Plant latent defense response to microbial non-pathogenic factors antagonizes compatibility

Unlike microbe-associated molecular patterns(MAMPs) that are readily targeted by host immunity,microbial non-pathogenic factors(NPFs) appear negligible as they do not elicit defense. Little is known about whether and how NPFs may be monitored by hosts to control compatibility. Herein, a forward genetic screening isolated an Arabidopsis mutant with a loss of plant-rhizobacteria mutualism, leading to the disclosure of a plant latent defense response(LDR) to NPFs. The activation of LDR in the mutant,named rol1 for regulator of LDR 1, is triggered by several non-pathogenic volatile organic compounds and antagonizes plant compatibility with the beneficial bacterium Bacillus amyloliquefaciens GB03. The activation of LDR in rol1 is mediated through the prokaryotic pathway of chloroplastic lipid *** rol1 root microbiome showed a reduced proportion of the Bacillaceae family. we propose that, parallel to the forefront immunity to MAMPs, LDR to certain NPFs provides a hidden layer of defense for controlling compatibility with commensal or beneficial microbes.

Sinopodophyllum hexandrum,a perennial rhizomatous herb that produces anti-cancer metabolite podophyllotoxin(PPT),is distributed in the Himalayan *** plant growth and PPT accumulation at higher(3300 m) versus lower elevation(2300 m) have been confirmed in our previous *** transcriptomic analysis,234 differentially expressed genes(DEGs) were observed and characterized with 171 up-regulated(UR)and 63 down-regulated(DR) when plants grown at 3,300 m vs 2,300 *** proteomic analysis,65 differentially expressed proteins(DEPs) were identified with 46 UR and19 *** annotation revealed that UR-key genes and proteins including:PLR,2-ODD,PAL,DIR,CYP719 A23,CAD and CYP71 CU1 directly participated in PPT *** classification showed that over 200 DEGs and 50 DEPs associated with growth promotion were classified into 8 categories:cell morphogenesis,bio-signaling,primary metabolism,photosynthesis/energy,transcription/polynucleotide metabolism,translation/protein synthesis,transport as well as stress *** transcriptomic and proteomic analysis provide insight into the mechanism of high elevation enhancing plant growth and PPT accumulation in ***.

为研究彩云木Synadenium grantii叶的化学成分及部分化合物体外抗炎活性,本研究采用硅胶、Sephadex LH-20等多种柱色谱进行分离纯化,通过理化性质和NMR等波谱数据鉴定化合物的结构,并对部分化合物进行抑制脂多糖诱导RAw 264.7细胞释放NO活性的测定。从彩云木叶二氯甲烷-甲醇(1∶1)提取物中共分离得到了10个化合物,分别鉴定为日尔曼醇(1)、大戟醇(2)、正二十八烷醇(3)、β-谷甾醇(4)、豆甾醇(5)、ingol-7,8,12-triacetate-3-phenylacetate(6)、山柰酚-3-O-α-L-吡喃鼠李糖苷(7)、胡萝卜苷(8)、3,4,3′-三甲氧基鞣花酸-4′-O-β-D-吡喃葡萄糖苷(9)、山奈酚-3-O-β-D-吡喃葡萄糖苷(10)。其中3和6~10为首次从该种植物中分离得到,3、6、8和9为首次从聚苞大戟属植物中分离得到,9抑制LPS诱导RAw 264.7细胞释放NO的IC50值为12.0±0.9μM,显示较好的体外抗炎活性。

Very long chain-saturated and-polyunsaturated fatty acids(VLC-SFA and VLC-PUFA, respectively) are a functionally important class of fatty acids containing 28 carbons or more in their acyl chain. They are synthesized by the elongation of very long fatty acids-4(ELOVL4) enzyme, expressed mainly in the brain, retina, skin, testes, and meibomian gland, where these fatty acids are found(Agbaga et al., 2008). Further, these organs exhibit tissuespecific VLC-PUFA and VLC-SFA biosynthesis and incorporation into complex lipids for specific functions. In the brain, skin, and Meibomian glands, the ELOVL4 mainly makes VLC-SFA, which are incorporated into complex sphingolipids. In the retina, the ELOVL4 makes VLC-PUFA that are incorporated into phosphatidylcholine, that are critical for visual function, while in testes and sperm, the VLC-PUFA are incorporated into sphingolipids that are critical for fertility(Yeboah et al., 2021).

AIM:To assess visual outcomes and satisfaction of a non-diffractive extended depth of focus(EDOF)intraocular lens(IOL)in individuals with ocular hypertension(OHT)and well-controlled mild glaucoma undergoing cataract ***:An investigator-initiated,single-center,prospective,interventional,noncomparative study conducted in Montreal,*** study enrolled 31 patients(55 eyes)with OHT or mild glaucoma who received a non-diffractive EDOF IOL(Acrysof IQ Vivity).participants underwent sequential cataract surgery with the Vivity ***-up evaluations occurred at 1d,1,and 3mo postoperatively,assessing uncorrected distance,intermediate,and near visual ***(QUVID:Questionnaire for visual disturbances and IOLSAT:Intraocular lens satisfaction)were administered pre and post-operatively to measure visual disturbances and spectacle independence in various *** parameters included intraocular pressure(IOP),glaucoma medications,spherical equivalence,mean deviation and pattern standard deviation or square root of lost variance on Octopus visual ***:At 1 and 3mo postoperatively,significant improvements were observed in uncorrected distance and intermediate visual *** independence was enhanced for distance and intermediate vision,especially in bright light ***-free intermediate vision was improved even in dim *** disturbances,particularly glare symptoms,were reduced,and there was a notable decrease in IOP and glaucoma medication burden at *** was more hazy vision postoperatively with no impact on visual acuity and visual ***:The non-diffractive EDOF lens improves distance and intermediate spectacle-free visual function in patients with OHT and well-controlled *** findings highlight significant improvements in visual acuity,reduced glare,enhanced spectacle independence,and improved visual performance in different lighting conditions.

The accurate simulation of wave propagation in real media requires properly taking the attenuation into account,which leads to wave dissipation together with its causal companion,wave *** this study,to obtain a weak formulation of heterogenous viscoacoustic wave propagation in an infinite domain,the viscoacoustic medium is first characterized by its frequency-dependent complex bulk compliance instead of the classically used complex bulk ***,a mechanical model using serially connected standard linear solids(SSLS)is built to obtain the rational approximation of the complex bulk compliance whose parameters are calculated via an adapted nonlinear optimization *** the obtained bulk compliance-based constitutive relation,a novel second-order viscoacoustic wave equation in the frequency domain is derived,of which the weak formulation can be physically explained as the virtual work equation and can thus be discretized using a continuous spectral element method in ***,a new method is introduced to address the convolution terms involved in the inverse Fourier transform,whose accurate time integration can then be achieved using an explicit time scheme,which avoids the transient growth that exists in the classical *** resulting full time-space decoupling scheme can handle wave propagation in arbitrary heterogeneous ***,to treat the wave propagation in an infinite domain,a perfectly matched layer in weak formulation is derived for the truncation of the infinite domain via complex coordinate stretching of the virtual work *** only minor modification,the resulting perfectly matched layer can be implemented using the same time scheme as for the wave equation inside the truncated *** accuracy,numerical stability,and versatility of the new proposed scheme are demonstrated with numerical examples.

Axonal regeneration following surgical nerve repair is slow and often incomplete,resulting in poor functional recovery which sometimes contributes to lifelong ***,there are no FDA-approved therapies available to promote nerve *** accelerates axonal regeneration,but systemic side effects presently outweigh its potential benefits for peripheral nerve *** authors describe herein a biodegradable polyurethane-based drug delivery system for the sustained local release of tacrolimus at the nerve repair site,with suitable properties for scalable production and clinical application,aiming to promote nerve regeneration and functional recovery with minimal systemic drug *** is encapsulated into co-axially electrospun polycarbonate-urethane nanofibers to generate an implantable nerve wrap that releases therapeutic doses of bioactive tacrolimus over 31 *** and drug loading are adjustable for applications in small and large caliber nerves,and the wrap degrades within 120 days into biocompatible *** released from the nerve wrap promotes axon elongation in vitro and accelerates nerve regeneration and functional recovery in preclinical nerve repair models while off-target systemic drug exposure is reduced by 80%compared with systemic *** its surgical suitability and preclinical efficacy and safety,this system may provide a readily translatable approach to support axonal regeneration and recovery in patients undergoing nerve surgery.
