Comparison of p53 and prohibitin expression in the spectrum of hepatitis, cirrhosis, and hepatocellular carcinoma

Aim:To investigate the correlation of p53 and prohibitin (PHB) expression in the spectrum of hepatitis, cirrhosis, and human hepatocellular carcinoma (HCC).methods:Hepatic biopsies from patients with HCC (n = 60), cirrhosis (CIR,n= 30), hepatitis C virus (HCV,n = 30), and normal livers (NL,n = 20) were examined for immunohistochemical expression of RelA/p65, tumor necrosis factor receptor-1 (TNFR1), TNF-related apoptosis-inducing ligand (TraIL), p53, and PHB. The samples were also analysed by nuclear factor kappa B (NFκB) Southwestern histochemistry and a transferase-mediated dUTP-biotin nick-end labelling ***: expression of NFκB and RelA/p65 was detected increasingly from NL to CIR, but had a diminished labelling in the HCC cases (P expression levels of TNFR1 and TraIL followed the same pattern (P eased in HCC, but was progressively reduced from CIR to NL (P ear expressions were ampliifed in cases of HCC, but diminished in NL, HCV, and CIR (Pese results suggest that in addition to well-understood sequences of proinlfammatory events such as TNF-induced NFκB activation and NFκB/TraIL pathway-mediated apoptosis, development of HCC is also inlfuenced by regulation ofp53 andPHB tumour suppressor function. Additional studies are necessary to explain the contradictory mechanisms of the tumour microenvironment observed in the sequence of HCV, CIR, and HCC.

Identification of IL11ra and meLK amplification in gastric cancer by comprehensive genomic profiling of gastric cancer cell lines

AIM To identify common copy number alterations on gastric cancer cell *** Four gastric cancer cell lines(ACP02, ACP03, AGP01 and PG100) underwent chromosomal comparative genome hybridization and array comparative genome hybridization. We also confirmed the results by fluorescence in situ hybridization analysis using the bacterial artificial chromosome clone and quantitative real time PCR *** The amplification of 9p13.3 was detected in all cell lines by both methodologies. An increase in the copy number of 9p13.3 was also confirmed by fluorescence in situ hybridization analysis. Moreover, the interleukin 11 receptor alpha(IL11ra) and maternal embryonic leucine zipper kinase(meLK) genes, which are present in the 9p13.3 amplicon, revealed gains of the meLK gene in all the cell lines studied. Additionally, a gain in the copy number of IL11 ra and meLK was observed in 19.1%(13/68) and 55.9%(38/68) of primary gastric adenocarcinoma samples, respectively. CONCLUSION The characterization of a small gain region at 9p13.3 in gastric cancer cell lines and primary gastric adenocarcinoma samples has revealed meLK as a candidate target gene that is possibly related to the development of gastric cancer.

Background The increase in the frequency and intensity of droughts is pointed out as one of the main factors altering biogeochemical cycles in the Amazon *** eco-nutritional approach using X-ray fluorescence micro-analysis(μXRF)is proposed to verify the long-and short-term effects of droughts on the growth and xylem nutrient concentrations of Cedrela fissilis *** Fourteen radii were selected from a tree-ring width chronology and X-rayed by Itrax *** of ring width,wood density,and concentrations of aluminum(Al),phosphorus(P),sulfur(S),calcium(Ca),potassium(K),manganese(Mn),iron(Fe)and strontium(Sr)together with Al/Ca,Ca/Mn,K/Ca,Sr/Ca and Mn/S ratios were constructed and correlated with precipitation,temperature,the difference between precipitation and potential evapotranspiration(P-PeT)and standardized precipitation-evapotranspiration index(SPeI).Results During dry years,*** showed narrower,less dense rings,lower Al,P,S and Ca,and higher K and Fe concentrations(the opposite was found in wet years).Ring width decreased(together with Al,P,S,K,Ca,Mn,Fe,Sr,Al/Ca,K/Ca and Sr/Ca)and wood density increased(together with Ca/Mn and Mn/S),which was associated with an increase in evapotranspiration and temperature over time,mainly since *** fissilis showed a tendency to increase its capacity for resistance,and a recovery and resilience in growth over time associated with responses in Al,Ca,P and ***,it showed a risk in the capacity for recovery of the pre-drought density values,associated with unsatisfactory responses in Al,Ca,K,Fe and *** This study is the first attempt to analyze tree-ring nutritional evidences of *** trees to climate sensitivity and resilience to drought,based on long-term data from seasonal moist tropical forests of the *** data suggested that *** is undergoing alterations in the concentration,use and redistribution of nutrients associated with increasing wood density and d

AIM:To analyze small intestinal bacterial overgrowth in school-aged children and the relationship between hydrogen and methane production in breath ***:This transversal study included 85 children residing in a slum and 43 children from a private school,all aged between 6 and 10 years,in Osasco,*** characterization of the groups,data regarding the socioeconomic status and basic housing sanitary conditions were *** data was obtained in children from both *** children completed the hydrogen(H 2) and methane(CH 4) breath test in order to assess small intestinal bacterial overgrowth(SIBO).SIBO was diagnosed when there was an increase in H 2 ≥ 20 ppm or CH 4 ≥ 10 ppm with regard to the fasting value until 60 min after lactulose ***:Children from the slum group had worse living conditions and lower nutritional indices than children from the private *** was found in 30.9%(26/84) of the children from the slum group and in 2.4%(1/41) from the private school group(P = 0.0007).Greater hydrogen production in the small intestine was observed in children from the slum group when compared to children from the private school(P = 0.007).A higher concentration of hydrogen in the small intestine(P e colon(P erved among the children from the slum group with SIBO when compared to children from the slum group without *** production was observed in 63.1%(53/84) of the children from the slum group and in 19.5%(8/41) of the children from the private school group(P methane production was observed in 38/58(65.5%) of the children without SIBO and in 15/26(57.7%) of the children with SIBO from the *** production of hydrogen was lower in methaneproducing children(P = 0.017).CONCLUSION:Children who live in inadequate environmental conditions are at risk of bacterial overgrowth and methane *** is a substrate for methane production in the colon.

Alterations in epigenetic control of gene expression play an important role in many diseases, including gastric cancer. Many studies have identified a large number of upregulated oncogenic mi RNAs and downregulated tumour-suppressor mi RNAs in this type of cancer. In this review, we provide an overview of the role of mi RNAs, pointing to their potential to be useful as diagnostic and/or prognostic biomarkers in gastric cancer. Moreover, we discuss the influence of polymorphisms and epigenetic modifications on mi RNA activity.

Healthcare access and the patterns of maternal health care utilization among poor and non-poor women living in urban areas in Portugal

Introduction: Studies on attitudes and practices are increasingly used but not specifically related to the motivations for the use of reproductive health care among women of fertile age, living in urban areas and in different social contexts. Objectives: The aim of this study was to estimate the associations between the variables of social status (degree of poverty in the studied groups) and the variables of fecundity (representations, tensions, practices and control of fertility) and, in addition, to compare access to health care in the different studied groups, assessing the association between use of maternal health care and poverty in urban areas. design: A case-control study was conducted in the Municipality of Lisbon, Portugal, with a total sample of 1513 women of fertile age: 499 cases of women considered very poor were selected from the database of beneficiaries of RSI (Social Welfare Payment for Inclusion);1014 controls (two controls for each selected case), divided as 507 poor women selected from the other beneficiaries of Santa Casa da Misericórdia in Lisbon and 507 non-poor women selected from four Health Centers from the Municipality of Lisbon, Portugal. A total of 1054 women answered the questionnaire: 304 cases (response rate of 61%) and 750 (response rate of 74%) controls. The statistical analysis involved descriptive analysis and multinomial logistic regression. Results: The analysis confirms the association between poverty and patterns and representations of fecun dity regarding pregnancy planning. The results of this study thus show the existence of different distributions on several variables and the gradients of poverty. Regarding access to health care, the major impact of poverty on women is limiting access to pharmaceuticals. The incapacity to afford the cost of health care appears as a central aspect of access to health care. The severe acute respiratory syndrome coronavirus 2(SARS-CovV-2)infection is associated with a hyperinflammatory state and lymphocytopenia,a hallmark that appears as both signature and prognosis of disease severity *** cytokine storm and a sustained inflammatory state are commonly associated with immune cell depletion,it is still unclear whether direct SARS-Cov-2 infection of immune cells could also play a role in this scenario by harboring viral *** found that monocytes,as well as both B and T lymphocytes,were susceptible to SARS-Cov-2 infection in vitro,accumulating double-stranded RNA consistent with viral RNA replication and ultimately leading to expressive T cell *** addition,flow cytometry and immunofluorescence analysis revealed that SARS-Cov-2 was frequently detected in monocytes and B lymphocytes from coronavirus disease 2019(CoVID-19)*** rates of SARS-Cov-2-infected monocytes in peripheral blood mononuclear cells from CoVID-19 patients increased over time from symptom onset,with SARS-CoV-2-positive monocytes,B cells,and CD4+T lymphocytes also detected in postmortem lung *** results indicated that SARS-CoV-2 infection of blood-circulating leukocytes in covID-19 patients might have important implications for disease pathogenesis and progression,immune dysfunction,and virus spread within the host.
