Modeling of genetic gain for single traits from marker-assisted seedling selection in clonally propagated crops

Seedling selection identifies superior seedlings as candidate cultivars based on predicted genetic potential for traits of ***,genetic potential is determined by phenotypic *** the availability of DNA tests for some agronomically important traits,breeders have the opportunity to include DNA information in their seedling selection operations—known as marker-assisted seedling selection.A major challenge in deploying marker-assisted seedling selection in clonally propagated crops is a lack of knowledge in genetic gain achievable from alternative *** models based on additive effects considering seed-propagated crops are not directly relevant for seedling selection of clonally propagated crops,as clonal propagation captures all genetic effects,not just *** study modeled genetic gain from traditional and various marker-based seedling selection strategies on a single trait basis through analytical derivation and stochastic simulation,based on a generalized seedling selection scheme of clonally propagated *** trait-test scenarios with a range of broad-sense heritability and proportion of genotypic variance explained by DNA markers were simulated for two populations with different segregation *** derived and simulated results indicated that marker-based strategies tended to achieve higher genetic gain than phenotypic seedling selection for a trait where the proportion of genotypic variance explained by marker information was greater than the broad-sense *** from this study provides guidance in optimizing genetic gain from seedling selection for single traits where DNA tests providing marker information are available.

Pelvic region bone density,soft tissue mass,and injury frequency in female professional ballet dancers and soccer athletes

We recently observed a high prevalence of low pelvic bone mineral density(BMD)in female professional ballet *** this population is susceptible to musculoskeletal overuse injuries,we aimed to determine which regions of the pelvis may be at greatest risk compared to general population females(GENPOP)as well as professional female soccer players(SOCCER,a comparison to other elite athletes regularly subjected to high degrees of loading).Three groups of age-matched females[(GENPOP;n=38,27±1yrs),(BALLET;single com-pany,n=36,26±3yrs),(SOCCER;single NWSL■club,n=34,25±1yrs)]consented to have their BMD and body composition assessed(DEXA,GE■).In addition to soft tissue and total and regional BMD analyses,a segmental analysis of the pelvis was performed to determine site-specific BMD for the iliac fossa,iliac fossa/iliac crest/ilium combined,pubic bone,ischium,and sacrum.A mixed-model ANOVA followed by a Tukey's post-hoc test was used to compare the groups(Type-I error;α=0.05).The BALLET group had lower pelvic BMD for all measures(Avg.%Diff.=15%-27%,p<0.001)compared to the SOCCER group and for the ischium(Avg.%Diff.=8%;p=0.007)and sacrum(Avg.%Diff.=7%;p=0.028)compared to the GENPOP *** BALLET group had lower lean mass for all measures compared to the other groups(Avg.%Diff.=12%-18%;p<0.01).Professional ballet performers exhibit reduced pelvic region soft tissue and site-specific BMD not previously detected using standard DEXA *** findings highlight which pelvic regions may benefit from preventative strength training and/or nutritional interventions.

An Extended Genetic Linkage Map for Watermelon Based on a Testcross and a BC<sub>2</sub>F<sub>2</sub>Population

There is limited genetic mapping data useful for breeding programs of watermelon. Introgression lines should be a use-ful tool for genetic studies and genetic enhancement of watermelon cultivars. In this study, we used an advanced re-combinant population (BC2F2) to identify and map chromosomal segments of the wild watermelon Citrullus lanatus var. citroides that were incorporated in the genome of the watermelon cultivar Crimson Sweet (Citrullus lanatus var. lana-tus). An advanced recombinant population (BC2F2) was constructed using a United States Plant Introduction (PI) 494817 (C. lanatus var. citroides) (known to have moderate resistance to bacterial fruit blotch) as a donor parent, and the elite watermelon cultivar Crimson Sweet (C. lanatus var. lanatus) as the recurrent parent. The genetic linkage map consists of 272 markers, including 89 sequence-related amplified polymorphism (SRAP), 72 targeted region amplifica-tion polymorphism (TRAP), and 111 high frequency oligonucleotide-targeting active gene (HFO-TAG) markers. The 272 markers were assembled into 51 linkage groups, covering a total genetic distance of 2162 cM, with an average genetic distance of 7.9 cM between markers. Also, we expended the genetic linkage map for watermelon derived from a testcross population {Griffin 14113 [C. lanatus var. citroide (L.H. Bailey) Mansf.] x watermelon cultivar New Hamp-shire Midget (C. lanatus var. lanatus)} x PI 386015 [C. colocynthis (L.) Schrad.]. The genetic linkage map based on the test cross population consists of 558 markers that cover a genetic distance of 2760.8 cM. This linkage map consists of 41 linkage group, including 10 large linkage groups (ranging from102-240 cM), nine intermediate size linkage groups (ranging from 62-93 cM), and 22 small linkage groups (ranging from 2-56 cM). Fluoxetine induces cytotoxic endoplasmic reticulum stress and autophagy in triple negative breast cancer

AIM: To investigate the mechanism of action of lipophilic antidepressant fluoxetine(FLX) in representative molecular subtypes of breast ***: The anti-proliferative effects and mechanistic action of FLX in triple-negative(SUM149PT) and luminal(T47D and Au565) cancer cells and nontransformed MCF10 A were investigated. Reverse phase protein microarray(RPPM) was performed with and without 10 μmol/L FLX for 24 and 48 h to determine which proteins are significantly changed. Viability and cell cycle analysis were also performed to determine drug effects on cell growth. Western blotting was used to confirm the change in protein expression examined by RPPM or pursue other signaling proteins. RESULTS: The FLX-induced cell growth inhibition in all cell lines was concentration- and time-dependent but less pronounced in early passage MCF10 A. In comparison to the other lines,cell growth reduction in SUM149 PT coincided with significant induction of endoplasmic reticulum(ER) stress and autophagy after 24 and 48 h of 10 μmol/L FLX,resulting in decreased translation of proteins along the receptor tyrosine kinase/Akt/mammalian target of rapamycin pathways. The increase in autophagy marker,cleaved microtubule-associated protein 1 light chain 3,in SUM149 PT after 24 h of FLX was likely due to increased metabolic demands of rapidly dividing cells and ER stress. Consequently,the unfolded protein response mediated by double-stranded RNA-dependent protein kinase-like ER kinase resulted in inhibition of protein synthesis,growth arrest at the G1 phase,autophagy,and caspase-7-mediated cell ***: Our study suggests a new role for FLX as an inducer of ER stress and autophagy,resulting in death of aggressive triple negative breast cancer SUM149 PT.

简介头颈部肿瘤病人需要营养健康的饮食,但由于恶性肿瘤和放射疗法对身体的影响,其所需的营养摄入也要相应改变。目前,通过健康行为干预以改善这类病人的营养状况是否有效,尚未研究。试验建立在数据基础上,评估由营养师提供健康行为干预以改善头顶部肿瘤患者营养不良状况的有效性,即膳食疗法。方法与分析采用阶梯式整群随机对照试验,对参加研究医院的肿瘤科大夫进行随机数字编号,并进行为期2天的膳食疗法培训,研究人员在培训期间进行督导,并提供2个月的强化训练。膳食疗法是基于行为改变理论的一种方法,包括:动机性访谈、认知行为治疗和临床实践。膳食疗法的作用是激发病人的食欲,帮助病人克服疼痛、黏膜炎、恶心、口干、味觉改变和食欲减退等障碍,改变其不良健康行为,由营养师对病人进行膳食指导。在澳大利亚选择5个地区,从每个地区的医院放射科选择18岁以上且患有头颈部肿瘤,准备进行放射治疗的患者,共400人参加研究。分别选择放疗的第1周和最后1周,放疗后的第4周和第12周四个时段进行评估。本次研究的主要成果是对头颈部肿瘤病人放疗后营养现状的评估。
