＜正＞Sensory hair cells of vertebrates convert mechanical energy into electrical signals for transmission to the central nervous *** hair cells also are postsynaptic to efferent neurons that project from the central nervous system to inhibit peripheral *** ions are essential cytoplasmic mediators of both afferent and efferent synaptic ***,like other so-called ’short’ receptors（taste cells,photoreceptors） hair cells are physically compact, possessing neither axons nor dendrites to provide spatially-segregated calcium signals for separate synaptic processes. Therefore,afferent excitation and efferent inhibition,often occurring at immediately adjacent synaptic sites,must rely on strong,specialized regulatory mechanisms that confer opposite functional significance to their specific calcium signals （reviewed in fuchs and Parsons,2006）.Small conductance（SK） calcium-sensitive potassium channels are activated by release of ACh from efferent ***-clamp recording from cochlear hair cells can examine the underlying dynamics of synaptic calcium signaling through activation of these synapse-specific potassium *** altering cytoplasmic calcium buffering,or the efficacy of internal calcium stores,we have explored the mechanisms that regulate SK channel activity at efferent synaptic *** such experiments,it is clear that SK channel gating depends on release from internal calcium stores,presumably the ’synaptoplasmic cistern’ lying immediately adjacent to,and coextensive with,the efferent ***,this synaptoplasmic cistern can be loaded by influx through voltage-gated calcium channels, suggesting that it also may be involved in regulating calcium signals at afferent synapses as ***,the specialized synaptic calcium signaling of hair cells relies on energy-dependent,near-membrane calcium stores that help to segregate local calcium signals.

The SARS-CoV-2 Omicron variants are notorious for their transmissibility,but little is known about their subgenomic RNA(sgRNA)*** study applied RNA-seq to delineate the quantitative and qualitative profiles of canonical sgRNA of 118 respiratory samples collected from patients infected with Omicron BA.2 and compared with 338 patients infected with non-variant of concern(non-VOC)-D614G.A unique characteristic profile depicted by the relative abundance of 9 canonical sgRNAs was reproduced by both BA.2 and non-VOCD614G regardless of host gender,age and presence of ***,such profile was lost in samples with low viral load,suggesting a potential application of sgRNA pattern to indicate viral activity of individual patient at a specific time point.A characteristic qualitative profile of canonical sgRNAs was also reproduced by both BA.2 and *** presence of a full set of canonical sgRNAs carried a coherent correlation with crude viral load(AUC¼0.91,95%CI 0.88–0.94),and sgRNA ORF7b was identified to be the best surrogate marker allowing feasible routine application in characterizing the infection status of individual *** potentials in using sgRNA as a target for vaccine and antiviral development are worth pursuing.

Very recently,unusual thrombotic events in combination with severe thrombocytopenia have been reported 1–2 weeks following SARS-CoV-2 vaccination with ChAdOx1 nCov-19(AstraZeneca).This condition,termed VITT(vaccine-induced immune thrombotic thrombocytopenia),has been related to high risk of fatal outcome with both ischaemic and haemorrhagic complications.12 Optimal treatment strategies still need to be elucidated—especially in case of cerebral sinus venous thrombosis(CSVT)with associated brain haemorrhage as the underlying thrombotic *** here report our clinical experience with two young women diagnosed with VITT-associated CSVT,treated with high-dose intravenous immunoglobulins(IVIGs),corticosteroids and argatroban in the hyperacute phase,followed by dabigatran resulting in excellent outcome.

BACKGROUND In adults,bowel intussusception is a rare diagnosis and is mostly due to an organic bowel *** rare cases,this is a complication of a percutaneously placed endoscopic gastro(jejunostomy)*** SUMMARY We describe a case of a 73-year-old patient with a history of myocardial infarction,chronic idiopathic constipation and Parkinson’s *** the admission of his Parkinson’s medication,a percutaneous endoscopic gastrostomy with jejunal extension(PEG-J)was *** patient presented three times at the emergency department of the hospital with intermittent abdominal pain with nausea and *** were no distinctive abnormalities from the physical and laboratory *** abdominal computed tomography scan showed a small bowel *** push endoscopy,a jejunal bezoar at the tip of the PEG-J catheter was found to be the cause of small bowel *** intussusception was resolved after removing the bezoar during push *** Endoscopic treatment of bowel intussusception caused by PEG-J catheter bezoar.

Free-space optical neural network based on thermal atomic nonlinearity

As artificial neural networks(ANNs)continue to make strides in wide-ranging and diverse fields of technology,the search for more efficient hardware implementations beyond conventional electronics is gaining *** particular,optical implementations potentially offer extraordinary gains in terms of speed and reduced energy consumption due to the intrinsic parallelism of free-space *** the same time,a physical nonlinearity—a crucial ingredient of an ANN—is not easy to realize in free-space optics,which restricts the potential of this *** problem is further exacerbated by the need to also perform the nonlinear activation in parallel for each data point to preserve the benefit of linear free-space ***,we present a free-space optical ANN with diffraction-based linear weight summation and nonlinear activation enabled by the saturable absorption of thermal *** demonstrate,via both simulation and experiment,image classification of handwritten digits using only a single layer and observed 6% improvement in classification accuracy due to the optical nonlinearity compared to a linear *** platform preserves the massive parallelism of free-space optics even with physical nonlinearity,and thus opens the way for novel designs and wider deployment of optical ANNs.

Cloning and characterization of a FLORICAULA/LEAFY ortholog, PFL, in polygamous papaya

The homologous genes FLORICAULA (FLO) in Antirrhinum and LEAFY (LFY) in Arabidopsis are known to regu- late the initiation of flowering in these two distantly related plant species. These genes are necessary also for the expression of downstream genes that control floral organ identity. We used Arabidopsis LFY cDNA as a probe to clone and sequence a papaya ortholog of LFY, PFL. It encodes a protein that shares 61% identity with the Arabidopsis LFY gene and 71% identity with the LFY homologs of the two woody tree species: California sycamore (Platanus racemosa) and black cottonwood (Populus trichocarpa). Despite the high sequence similarity within two conserved regions, the N-terminal proline-rich motif in papaya PFL differs from other members in the family. This difference may not affect the gene function of papaya PFL, since an equally divergent but a functional LFY ortholog NEEDLY of Pinus radiata has been reported. Genomic and BAC Southern analyses indicated that there is only one copy of PFL in the papaya genome. In situ hybridization experiments demonstrated that PFL is expressed at a relatively low level in leaf primordia, but it is expressed at a high level in the floral meristem. Quantitative PCR analyses revealed that PFL was expressed in flower buds of all three sex types - male, female, and hermaphrodite with marginal difference between hermaphrodite and unisexual flowers. These data suggest that PFL may play a similar role as LFY in flower development and has limited effect on sex differentiation in papaya.

BACKGROUND Careful selection of hepatocellular carcinoma(HCC)patients prior to chemoembolization treatment is a daily reality,and is even more necessary with new available therapeutic options in *** propose two new models to better stratify patients and maximize clinical benefit:“6 and 12”and“pre/post-TACE-predict”(TACE,transarterial chemoembolization).METHODS We evaluated and compared their performance in predicting overall survival with other systems{Barcelona Clinic Liver Cancer(BCLC),Albumin-Bilirubin(ALBI)and NIACE[Number of tumor(s),Infiltrative HCC,alpha-fetoprotein,Child-Pugh(CP),and performance status]}in two HCC French cohorts of different stages enrolled between 2010 and *** The cohorts included 324 patients classified as BCLC stages A/B(cohort 1)and 137 patients classified as BCLC stages B/C(cohort 2).The majority of the patients had cirrhosis with preserved liver function.“Pre-TACE-predict”and“6 and 12”models identified three distinct categories of patients exhibiting different prognosis in cohort ***,their prognostic value was no better than the BCLC system or NIACE *** function based on CP and ALBI grades significantly impacted patient ***,the“post-TACE-predict”model had a higher predictive value than other *** stratification ability as well as predictive performance of these new models in an intermediate/advanced stage population was less efficient(cohort 2).CONCLUSION The newly proposed“Pre-TACE-predict”and“6 and 12”models offer an interesting stratification into three categories in a recommended TACE population,as they identify poor candidates,those with partial control and durable *** models'contribution was reduced in a population with advanced stage HCCs.
