There has been a growing appreciation for freezing of gait as a disabling symptom that causes a significant burden in Parkinson’s disease. Previous research has highlighted some of the key components that underlie the phenomenon, but these reductionist approaches have yet to lead to a paradigm shift resulting in the development of novel treatment strategies. Addressing this issue will require greater integration of multi-modal data with complex computational modeling, but there are a number of critical aspects that need to be considered before embarking on such an approach. This paper highlights where the field needs to address current gaps and shortcomings including the standardization of definitions and measurement, phenomenology and pathophysiology, as well as considering what available data exist and how future studies should be constructed to achieve the greatest potential to better understand and treat this devastating symptom.

Comparative Proteomics Analysis of Exhaled Breath Condensate in Lung Cancer Patients

The prognosis for patients with non-small cell lung cancer (NSCLC) remains poor in spite of better treatments. This relates mainly to the fact that the majority of patients present with advanced disease. There is a need to identify tools which can improve screening for lung cancer in the at risk patient population. The aim of this study was to compare the breath proteomic profile of NSCLC patients with healthy control subjects to explore the potential of new biomarkers of lung cancer. Comparative proteomic analysis of exhaled breath condensate (EBC) between 14 patients with NSCLC and 13 healthy control subjects were carried out using LTQ FT Ultra mass spectrometry and database searching to determine any unique proteins. In total, 29 unique proteins were identified using multiple protein identification algorithms. A comparison of lung cancer, smoker, and ex-smoker proteomes showed that 18 proteins were shared among the three groups. While one unique protein was found in smokers and lung cancer patients, four proteins were unique to ex-smokers. This data set provides a foundation for evaluation of these proteins from EBC as potential biomarkers for non-invasive lung cancer diagnosis.

Influence of inter-grain cementation stiffness on the effective elastic properties of porous Bentheim sandstone

Effective elastic properties of porous media are known to be significantly influenced by *** this paper,we investigated the influence of another critical factor,the inter-grain cementation stiffness,on the effective elastic properties of a granular porous rock(Bentheim sandstone)using an advanced numerical workflow with realistic rock microstructure and a theoretical ***,the disparity between the experimentally tested elastic properties of Bentheim sandstone and the effective elastic properties predicted by empirical equations was ***,a micro-computed tomography(CT)-scan based approach was implemented with digital imaging software AVIZO to construct the 3d(three-dimensional)realistic microstructure of Bentheim *** microstructural model was imported to a mechanics solver based on the 3d finite element model with inter-grain boundaries modelled by cohesive *** simulations were run to test the effective elastic properties for different shear and normal intergrain cementation ***,a relation between the macroscale Young’s modulus and inter-grain cementation stiffness was derived with a theoretical model which can also account for porosity *** the numerical and theoretical results indicate the influence of the inter-grain cementation stiffness,on the effective elastic properties is significant for porous *** calibrated normal and shear stiffnesses at the inter-grain boundaries are 1.2×10^(5) and 4×10^(4) GPa/m,respectively.

Tumour Necrosis Factor Alpha and Oxidative Stress in the Breath Condensate of Those with Non-Small Cell Lung Cancer

Background and Aims: Lung cancer is a leading cause of cancer mortality worldwide and is associated with the release of tumour necrosis factor-α (TNF-α), subsequent cellular apoptosis and the generation of oxidative stress. Exhaled breath condensate (EBC) analysis is a non-invasive method for sampling biofluids from the lower respiratory tract. This study aimed to evaluate possible biomarkers of lung cancer by measuring the levels of TNF-α and the oxidation of ascorbic acid in EBC. Patients with lung cancer were enrolled into the study prior to treatment, during treatment and post-treatment, and results compared with an age-matched control population. Material and Methods: Patients with Stage II-IV non small cell lung cancer (NSCLC) were recruited prior to and at stages of their treatment. EBC levels of TNF-α, and rate of ascorbic acid oxidation were measured. Results: A total of 19 patients with NSCLC (mean age 71.37 ± 7.77 yrs) and 25 age-matched control subjects were enrolled. Levels of EBC TNF-α were elevated in the EBC of patients with lung cancer compared with control subjects (1.02 ± 0.07 pg/ml vs. 0.51 ± 0.06 pg/ml, p d oxidation was significantly greater in the EBC of patients with lung cancer compared with control subjects (2.20% [0.4 – 11.0] vs. 1.00% [0.1 – 8.5], p = 0.0244). Conclusion: TNF-α and the rate of ascorbic acid oxidation was elevated in the EBC of patients with lung cancer regardless of treatment. Longitudinal studies in a larger population are required to evaluate these markers for the effect of treatment and prognosis.

The nature of signature inversion in the πg_(9/2)vh_(11/2) bands of odd–odd ~(98,102)Rh nuclei is studied. Calculations are performed by using a triaxial rotor plus two-quasiparticle model and are compared with the experimentally observed signature inversions. The calculations reproduce well the observations and suggest that, in these bands, the signature inversion can be interpreted mainly as a competition between the Coriolis and the proton–neutron residual interactions in low K space. The triaxiality applied in the Hamiltonian enlarges the amplitudes of high spin signature zigzags at small triaxial deformation and might push the signature inversion point to higher spin at large triaxial deformation.

We consider a continuous review perishable （s,S） inventory system with a service facility consisting of a waiting hall of finite capacity and a single server. We assume two types of customers, ordinary and negative, arrive according to a Markovian Arrival Process （MAP）. An ordinary customer joins the queue and a negative customer instead of joining the queue removes one ordinary customer from the queue. The removal rule adopted in this paper is RCE （removal of a customer from the end）. The individual customer＇s unit demand is satisfied after a random time of service which is assumed to have a phase-type distribution. The life time of each item and the lead time of the reorders have been assumed to be independent exponential distributions. The joint probability distribution of the number of customers in the system and the inventory level is obtained for the steady state case. Various stationary system performance measures are computed and the total expected cost rate is calculated. The results are illustrated numerically.

Artificial lights improve the catchability of snow crab (Chionoecetes opilio) traps

This study investigated the behaviour and commercial catchability of snow crab(Chionoecetes opilio)in response to different low-powered LEd lights under laboratory and field *** created a novel choice-experiment in a laboratory setting in which we investigated the behaviour of snow crab in response to coloured LEd *** results showed that snow crab movement was dependent on light colour,with animals choosing to move toward blue and white lights,away from purple lights,and no detectable effect for green and red *** then conducted two field experiments to investigate the effect of the same LEd lights on the catch rates of commercial traps during the 2016 snow crab fishery on the east coast of Newfoundland and *** from the first field experiment showed that adding white and purple LEd lights into baited traps significantly improved Catch Per Unit Effort(CPUE)by 77% and 47% *** from the second field experiment showed that unbaited traps equipped with only LEd lights(no bait),could also catch snow crab in comparable amounts to traditional baited traps,with soak time and depth explaining some of the variation in *** together,these experiments suggest that fishing enterprises can improve their catching performance and profitability by adding LEd lights to their traps,or by using LEd lights as a bait replacement.

Inhibition of Rgs10 Expression Prevents Immune Cell Infiltration in Bacteria-induced Inflammatory Lesions and Osteoclast-mediated Bone destruction

Regulator of G-protein Signaling 10 （Rgsl0） plays an important function in osteoclast differentiation. However, the role of Rgsl0 in immune cells and inflammatory responses, which activate osteoclasts in inflam- matory lesions, such as bacteria-induced periodontal disease lesions, remains largely unknown. In this study, we used an adeno-associated virus （AAV-） mediated RNAi （AAV-shRNA-Rgs10） knockdown approach to study Rgsl0＇s function in immune cells and osteoclasts in bacteria-induced inflammatory lesions in a mouse model of periodontal disease. We found that AAV-shRNA-Rgs10 mediated Rgs10 knockdown impaired osteoclastogenesis and osteoclast-mediated bone resorption, in vitro and in vivo. Interestingly, local injection of AAV-shRNA-Rgs10 into the periodontal tissues in the bacteria-induced inflammatory lesion greatly decreased the number of dendritic cells, T-cells and osteoclasts, and protected the periodontal tissues from local inflammatory damage and bone destruction. Importantly, AAV-mediated Rgs10 knockdown also reduced local expression of osteoclast markers and pro-inflammatory cytokines. Our results demonstrate that AAV- shRNA-Rgs10 knockdown in periodontal disease tissues can prevent bone resorption and inflammation simultaneously. Our data indicate that Rgsl0 may regulate dendritic cell proliferation and maturation, as well as the subsequent stimulation of T-cell proliferation and maturation, and osteoclast differentiation and acti- vation. Our study suggests that AAV-shRNA-Rgs10 can be useful as a therapeutic treatment of periodontal disease.

Brominated disinfection by-products(Br-dBPs)can form during the chlorination of drinking water in treatment plants(dWTP).Regulations exist for a small subset of Br-dBPs;however,hundreds of unregulated Br-dBPs have been detected,and limited information exists on their occurrence,concentrations,and seasonal ***,a data-independent precursor isolation and characteristic fragment(dIPIC-Frag)method were optimized to screen chlorinated waters for *** were 553 Br-dBPs detected with m/z values ranging from 170.884 to 497.0278 and chromatographic retention times from 2.4 to 26.2 *** MS2 information,structures for 40 of the 54 most abundant Br-dBPs were *** method was then applied to a year-long study in which raw,clear well,and finished water were analyzed *** 54 most abundant unregulated Br-dBPs were subjected to trend ***-dBPs with higher oxygen-to-carbon(O/C)and bromine-to-carbon(Br/C)ratios increased as water moved from the clear well to the finished stage,which indicated the dynamic formation of *** trends of unregulated Br-dBPs were compared to raw water parameters,such as natural organic matter,temperature,and total bromine,but no correlations were *** was found that total concentrations of bromine(TBr)in finished water(0.04–0.12 mg/L)were consistently and significantly greater than in raw water(0.013–0.038 mg/L,Pduction of bromine during the disinfection *** of TBr in treatment units,rather than raw water,were significantly correlated to 34 of the Br-dBPs atα?*** study provides the first evidence that monthly trends of unregulated Br-dBPs can be associated with the concentration of TBr in treated waters.
