In vivo study of conductive 3D printed PCL/MWCNTs scaffolds with electrical stimulation for bone tissue engineering

Critical bone defects are considered one of the major clinical challenges in reconstructive bone *** combination of 3D printed conductive scaffolds and exogenous electrical stimulation(ES)is a potential favorable approach for bone tissue *** this study,3D conductive scaffolds made with biocompatible and biodegradable polycaprolactone(PCL)and multi-walled carbon nanotubes(MWCNTs)were produced using the extrusion-based additive manufacturing to treat large calvary bone defects in *** results show that the use of PCL/MWCNTs scaffolds and ES contributes to thicker and increased bone tissue formation within the bone *** and mineralization are also significantly promoted using high concentration of MWCNTs(3 wt%)and ***,scaffolds favor the tartrate-resistant acid phosphatase(TRAP)positive cell formation,while the addition of MWCNTs seems to inhibit the osteoclastogenesis but present limited effects on the osteoclast functionalities(receptor activator of nuclear factor κβ ligand(RANKL)and osteoprotegerin(OPG)expressions).The use of ES promotes the osteoclastogenesis and RANKL expressions,showing a dominant effect in the bone remodeling *** results indicate that the combination of 3D printed conductive PCL/MWCNTs scaffold and ES is a promising strategy to treat critical bone defects and provide a cue to establish an optimal protocol to use conductive scaffolds and ES for bone tissue engineering.

Feasibility of performing the 3-minute step test with remote supervision in children and adolescents with cystic fibrosis: A comparative study

Importance:The 3-min step test is a simple option to monitor submaximal exercise capacity,although its use via remote video monitoring has not been investigated in children with cystic fibrosis(CF).Objective:This study aimed to assess the feasibility and reproducibility of performing the 3-min step test with remote ***:A cross-sectional study including CF patients(6-18 years)from two CF services were ***,anthropometric,clinical,and lung function data were collected and two 3-min step tests were performed:(i)in-person supervision,and(ii)remotely supervised by video *** and after the tests,heart rate(HR),oxygen saturation(SpO_(2)),and the Borg score for dyspnea and lower limb fatigue were ***:Twenty-three patients(10.7±3.7 years)with a mean FEV1 of 89.5%±23.2%were *** were no significant differences between tests,with mean differences(95%confidence intervals)in final HR of-3.3(-8.9,2.4),change in HR of-1.9(-6.1,2.1),final SpO_(2) of 0.3(-0.4,1.0),and final dyspnea of 0.1(-0.8,0.9).The intraclass correlation coefficient was 0.852(final HR),0.762(final SpO_(2)),and 0.775(final lower limb fatigue).Significant and moderate correlations were found between tests for final HR(r=0.75),change in HR(r=0.61),and final SpO_(2)(r=0.61).The Bland-Altman analysis showed a mean difference in final SpO_(2) between tests of 0.3%(limit of agreement-3.0%,3.5%).Interpretation:Physiological responses between tests were similar,indicating it was feasible to perform the 3-min step test with remote supervision in CF children.

Esophageal atresia (EA) with or without tracheoesophageal fistula (TEF) is the most common congenital anomaly of the esophagus. The improvement of survival observed over the previous two decades is multifactorial and largely attributable to advances in neonatal intensive care, neonatal anesthesia, ventilatory and nutritional support, antibiotics, early surgical intervention, surgical materials and techniques. Indeed, mortality is currently limited to those cases with coexisting severe life-threatening anomalies. The diagnosis of EA is most commonly made during the first 24 h of life but may occur either antenatally or may be delayed. The primary surgical correction for EA and TEF is the best option in the absence of severe malformations. There is no ideal replacement for the esophagus and the optimal surgical treatment for patients with long-gap EA is still contro-versial. The primary complications during the postoperative period are leak and stenosis of the anastomosis, gastro-esophageal reflux, esophageal dysmotility, fistula recurrence, respiratory disorders and deformities of the thoracic wall. Data regarding long-term outcomes and follow-ups are limited for patients following EA/TEF repair. The determination of the risk factors for the complicated evolution following EA/TEF repair may positively impact long-term prognoses. Much remains to be studied regarding this condition. This manuscript provides a literature review of the current knowledge regarding EA.

Tertiary peritonitis: A disease that should not be ignored

Intra-abdominal infections can be classified into uncomplicated or complicated(peritonitis). Peritonitis is divided into primary, secondary, and tertiary. Tertiaryperitonitis is the less common but the most severe among peritonitisstratifications, being defined as a recurrent intra-abdominal infection that occurs48 h after a well-succeeded control of a secondary peritonitis. This disease has acomplex pathogenesis that is closely related to the capacity of the peritonealcavity to activate immunological processes. Patients who progress to persistentperitonitis are at an increased risk of developing several infectious complicationssuch as sepsis and multiple organ failure syndrome. Moreover, tertiary peritonitisremains an important cause of hospital death mainly among patients withassociated risk factors. The microbiological profile of organisms causing tertiaryperitonitis is often different from that observed in other types of peritonitis. Inaddition, there is a high prevalence of multidrug-resistant pathogens causing thiscondition, and an appropriate and successful clinical management depends on anearly diagnosis, which can be made easier with the use of clinical scorespresenting a good prediction value during the intensive care unit ***, immediate therapy should be performed to control theinfectious focus and to prevent new recurrences. In this sense, the treatment isbased on initial antimicrobial therapy and well-performed peritoneal drainage.

Surface runoff and soil erosion in a natural regeneration area of the Brazilian Cerrado

The Brazilian Cerrado has been converted to farmland,and there is little evidence that this expansion will decrease,mainly because agriculture is the country's main economic ***,the impacts of intense modification of land use and land cover on surface runoff and soil erosion are still poorly un-derstood in this ***,we assessed surface runoff and soil loss in a woodland Cerrado area under a former pasture area,which was abandoned and has undergone a natural regeneration process for 7 years(RC).Its results were compared with that found in an undisturbed area of woodland Cerrado(CE),40-month-old eucalyptus(3.0×1.8 m)(EU),and pasture under rotational grazing(PA).The study was conducted on Red Acrisol located in the Brazilian *** performed rainfall simulations on a plot of 0.7 m2 and using three constant rainfall intensities of 60,90,and 120 mm h-1 for 1 *** each rainfall intensity,we carried out four repetitions using different plots in each treatment,i.e.12 plots per treat-ment studied and 48 plots in *** noted that the soil physical properties were improved in RC and,consequently,water infiltration and soil erosion control;RC presented surface runoff and soil loss different from EU and PA(a=0.05).The macroporosity and soil bulk density affected surface runoff in RC and PA because the RC was used as pasture and is currently regenerating back to the cerrado *** the rainfall intensity increased,EU became more similar to PA,which showed the highest surface runoff and soil *** findings indicate that natural regeneration processes(pasture to the cerrado vegetation)tend to improve the soil ecosystem services,improving infiltration and reducing surface runoff and soil erosion.

Indicator of Agriculture Vulnerability to Climatic Extremes. A Conceptual Model with Case Study for the Northeast Brazil

The Northeast Brazil (NEB) is known for its temporal and spatial variability of precipitation. Several studies have investigated this variability in order to understand the damaging episodes such as droughts and floods. The phenomenon of drought in the NEB is a complex topic due to affecting?millions of people and being?the object of study in several fields of knowledge. One way to try to argue about this phenomenon is through the concept of vulnerability. The “operability” of this broad concept in natural disasters is a complex task. In order to measure an indicator of vulnerability it is necessary large amount of data from different areas of knowledge, among which?include: meteorology, socio-environmental, economic, public health, among other areas. The main objective of this study is to create an index of vulnerability to climate extremes (drought and flood) for the NEB and to compare this rate with those found in the scientific literature. The data that will be used in this study are from ANA (Agência Nacional das águas), IBGE (Instituto Brasileiro de Georgrafia e Estatística), and Ministry of National integration.

Microcalorimetric Study of Acetylcholine and Acetylthiocholine Hydrolysis by Acetylcholinesterase

Acetylcholinesterase (AChE) is an important enzyme responsible for the cleavage of acetylcholine. Studies of the activity of this enzyme use an artificial substrate, acetylthiocholine, because a product of its catalysis, thiocholine, readily generates a light absorbing product upon reaction with Elman’s reagent 5,5’-dithiobis-(2-nitrobenzoic acid (DTNB). The hydrolysis of acetylcholine cannot be assayed with this method. The isothermal titration calorimetry can assay the hydrolysis of both substrates, without requiring additional reagents other than the enzyme and the substrate. To compare kinetic values obtained in the hydrolysis of acetylcholine (ACh) and acetylthiocholine (ATCh), with carbaryl acting as inhibitor, a calorimetric technique was used to evaluate kinetic properties of the two reactions. This method can show the hydrolysis of both substrates by the heat exchange that occurs during catalysis. In addition, it allowed the assessment of the AChE inhibition by carbaryl, a common insecticide. The results show a similarity between values obtained with both substrates, which are slightly higher for acetylcholine, the enzyme natural substrate. Enzymatic parameters values from ATCh and ACh were similar to each other and inhibitory constants using carbaryl were also similar, displaying that any approach to ACh is feasible using ATCh. The results obtained from ITC show the precision achieved by the calorimetric method.

Association between Helicobacterpylori, Epstein-Barr virus, human papillomavirus and gastric adenocarcinomas

AIM TO correlate Helicobacter pylori （H. pylorli）, EpsteinBarr virus （EBV） and human papillomavirus （HPV） with gastric cancer （GC） cases in Para State, *** Tissue samples were obtained from 302 gastric adenocarcinomas. A rapid urease test was used to detect the presence of H. pylori, and the presence of the cagA gene in the HP-positive samples was confirmed by PCR. An RNA in situ hybridization test designed to complement Eberl RNA was used to detect the presence of EBV in the samples, and the L1 region of HPV was detected using nested PCR. Positive HPV samples were genotyped and analyzed for E6 and E7 viral gene expression. Infections were also correlated with the clinical and pathological characteristics of the *** The majority of the 302 samples analyzed were obtained from men （65%） aged 55 years or older （67%） and were classified as the intestinal subtype （55%）. All three pathogens were found in the samples analyzed in the present study （H. pylori： 87%, EBV： 20%, HPV： 3%）. Overall, 78% of the H. pylori-positive （H. pylorl＋） samples were cagA＋（H. pylori-cagA＋）, and there was an association between the cytotoxic product of this gene and EBV. Coinfections of H. pylori-cagA＋ and EBV were correlated with the most advanced tumor stages. Although only 20% of the tumors were positive for EBV, infection with this virus was associated with distant metastasis. Only the HPV 16 and 18 strains were found in the samples, although no expression of the E6 and E7 oncoproteins was detected. The fundus of the stomach was the region least affected by the *** HPV was not involved in gastric tumorigenesis. Prophy- lactic and therapeutic measures against H. pylori and EBV may prevent the development of GC, especially the more aggressive forms.

This research aimed to analyze the possibility to estimate and automatically map large areas of soybean cultivation through the use of MODIS(Moderate-Resolution Imaging Spectroradiometer)*** major techniques were used:GEOgraphic-Object-Based Image Analysis(GEOBIA)and Data Mining(DM).In order to obtain the images,the segmentation algorithm implemented by Definiens Developer was used.A decision tree(DT)was created from a training set previously ***-series of images from the MODIS sensor aboard the Terra satellite were acquired in order to represent the wide variation of the vegetation pattern along the soybean crop *** time-series data were used only for the CEI ***,to compare the results obtained from GEOBIA,the slicing technique was used at the CEI *** the training,the DT was applied to the vegetation indices generating the thematic map of the spatial distribution of *** accordance with the error matrix and kappa parameter analysis,tests for statistical significance were *** indicate that the classification achieved by Kappa coefficients is *** short,the obtained results proved that combining vegetation indices and time-series data using GEOBIA return promising results for mapping soybean plantation on a regional scale.
