Epac2-deficiency leads to more severe retinal swelling, glial reactivity and oxidative stress in transient middle cerebral artery occlusion induced ischemic retinopathy

Ischemia occurs in diabetic retinopathy with neuronal loss, edema, glial cell reactivity and oxidative stress. Epacs, consisting of Epac 1 and Epac2, are cAMP mediators playing important roles in maintenance of endothelial barrier and neuronal functions to investigate the roles of Epacs in the pathogenesis of ischemic retinopathy, transient middle cerebral artery occlusion （tMCAO） was performed on Epacl-deficient （Epacl-/- ） mice, Epac2-deficient （Epac2-/-） mice, and their wild type counter-parts （Epacl＋/＋ and Epac2＋/＋）. two-hour occlusion and 22-hour reperfusion were conducted to induce ischemia/reperfusion injury to the retina. After tMCAO, the contralateral retinae displayed similar morphology between different genotypes. Neu-ronal loss, retinal edema and increase in immunoreactivity for aquaporin 4 （AQP4）, glial fibrillary acidic protein （GFAP）, peroxiredoxin 6 （Prx6） were observed in ipsilateral retinae. Epac2 / ipsilateral retinae showed more neuronal loss in retinal ganglion cell layer, increased retinal thickness and stronger immunostaining of AQP4, GFAP, and Prx6 than those of Epac2＋/＋. However, Epacl-/- ipsilateral retinae displayed similar pathology as those in Epacl＋/＋ mice. Our observations suggest that Epac2-deficiency led to more severe ischemic retinopathy after retinal ischemia/reperfusion injury.

AIM： to analyze the modulation of gene expression profile associated with inhibition of liver regeneration in hepatitis B X （HBx）-expressing transgenic mice. MEtHODS： Microarray technology was performed on liver tissue obtained from 4 control （LacZ） and 4 transgenic mice （HBx-LacZ）, 48 h after partial hepatectomy. the significance of the normalized logratios was assessed for each gene, using robust t-tests under an empirical Bayes approach. Microarray hybridization data was verified on selected genes by quantitative PCR. RESULtS： the comparison of gene expression patterns showed a consistent modulation of the expression of 26 genes, most of which are implicated in liver regeneration. Up-regulated genes included DNA repair proteins （Rad-52, MSH6） and transmembrane proteins （syndecan 4, tetraspanin）, while down-regulated genes were connected to the regulation of transcription （histone deacetylase, Zfp90, MyoDl） and were involved in the cholesterol metabolic pathway and isoprenoidbiosynthesis （farnesyl diphosphate synthase, Cyp7b1, geranylgeranyl diphosphate synthase, SAA3）. CONCLUSION： Our results provide a novel insight into the biological activities of HBx, implicated in the inhibition of liver regeneration,

tuberculosis(tB)continues to disproportionately affect Inuit populations in Canada with some communities having over 300 times higher rate of active tB than Canadian-born,non-Indigenous *** tuberculosis Elimination Framework has set the goal of reducing active tB incidence by at least 50%by 2025,aiming to eliminate it by *** these goals are achievable with available resources and treatment regimens currently in practice has not been *** developed an agent-based model of tB transmission to evaluate timelines and milestones attainable in Nunavut,Canada by including case findings,contact-tracing and testing,treatment of latent tB infection(LtBI),and the government investment on housing infrastructure to reduce the average house-hold *** model was calibrated to ten years of tB incidence data,and simulated for 20 years to project program *** found that,under a range of plausible scenarios with tracing and testing of 25%e100%of frequent contacts of detected active cases,the goal of 50%reduction in annual incidence by 2025 is not *** active tB cases are identified rapidly within one week of becoming symptomatic,then the annual incidence would reduce below 100 per 100,000 population,with 50%reduction being met between 2025 and *** tB from Inuit populations would require high rates of contacttracing and would extend beyond *** findings indicate that time-to-identification of active tB is a critical factor determining program effectiveness,suggesting that investment in resources for rapid case detection is fundamental to controlling tB.

Modelling Dam Break Evolution over a Wet Bed with Smoothed Particle Hydrodynamics: A Parameter Study

When investigating water flow in spillways and energy dissipation, it is important to know the behavior of the free surfaces. to capture the real dynamic behavior of the free surfaces is therefore crucial when performing simulations. today, there is a lack in the possibility to model such phenomenon with traditional methods. Hence, this work focuses on a parameter study for one alternative simulation tool available, namely the meshfree, Lagrangian particle method Smoothed Particle Hydrodynamics (SPH). the parameter study includes the choice of equation-of-state (EOS), the artificial viscosity constants, using a dynamic versus a static smoothing length, SPH particle spatial resolution and the finite element method (FEM) mesh scaling of the boundaries. the two dimensional SPHERIC Benchmark test case of dam break evolution over a wet bed was used for comparison and validation. the numerical results generally showed a tendency of the wave front to be ahead of the experimental results, i.e. to have a greater wave front velocity. the choice of EOS, FEM mesh scaling as well as using a dynamic or a static smoothing length showed little or no significant effect on the outcome, though the SPH particle resolution and the choice of artificial viscosity constants had a major impact. A high particle resolution increased the number of flow features resolved for both choices of artificial viscosity constants, but at the expense of increasing the mean error. Furthermore, setting the artificial viscosity constants equal to unity for the coarser cases resulted in a highly viscous and unphysical solution, and thus the relation between the artificial viscosity constants and the particle resolution and its impact on the behavior of the fluid needed to be further investigated.

Adsorption Kinetics and thermodynamics Study of Butylparaben on Activated Carbon Coconut Based

In this work, low cost coconut biochar based activated carbon (CBAC) was used for adsorption of Butylparaben (BPB) from aqueous medium. the prepared CBAC was characterized using BEt, Boehm analysis and the adsorption equilibrium, kinetics and thermodynamics studies of BPB adsorption were carried out. During batch adsorption runs, the effects of factors, such as contact time (0 - 300 min), CBAC dose (200 - 800 mg), pH (3 - 11) and solution temperatures (303 - 348 K) were investigated on BPB removal. Experimental results reveal that the BPB removal efficiency on CBAC is higher than 97% under acidic and neutral conditions. Equilibrium data were fitted by Langmuir, Freundlich and temkin isotherm models with correlation coefficient more than 0.9. the pseudo-second order kinetic model was observed to fit well the adsorption data. thermodynamic analysis shows positive values of standard Gibb’s free energy, suggesting the non-spontaneity of the process. the changes in enthalpy (0.2 ***-1) and entropy (19 ***-1) were found to be endothermic with an increase of randomness. the high adsorption efficiency of the synthesized coconut biochar materials with low cost indicates that it may be a promising adsorbent for removing organic compounds.
