Botanic Garden Collections—An Under-Utilised Resource

Botanic gardens and arboreta around the world are repositories of diverse collections of useful plants in their gardens and seed banks. However, the crop and forestry communities often overlook these collections, and so they are an underutilised resource. For example, in analysis of the ex situ conservation status of 6,941 socio-economically important plant taxa using data from forestry and crop collections, but omitted collections in botanic gardens and arboreta. In this paper, we compared the socio-economically important taxa identified by Khoury et al. from GRIN-Global World Economic Plants (WEP) with data on living and seed collections held in botanic garden and arboreta, as recorded in BGCI’s global PlantSearch and ThreatSearch datasets. This analysis produced an assessment of the proportion of these taxa that are found in botanic gardens and arboreta, the number of gardens or arboreta they are found in and what potential they have to contribute to research, conservation and sustainable use. We also compared the species conservation comprehensiveness assessments carried out by Khoury et al. with the threatened status of those species, according to the IUCN Red List and other threat assessment methodologies in order to ascertain whether threatened, useful species are well-conserved in botanic gardens. At least 6017 of the 6941 socio-economically important WEP taxa (86.7%) were currently found in the living and seed collections of botanic gardens and arboreta with 1456 taxa (21%) held in >40 collections. Khoury et al. suggested that 6748 of the WEP taxa are of either medium or high conservation priority. However, our analysis suggested that just 1153 taxa have been assessed as threatened at a national or international level. Fibrogenesis imperfecta ossium is a rare disorder of bone usually characterized by marked osteopenia and associated with variable osteoporosis and osteosclerosis, changing over time. Histological examination shows that newly formed collagen is abnormal, lacking birefringence when examined by polarized light. The case presented demonstrates these features and, in addition, a previously undocumented finding of a persistent marked reduction of the serum C3 and C4. Osteoblasts established in culture from a bone biopsy showed abnormal morphology on electron microscopy and increased proliferation when cultured with benzoylbenzoyl-ATP and 1,25-dihydroxyvitamin D, contrasting with findings in normal osteoblasts in culture. A gene microarray study showed marked upregulation of the messenger RNA(mRNA) for G-protein-coupled receptor 128(GPR 128), an orphan receptor of unknown function and also of osteoprotegerin in the patient's osteoblasts in culture. When normal osteoblasts were cultured with the patient's serum, there was marked upregulation of the mRNA for aquaporin 1. A single pathogenetic factor to account for the features of this disorder has not been defined, but the unique findings described here may facilitate more definitive investigation of the abnormal bone cell function.

By finding some important culture conditions as below, we succeeded in experimentally controlling the whole life history of a dioecious marine green alga, Bryopsis plumosa （hudson） C. Agardh. In this study, we focused on the primary and secondary sex ratios （i.e. at inception and maturity） using these culture techniques. Gametogenesis was induced by culturing haploid gametophytes with Provasoli＇s enriched seawater （PES） medium under a 14：10 h light： dark cycle at 14 ℃. Formed zygotes grew into diploid sporophytes, which were cultured for 3 months with PES medium under a 14：10 h light ： nbsp;dark cycle at 18℃. Then they were transferred into Schreiber medium and cultured under a 10：14 h light： dark cycle at 22℃. Within 1 week, zoosporogenesis was observed. Zoospores were released within a couple of days. Each zoospore soon germinated and grew into a unisexual gametophyte. The primary sex ratio was examined in gametophytes that originated from a single sporophyte. The secondary sex ratio was studied in the field. Both were estimated as 1：1. Synchronized meiotic cell divisions might occur during zoosporogenesis dividing each sex-determining factor evenly among zoospores. Given the equal sex ratio at maturity, there seems to be no environmental factor that differentially affects the survival of male or female gametophytes in nature.

Acne vulgaris is the most common skin condition associated with inflammation of the pilosebaceous unit and affects all ethnic and age groups, independent of sex, nationality, or socioeconomic status. Treatment usually includes oral and/or physical and/or topical interventions—the last can be obtained through commercial preparations in fixed doses or as compounded creams/gels, with personalized qualitative and quantitative composition, to be unique to each patient. In this sense, ready-to-use vehicles play an important role as a timesaving strategy and to ensure maximum results from the treatment. In this paper, we present CleodermTM Clarifying Cream, a ready-to-use, functional semisolid vehicle for acne treatments and topical products for oily skin, to be used by compounding pharmacies. It contains ingredients that can potentiate the effects of the active ingredients added, and has a light and pleasant skin feel. The current body of evidence shows that CleodermTM Clarifying Cream can be an important strategy for compounding personalized acne treatments due to its multiple positive roles on decreasing sebum production, lipid peroxidation, and reactive oxygen species, inhibition of Cutibacterium acnes proliferation, and control of inflammation.

Safety Assessment of a Novel Topical Vehicle for Personalized Treatments in Acne

Acne vulgaris is a common inflammatory disease of the pilosebaceous unit, with a profound impact on the quality of life of patients. In this work, we aimed to study the safety characteristics of CleodermTM, a novel functional semisolid vehicle developed for the compounding of treatments for acne and oily skin. The primary and cumulative irritation and sensitization potential were evaluated in 54 healthy volunteers in a comparative single-blinded study, while the acnegenic and comedogenic potential of the product were assessed in 31 healthy volunteers with combination to oily skin through a non-comparative study. CleodermTM was applied (0.05 g/cm2) in filter paper discs in both studies. For the evaluation of the irritation and sensitization potential, the investigational product, or a control (0.9% NaCl) was applied to the right or left scapular area of the study subjects. No clinical reactions were observed during the induction or challenge phases, suggesting that CleodermTM presents no irritating or allergenic potential. Regarding acnegenicity and comedogenicity, no significant increase in the number of acne lesions linked to the application of the product in four areas of the face was reported, indicating that CleodermTM appears not to display acnegenic and comedogenic potential. Furthermore, over 60% of the study subjects exhibited a decrease in the number of acne lesions after product use. These results show that CleodermTM exhibited no irritating, sensitizing, acnegenic or comedogenic potential, and suggest that it can be considered safe under the study conditions and that the claims dermatologically tested, clinically tested, and non-comedogenic/acnegenic can be supported.
