A novel aeration strategy in repeated-batch fermentation for efficient ethanol production from sweet sorghum juice

To improve the efficiency of ethanol production in a batch fermentation from sweet sorghum juice under a very high gravity(VHG)condition(~290 g/L of total sugar)by Saccharomyces cerevisiae NP01,repeatedbatch fermentation under an aerated condition(2.5 vvm for the first 4 h during every cycle)was done in a5-L *** average ethanol concentration(P),productivity(Qp)and yield(Yp/s)for five successive cycles were 112.31 g/L,1.55 g/L·h^-1 and 0.44,respectively with 80.97%sugar *** complete sugar consumption,the total sugar of the juice was reduced to a high gravity(HG)level(~240 g/L).The results showed that yeast extract was not necessary for ethanol production,and aeration during every other cycle i.e.,alternating cycles,was sufficient to promote both yeast growth and ethanol *** average P,Qpand Yp/svalues for eight successive cycles with aeration during alternating cycles were97.58 g/L,1.98 g/Láh and 0.41,respectively with 91.21%sugar *** total fatty acids in the yeast cells under the aerated condition were^50%higher than without aeration,irrespective the initial sugar concentration,whereas the ergosterol contents under aeration condition were^29%to 49%higher than those without aeration.

AIM: To prove that the terminal ileum is intussuscepted into the cecum creating the ileocecal junction,contrary to previous valvular concept which has been widely believed. METHODS: This study is based on gross and microscopic examinations of fresh specimens derived from colonic operations (right hemicolectomy or subtotal colectomy). Data compiled from observing and dissecting of specimens of seven patients are used to examine both gross and microscopic appearance of ileocecal junction. RESULTS: Intussusception of the terminal ileum was found in every specimen. However,the length of intussusception was different in each specimen. CONCLUSION: Gross and microscopic appearance studies suggest that the terminal ileum is intussuscepted into the cecum.

Progressive familial intrahepatic cholestasis （PFIC） type 2 is caused by mutations in ABCB11, which encodes bile salt export pump （BSEP）. We report a Thai female infant who presented with progressive cholestatic jaundice since 1 mo of age, with normal serum y-glutamyltransferase. Immunohistochemical staining of the liver did not demonstrate BSEP along the canaliculi, while multidrug resistance protein 3 was expressed adequately. Novel mutations in ABCB11, a four-nucleotide deletion in exon 3, c.90_93delGAAA, and a single-nucleotide insertion in exon 5, c.249_250insT, were identified, with confirmation in her parents. These mutations were predicted to lead to synthesis of truncated forms of BSEP. Immunostaining and mutation analysis thus established the diagnosis of PFIC type

This paper presents a study of fractional order quadrature oscillators based on current-controlled current follower transconductance amplifiers (CCCFTA). The design realisation and performance of the fractional order quadrature oscillators have been presented. The quadrature oscillators are constructed using three fractional capacitors of orders α = 0.5. The fractional capacitor is not available on the market or in the PSPICE program. Fortunately, the fractional capacitor can be realised by using the approximate method for the RC ladder network approximation. The oscillation frequency and oscillation condition can be electronically/orthogonally controlled via input bias currents. Due to high-output impedances, the proposed circuit enables easy cascading in current-mode (CM). The PSPICE simulation results are depicted, and the given results agree well with the anticipated theoretical outcomes.

太阳p模频率随太阳磁活动的变化被称为频率漂移.频率漂移被认为是起源于太阳近表面的一个薄层受到的扰动.利用磁活动指数和太阳模型来计算频率漂移的方法之前已经建立起来了.在这种方法中,频率漂移值依赖于磁活动强度和扰动源位置.磁活动强度正比于磁活动指数.扰动源的位置之前被认为是固定的.研究了扰动源位置的变化.利用观测的太阳p模震动频率和Ca II指数,发现了扰动源的位置可能是随着太阳磁活动而变化的.在太阳活动极大时,扰动源位置较深,而在太阳活动极小时,扰动源位置较浅.

The Prediction Factors of Pre-XDR and XDR-TB among MDR-TB Patients in Northern Thailand

Background: Molecular diagnosis based on the detection of mutations conferring genetic drug resistance is useful for early diagnosis and treatment of Pre-XDR and XDR-TB patients. However, the study of mutation as a marker to predict Pre-XDR and XDR-TB is rare. Methods: Thirty-four Mycobacterium tuberculosis (MTB) isolates from MDR, Pre-XDR and XDR-TB patients in the upper north of Thailand, who had been identified for drug susceptibility using the indirect agar proportion method from 2005-2012, were examined for genetic site mutations of katG, inhA, and ahpC for isoniazid (INH) drug resistance, rpoB for rifampicin (RIF) drug resistance, gyrA for ofloxacin (OFX), and rrs for kanamycin (KAN). Associations between resistant genes and Pre-XDR and XDR-TB in the MDR patients were performed using exact probability tests. Univariable logistic regression was used to quantify the strength of association between the gene mutation with Mycobacterium tuberculosis and the prevalence of Pre-XDR and XDR-TB in the MDR patients. Results: The mutations in the region of the rpoB gene at codon 445 (C445T) in the Pre-XDR or XDR-TB patients were significantly 20.6 times more prevalent among the MDR-TB patients. The inhA gene mutation at codon 114 (T114G) was also significantly 8.1 times more prevalent. Conclusion: The findings can be used to predict the odds of Pre-XDR and XDR-TB in MDR-TB patients, as a guide for prevention and treatments.

草菇锰超氧化物歧化酶基因异源表达条件优化及其耐胁迫功能分析

根据草菇(Volvariella volvacea)的锰超氧化物歧化酶(Mn-SOD)基因序列,构建重组质粒pBAR GPE1/Mn-SOD,转化大肠杆菌(Escherichia coli)Stbl3菌株,采用单因素实验对诱导剂IPTG浓度(0、0.2、0.5、1.0、1.5、2.0 mmol·L^-1)、诱导培养时间(0、2、4、6、8、10 h)及MnCl2浓度(0、0.2、0.5、1.0、1.5、2.0 mmol·L^-1)进行优化,以获得最优表达条件。在最优条件下分别进行耐热性、耐冷性及耐盐性分析,测定重组菌pBAR GPE1/Mn-SOD和对照菌pBAR GPE1的SOD酶活力及生长情况。结果表明:最优表达条件为IPTG浓度1 mmol·L^-1,诱导培养时间6 h,不需要添加MnCl2;在最优条件下,与对照菌相比,发现重组菌的SOD酶活力显著提高,生长情况较好,提高了转化菌株的耐热性、耐冷性和耐盐性。

为探究草菇生产中产量不稳定的内在原因,本研究通过测定废棉培养料不同时期的理化性状、木质纤维素含量及降解酶活性,对草菇(Volvariella volvacea)低产和高产批次基质的降解情况进行了对比分析。结果表明:在培养料的整个发酵阶段,两批次的含水量和碳氮比均逐渐降低,灰分含量、含氮量则逐渐升高,同时高产批次的羧甲基纤维素酶(CMCase)和木聚糖酶活性一直显著高于低产批次。发酵结束时,高产批次的碳氮比较低产批次高4.60,p H值和含水量无明显差异。草菇生长发育阶段,高、低产两批次碳氮比的下降幅度分别为25.71%和12.77%,且前者纤维素相对含量的下降幅度显著大于后者。原基形成时低产批次的漆酶(Laccase)活性是高产批次的2.07倍,采收时低产批次子实体的平均单重较高产批次高6.66 g。本研究对草菇栽培基质的科学优化及工厂化生产进程的推进具有重要参考意义。
