Osteoarthritis (OA), the most common form of joint disease, is characterized clinically by joint pain, stiffness,and deformity. OA is now considered a whole joint disease;however, the breakdown of the articular cartilage remains themajor hallmark of the disease. Current treatments targeting OA symptoms have a limited impact on impeding orreversing the OA progression. Understanding the molecular and cellular mechanisms underlying OA development isa critical barrier to progress in OA therapy. Recent studies by the current authors’ group and others have revealedthat the nuclear factor of activated T cell 1 (NFAT1), a member of the NFAT family of transcription factors, regulatesthe expression of many anabolic and catabolic genes in articular chondrocytes of adult mice. Mice lacking NFAT1exhibit normal skeletal development but display OA in both appendicular and spinal facet joints as adults. Thisreview mainly focuses on the recent advances in the regulatory role of NFAT1 transcription factor in the activities ofarticular chondrocytes and its implication in the pathogenesis of OA.

Granular titanite from the Roter Kamm crater in Namibia:Product of regional metamorphism,not meteorite impact

Accessory minerals with so-called granular texture have risen in importance as geochronological tools for U-Pb dating of meteorite impact events. Grain-scale recrystallization, typically triggered by a combination of high-strain deformation and post-impact heating, can create a polycrystalline microstructure consisting of neoblasts that expel radiogenic Pb, which are thus ideal for isotopic dating. While granular domains in zircon and monazite from shocked rocks have been demonstrated to preserve impact ages,few U-Pb dating studies have been conducted on granular microstructures in titanite(CaTiSiO;). Here we report the occurrence of granular-textured titanite from ~2020 Ma granite basement rock exposed in the rim of the 4–5 Ma Roter Kamm impact structure in Namibia. Orientation mapping reveals two microstructurally distinct titanite populations: one consisting of strained/deformed grains, and the other consisting of grains that comprise aggregates of strain-free neoblasts. In situ U-Pb geochronology on 37 grains shows that most grains from both titanite populations yield indistinguishable U-Pb dates of ca.1025 Ma, consistent with the observed microstructures forming during the Mesoproterozoic Namaqua Orogeny. Only four grains preserved older age domains, recording ca. 1875 Ma Paleoproterozoic metamorphism. Two significant observations emerge:(1) none of the analyzed titanite grains yield the 2020 Ma igneous crystallization age previously established from zircon in the same sample, and(2) no age-resetting was detected that could be attributed to the 4 to 5 Ma Roter Kamm impact *** the similarity of the neoblastic microstructure to minerals from other sites with an established impact provenance, the granular texture and near-complete Pb-loss in titanite from Roter Kamm granite instead records a Paleo-to Mesoproterozoic polymetamorphic history, rather than Miocene age shockrelated processes. Impact of Long-Term Treatment with OROS Methylphenidate on Pubertal Development in Adolescents with ADHD

The objective of this study was to examine whether stimulants impact pubertal development in adolescent Attention Deficit Hyperactivity Disorder (ADHD), an understudied subject. Pubertal staging data were collected during a 2-year open study of extended release methylphenidate in adolescents (N = 111) with ADHD. Tanner stages were compared to national estimates. The sample was primarily male, Caucasian, and a mean age of 14.8 years at baseline. The baseline Tanner stage for 70% of subjects was consistent with chronological age. For the majority of subjects who reached 12 - 20 months (N = 25) or 24 months (N = 21) endpoints, the Tanner stage at respective endpoints was consistent with age. We found that progression in Tanner stage was not associated with OROS MPH duration or dose (p > 0.10). Long-term treatment with extended release methylphenidate did not appear to adversely impact pubertal development in this sample of adolescents with ADHD.

High-biomass red tides occur frequently in some semi-enclosed bays of Hong Kong where ambient nutrients are not high enough to support such a high phytoplankton biomass. These high-biomass red tides release massive inorganic nutrients into local waters during their collapse. We hypothesized that the massive inorganic nutrients released from the collapse of red tides would fuel growth of other phytoplankton species. This could influence phytoplankton species composition. We tested the hypothesis using a red tide event caused by Mesodinium rubrum(M. rubrum) in a semi-enclosed bay, Port Shelter. The red tide patch had a cell density as high as 5.0×10~5 cells L^(-1), and high chlorophyll a(63.71 μg L^(-1)). Ambient inorganic nutrients(nitrate:NO_3^-, ammonium: NH_4^+, phosphate: PO_4^(3-), silicate: SiO_4^(3-)) were low both in the red tide patch and the non-red-tide patch(clear waters outside the red tide patch). Nutrient addition experiments were conducted by adding all the inorganic nutrients to water samples from the two patches followed by incubation for 9 days. The results showed that the addition of inorganic nutrients did not sustain high M. rubrum cell density, which collapsed after day 1, and did not drive M. rubrum in the non-red-tide patch sample to the same high-cell density in the red tide patch sample. This confirmed that nutrients were not the driving factor for the formation of this red tide event, or for its collapse. The death of M. rubrum after day 1 released high concentrations of NO_3^-,PO_4^(3-), SiO_4^(3-), NH_4^+, and urea. Bacterial abundance and heterotrophic activity increased, reaching the highest on day 3 or 4, and decreased as cell density of M. rubrum declined. The released nutrients stimulated growth of diatoms, such as Chaetoceros affinis var. circinalis, Thalassiothrix frauenfeldii, and Nitzschia sp., particularly with additions of SiO_4^(3-) treatments, and other species. 采用细胞病变法(cytopathologic effect assay,CPE)对中医常用的44种清热解毒中药进行了抗呼吸道合胞体病毒感染的研究。其中27种中药显示很强或中等程度的抗呼吸道病毒感染的活性,其50%抑制浓度(50%inhibition concentration,IC)范围为:6.3～52.1μmg/ml,选择指数(selectivity index,SI)范围为:2.0～32.1。对具有较强活性的苦参(sophora flavescens Ait.)和黄芩(Scutellaria baicalensis Georgi)提取物进行活性成分跟踪分离,得到了具有强抗呼吸道合胞体病毒感染的5个化合物:anagyrine(2),oxymatrine(7),sophoranol(10),wogonin(12)和oroxylin A(13)。

We investigated the seasonal dynamics of flow cytometrically-defined populations of viruses, heterotrophic bacteria, and the picoeukaryotic and prokaryotic phytoplankton at three sites in the temperate oligotrophic inverse estuary of spencer Gulf (South Australia). We consistently identified two sub-populations of viruses, three sub-populations of heterotrophic bacteria, one population of picoeukaryotic phytoplankton and two populations of prokaryotic phytoplankton (cyanobacteria Prochlorococcus and Synechococcus). Both the cytometric community composition and the abundance of viruses, heterotrophic bacteria and both prokaryotic (Synechococcus and Prochlorococcus) and eukaryotic picophytoplankton were consistent with previous observations conducted in South Australian continental shelf waters. Noticeably LDNA bacteria (i.e. inactive or dormant cells) were consistently significantly the most abundant group of heterotrophic bacteria (totaling from 29% to 68% of total bacterial abundance) and were up to 10-fold more abundant than that previously reported in South Australian continental shelf waters, including the nearby Saint Vincent Gulf. These results suggest an overall low activity of the microbial community, and are consistent with previous evidence that LDNA cells may play a greater role in heterotrophic processes than HDNA cells in oligotrophic waters. In an attempt to further assess the qualitative and quantitative nature of the mortality of these organisms, we used a specific dilution assay to assess the relative contribution of viruses and microzooplankton grazers to the mortality rates of heterotrophic bacteria, and picoeukaryotic and prokaryotic phytoplankton. We consistently reported site-specific, population specific and sea-son-specific viral lysis and grazing rates of heterotrophic bacteria and the picoeukaryotic and prokaryotic (cyanobacteria Prochlorococcus and Synechococcus) phytoplankton across sites and seasons.
