Isolation of high quality DNA from multiple samples can be both time-consuming and expensive. We have developed a combined protocol to reduce the time component of the hexadecyltrimethylammonium bromide （CTAB） extraction method and reduced costs by regenerating the silica columns used to purify genomic DNA. We present data that shows, by in- creasing the temperature used during the CTAB method, the time required to extract crude genomic DNA can be reduced. We show that silica columns can be regenerated using HCI and still maintain their DNA-binding capacity. Furthermore, we show both spectrophotometrically, and by restriction enzyme cutting, that the quality of the eluted DNA is high. Critically, using both genomic DNA from pea and perennial ryegrass we demonstrate, using species-specific PCR primers, that there is no carry-over of DNA from repeated use of a single column. The main advantages of the method are high yield, high quality, cost effectiveness and time-saving. This method could satisfy demand when large numbers of plant genomic DNA samples are required, for example from targeting induced local lesions in genomes （TILLING） populations.

Drought periods are becoming more extreme worldwide and the ability of plants to contribute towards atmospheric flux is being compromised. Properly functioning stomata provide an exit for water that has been absorbed by the roots, funneled into various cell parts, and eventually released into the atmosphere via transpiration. By observing the effects that weather conditions such as climate change may have on stomatal density, distribution, and functioning, it may be possible to elucidate a portion of the mechanisms trees use to survive longer periods of water stress. This study analyzed stomatal density （SD）, stomatal conductance （gs ）, CO2 assimilation （A）, instantaneous water-use efficiency （WUEi ）, and transpiration （E） rates in six native tree species in the Midwestern USA and showed that trees within the same ecotype followed similar trends, but that trees within the same family did not when exposed to identical greenhouse conditions. Naturally drought tolerant tree species demonstrated lower g s and higher WUEi , while intolerant species had higher SD. This study showed negative or no correlation between SD and g s , A, E, and WUEi and positive correlations between E and A and gs and E.

Pancreatic cancer ranks tenth in terms of newly diagnosed cases, but just 10%–15% of these patients can undergo resection. Survival after curative surgery is dismal, as recurrences occur either locally or in the liver. Adjuvant treatment with either chemotherapy or chemoradiation (with or without maintenance chemotherapy) has been employed, to improve the poor prognosis. Justification for the use of chemoradiation, with follow on chemotherapy, is based on the results of an underpowered 1987 GITSG study, which closed prematurely and compared intervention to observation. There has been no survival advantage demonstrated in the one randomized controlled trial that examined chemoradiation compared to chemotherapy. There is a clear cut survival advantage however with chemotherapy compared to observation, based on the results from two large randomized controlled trials, and supported by an individual patient data meta-analysis. The standard of care for adjuvant therapy based on level I evidence (from the ESPAC-1 trial) is post operative chemotherapy using 5-Fluorouracil with folinic acid providing a best estimate of 29% five years survival.

Carbon dioxide-abated hydrogen can be synthesised via various processes,one of which is sorption enhanced steam methane reforming(SE-SMR),which produces separated streams of high purity H_(2) and CO_(2).Properties of hydrogen and the sorbent material hinder the ability to rapidly upscale SE-SMR,therefore the use of artificial intelligence models is useful in order to assist scale *** of a data driven soft-sensor model over ther-modynamic simulations,is the ability to obtain real time information dependent on actual process *** this study,two soft sensor models have been developed and used to predict and estimate variables that would otherwise be difficult direct *** artificial neural networks and the random forest models were devel-oped as soft sensor prediction *** were shown to provide good predictions for gas concentrations in the reformer and regenerator reactors of the SE-SMR process using temperature,pressure,steam to carbon ratio and sorbent to carbon ratio as input process *** models were very accurate with high R^(2) values,all above 98%.However,the random forest model was more precise in the predictions,with consistently higher R^(2) values and lower mean absolute error(0.002-0.014)compared to the neural network model(0.005-0.024).

Objective:To investigate malaria prevalence and socio-economic conditions in East Timor. Methods:Blood samples were collected from 650 individuals distributed among six districts in East Timor.434 and 216 individuals were sampled by passive and active case detection, respectively(PCD,ACD).Results:The results showed that the plasmodium infection prevalence was 18.9%with predominance of Plasmodium falciparum(***) infections(60.2%). The majority of cases were detected in children between 2-14 years old in both PCD and ACD. Participants residing in Same and Lospalos were at a higher risk of *** percentage of infections associated to the lack of bed net use was twice as high as users(25.2%vs 12.5%). The prevalence of malaria among participants who lived in brick/cement houses(8.3%) was two times less than those who lived in other types of houses(3.7%).There was a significantly lower prevalence of infection detected in individuals living in complete housing constructions with absence of domestic animals in or near the ***:The major risk factors for malaria in East Timor are age,lack of bed net use,incomplete housing constructions and exposure to mosquitoes.
