine aminotransferase, calcium, lactate, pH, bicarbonate, and base excess. RESULTS: IPC increased the mean portal vein flow at 4 hours and 12 hours after reperfusion. IPC recovered 78% of the meanportal vein flow at 12 hours after reperfusion. IPC decreased the levels of aspartate aminotransferase, alanine aminotransferase and lactate, and increased the levels of ionized calcium, bicarbonate and base excess at 12 hours after reperfusion. CONCLUSIONS: This study demonstrated that IPC increases portal vein flow and enhances hepatoprotective effects in liver ischemia reperfusion. The better recovery of portal vein flow after IPC may be correlated with the lower levels of transaminases and with the better metabolic profile.

Healthcare Professionals’ Adherence to Contact Precautions at a Maternal and Child Teaching Hospital

Background: Healthcare-associated infections affect hundreds of millions of patients worldwide. Children have greater susceptibility to healthcare-associated infections due to the immaturity of their immune system. Contact precautions aim to promote safety, protection and prevention of contamination. Thus, the objective of this study was to verify adherence to contact precaution measures, as well as compliance to the use of personal protective equipment. Methods: This cross-sectional observational study was carried out from July to October 2019 using a checklist to evaluate health professionals’ adherence to hand hygiene procedures and the use of gloves and surgical gowns when assisting children on contact precautions. Results: A total of 941 observations were carried out in a total of 300.532 hours. Hand hygiene was performed before and after contact with the patient in 58.84% and 75.09% of the cases, respectively and a surgical gown was used in 86.40% of the cases. The use of gloves was the variable most adhered to by professionals (87.57%). Intensive care unit professionals were the care workers who most complied with the regulation regarding hand hygiene after contact with the patient (p = 0.009) and the use of the surgical gown (p < 0.001). The correct hand hygiene technique was the recommendation with least adherence. Non-compliance to the hand hygiene technique was statistically significant among intensive care unit professionals (p = 0.002). Conclusions: Adherence to hand hygiene before contact with the patient and compliance with the hand hygiene technique were neglected by most professionals. However, there was good adherence to the use of surgical gloves and gowns, as well as high compliance to the techniques of removing these items. .

Hybrid Rice Seed Treatment with Pesticides Improves Its Physiological Quality and Performance

Pathogens in the soil or transmitted by seeds can reduce the stand of plants, affecting the yield of the crop. Furthermore, in the case of rice, the low temperature is a limiting factor in culture. The treatment of seeds with insecticides is an alternative to reduce the damage caused by pests incidents during the early stages of plant development and in some cases, may assist in the germination of seeds exposed to low temperatures. In this sense, the objective of this study was to evaluate the influence of seed treatment with insecticides in the physiological quality and yield of hybrid rice seeds, cv. Inov CL. For this, three experiments were conducted with treated seeds in different doses of thiamethoxam + lambda-cyhalothrin and fipronil totaling seven treatments, including the control (no product). In the first experiment, it was determined seed germination to 10°C, 13°C, 16°C, 20°C and 25°C. In the second experiment, it was assessed seed vigor through accelerated aging, cold test and emergency field. The third experiment was carried out into the field to observe the number of panicles m-2 and crop yield. It is concluded that treatment of hybrid rice seeds, cv. Inov CL with thiamethoxam + lambda-cyhalothrin and fipronil increases the germination percentage of seed and early seedling performance. Treatment of hybrid rice seeds, cv. Inov CL with thiamethoxam + lambda-cyhalothrin provides greater germination and early seedling performance compared to treatment with fipronil. Treatment of hybrid rice seeds, cv. Inov CL with thiamethoxam + lambda-cyhalothrin in dosages of 500 mL 100 kg-11 of seeds or 500 mL 45 kg-1 of seeds increases grain yield.

Comparative Dimensional and Microscopic Analysis of the Strength and Hardness of Internal Fixation Plates

The fixation stability achieved with the use of plates and screws in oral and maxillofacial surgery is a decisive factor in treatment success. The mechanical and structural properties of the internal fixation materials have direct influence on the dimensional stability and resistance of a fixation system, thus influencing treatment outcomes. This study proposed to analyze the dimensional and resistance patterns of titanium plates used for obtaining stable fixation in orthognathic surgery and craniofacial trauma. For this study, 30 conventional 2.0 mm straight four hole plates with bridge from three brands commercialized in Brazil, were subjected to macroscopic, microscopic, strength and hardness analysis. The dimensional measurements were performed using a digital caliper. Energy-dispersive X-ray spectroscopy analysis was performed by scanning electron microscopy to analyze the chemical composition of the samples. The mechanical resistance tests were performed with a universal testing machine. The samples were then submitted to Vickers hardness analysis, complying with the standards of ASTM E92. The data collected from the dimensional study was submitted to statistical analysis of the coefficient of variation, while the values obtained during the mechanical tests were analyzed by variance (ANOVA) and Tukey’s test (p 0.05). The sample groups presented different performances in resistance, hardness, size and surface, even though they were reported to be chemically similar compounds that allowed us to conclude the plates from Group 2 were more resistant than groups 1 and 3.

Disease-resistant genes play an important role in defending against a variety of pathogens and insect pests in plants. Most of the disease-resistant genes encode pro- teins with conserved leucine rich repeat and nucleotide binding site domains. In this study, we cloned and char- acterized gene-resistant analogs （RGAs） from Eucalyptus grandis using degenerate PCR, with primers specifically targeting these two domains. The amplified fragments were cloned into the pGEM-T vector and transformed into Escherichia coli. Among the 90 clones obtained, 13 were sequenced and compared with each other and with previ- ously identified gene-resistant diseases. A BLASTX search in GenBank revealed high similarities among the con- served domains of these cloned genes with RGA genes. Some clones, however, showed no significant similarity with DNA sequences in GenBank. Southern blotting ana- lysis identified several polymorphic RFLP loci between distinct genotypes. However, none of them co-segregated with the Puccinia psidii Winter resistance gene 1 （Ppr1） in a population study.

Isoenzyme Expression in Bean Seed Germination Treated with Thiamethoxam with and without Drought Stress

The thiamethoxam acts as enhancer, allowing maximizing the expression of seed vigor. Isozymes are products of gene expression, highly influenced by the environment, because genes that control its expression are manifested in certain stages of development and in specific organs and tissues, or even under the action of certain stimulus. This study evaluated the isoenzyme expression in bean seedlings from seeds treated with thiamethoxam. Bean seeds were used, cultivar Pérola and IAPAR Siriri, submitted or not to drought stress. Seeds were treated at doses of thiamethoxam: 0, 1, 2, 3, 4 and 5 mL·kg-1 of seed. Isozymes were extracted from seedlings collected five days after sowing. We evaluated the expression of isoenzymes: Glutamate Oxalacetate transaminase (GOT), acid phosphatase (FAC), peroxidase (PO), Esterase (EST), malate dehydrogenase (MDH) and glutamate dehydrogenase (GTDH). Interpretation of the results was based on visual analysis of electrophoresis gels, considering the presence/absence and the intensity of each electrophoretic band. Bean seeds treated with thiamethoxam with doses up 3 mL·kg-1 of seed are higher germination untreated, even when submitted to drought stress. The expression of isozymes GOT, FAC, PO, EST, MDH and GTDH is not affected significantly in bean seedlings of Pérola and IPR Siriri under different doses of thiamethoxam with and without drought stress.

The EFVM （Vitoria Minas Railroad） is one of the main railways in Brazil. It transports freight trains of ore, 220 wagons each. These wagons have 2 boogies of 2 axles each and 32 metric tons on metre gauge. Elastic strains were measured on a special part of this railway due to these trains. The main load to evaluate stresses and strains was a G 16 Locomotive, a C-C kind from Vale, a Brazilian Company. The measurements were obtained by dynamic deflectometer installed on a main line of this railway, near Ipatinga, a city from Minas Gerais, one of Brazil states. This track was equipped to obtain stresses under an equal repeated static load A simulation of the stresses was made under critical strain by Ferrovia 1.0 software. It was also made an evaluation of unequal results from neighbor sleepers taking in comparison two equipped parts of this railway, one with compacted ballast and no compaction to the other. The results were strain limited, avoiding breakage or damage to the studied rails. This work analyses these measurements focusing on the improvement of track quality.
