Utilizing anaerobic metabolisms for the production of biotechnologically relevant products presents potential advantages,such as increased yields and reduced energy ***,lower energy dissipation may indicate that certain reactions are operating closer to their thermodynamic *** stoichiometric analyses and genetic modifications are frequently employed in metabolic engineering,the use of thermodynamic tools to evaluate the feasibility of planned interventions is less *** this study,we propose a novel metabolic engineering strategy to achieve an efficient anaerobic production of poly-(R)-3-hydroxybutyrate(PHB)in the model organism Escherichia *** approach involves re-routing of two-thirds of the glycolytic flux through non-oxidative glycolysis and coupling PHB synthesis with NADH *** complemented our stoichiometric analysis with various thermodynamic approaches to assess the feasibility and the bottlenecks in the proposed engineered *** to our calculations,the main thermodynamic bottleneck are the reactions catalyzed by the acetoacetyl-CoAβ-ketothiolase(EC 2.***.1.9)and the acetoacetyl-CoA reductase(EC 1.***.1.36).Furthermore,we calculated thermodynamically consistent sets of kinetic parameters to determine the enzyme amounts required for sustaining the conversion *** the case of the engineered conversion route,the protein pool necessary to sustain the desired fluxes could account for 20%of the whole cell dry weight.

Developing More Effective Enhanced Biochar Fertilisers for Improvement of Pepper Yield and Quality

Utilization of biochar at high application rates can increase soil C and crop yields, decrease greenhouse gas emissions and reduce nutrient run-off from soils. However, the high application rate of 10 t ha-1may not return a profit to the farmer due to the high cost of biochar. In this study biochar was modified through pre-treating the biomass and post-treating with phosphoric acid, minerals and different chemical fertilisers to study the effects of two new enhanced biochar fertilisers on the yield and quality of green pepper in a field experiment with 5 fertilisation treatments and 3 replications. The two new biochar fertilisers significantly(P < 0.05) increased the yield of green pepper(11.33–11.47 t ha-1), compared with the conventional chemical fertiliser(9.72 t ha-1). The biochar fertiliser treatments improved the vitamin C content of green pepper from 236.99 to 278.28 mg kg-1, and also significantly(P < 0.05) reduced the nitrate content from 132.32 to 101.92 mg kg-1, compared with chemical fertiliser. This study indicated that, compared to the use of conventional chemical fertiliser, all of the biochar fertiliser treatments could significantly improve the yield and quality of green pepper.

本研究将不同浓度改性不溶性大豆纤维(ISF)添加至凝固型酸奶以探究其对酸奶结构、流变特性和水分稳定性的影响。与原味酸奶相比,低浓度(0.25%和0.50%,w/v) ISF诱导形成以蛋白为主导的均匀网络结构,网络中嵌入密集的孔隙或空腔;高浓度(0.75%、1.00%和1.25%,w/v)ISF与蛋白共同形成了由蛋白网络结构、ISF-蛋白聚集体静电复合物及ISF类凝胶结构三部分介导的非均一网络结构。振荡时间扫描显示,随着ISF浓度从0%增加至0.50%,酪蛋白胶束体系溶胶-凝胶转变时间逐渐提前,而随着浓度继续增加至1.25%时转变时间发生推迟。ISF呈浓度依赖性地提高了酸奶凝胶的结构强度和表观粘度,这不仅由酸奶的结构特征决定,而且与ISF自身在水相中的粘弹性和粘度密切相关。ISF通过增强凝胶基质与水分结合力提高了酸奶的水分稳定性,在离心力作用下(1000g,20min),添加ISF凝固型酸奶的持水能力显著高于原味酸奶(P <0.05),最高可达84.5%。本研究证实了ISF作为发酵酸奶功能配料的积极作用,并对富含纤维"清洁标签"酸奶的生产具有一定指导意义。

Aims: A prospective study to assess sexual activity in patients with pelvic organ prolapse (POP) and evaluate changes in sexual functioning after surgery. Secondary outcomes were quality of life and anatomical results. Methods: Patients completed the validated Leiden Questionnaire (LQ) and the short form 36 (SF-36) pre- and two months postoperatively. SPSS 19.0 was used for statistical analysis. Results: 74 patients (average age 61.5 years) were approached, of whom 33.8% were sexually inactive due to other reasons than POP. 23 patients (average age 56.7 years) were included for further analysis. Postoperatively we found a significant improvement for orgasm during intercourse. The remaining domains didn’t change significantly. A trend is noted, indicating that sexual activity stayed unchanged in 82.6% of patients. Dyspareunia improved in 47.8% of patients, lubrication in 34.8% and sexual satisfaction in 40%. Quality of life improved in 73.9%. We found a significant improvement in POP grading. Conclusion: More than a third of the patients presenting with POP are not sexually active due to other reasons than POP. POP surgery appears to improve lubrication, dyspareunia, sexual satisfaction and orgasm during intercourse in a considerable part of patients. Furthermore, there’s no negative influence on sexual activity. POP surgery improves quality of life in the majority of patients.

Adsorption of Hydrazoic Acid on Pristine Graphyne Sheet： A Computational Study

Herein we have investigated the interaction between hydrazoic acid(HN3) and a pristine graphyne system based on density functional theory(DFT) method using generalized gradient approximation. The van der Waals dispersion correction is also considered for predicting the possibility of using the graphyne system for detection of hydrazoic acid. Pristine graphyne has a band gap of 0.453 eV, which decreases to 0.424 eV when HN3 is adsorbed on graphyne. The electrical conductivity of HN3-adsorbed graphyne is greater than that of its pristine counterpart. Charge transfer analysis reveals that the HN3-adsorbed graphyne system behaves as an n-type semiconductor; however, its pristine analogue acts as an intrinsic semiconductor. Pristine graphyne has zero dipole moment; however, its interaction with HN3 increases its dipole moment. The electronic properties of graphyne is significantly influenced by the presence of HN3, leading to the possibility of designing graphyne-based sensors for HN3 detection.

Background: In more than 50% of male subfertility, the aetiology remains unknown. Antisperm antibodies (ASA) might be involved, however the exact role of ASA in unexplained male subfertility is not clear, yet. Objective: The aim of this study was to examine 1) the prevalence of ASA in subfertile men, 2) the possible causes of the presence of ASA, and 3) the influence of ASA on sperm parameters and fertilization including assisted reproductive technologies (ART) and pregnancy outcomes. Study Design, Size, Duration: In this retrospective single center study, all men with semen analyses between January 2003 and December 2017 were included as well as all subfertile couples getting treatment if at least one sperm analysis showed a spermMar test ≥50%. Methods: Collected parameters were: intoxications, medication and professions that could have an adverse effect on fertility, sperm parameters, the type and amount of ART, mode of conception, pregnancy rates and outcomes. Results: 3098 semen analyses were performed. In total, 233 ASA positive men were observed, including 175 subfertile couples with an ASA positive man in the additional analyses.
