Tendinopathy,which is characterized by the ectopic ossification of tendon,is a common disease occurring in certain population,such as athletes that suffer from repetitive tendon ***,the molecular mechanism underlying the pathogenesis of tendinopathy caused by the overuse of tendon is still ***,we found that the mechanosensitive miRNA,miR-337-3p,had lower ex.ression under uniax.al cyclical mechanical loading in tendon-derived stem cells(TDSCs)and negatively controlled chondro-osteogenic differentiation of ***,downregulation of miR-337-3p ex.ression was also observed in both rat and human calcified tendons,and overex.ressing miR-337-3p in patellar tendons of rat tendinopathy model displayed a robust therapeutic ***,we found that the proinflammatory cytokine interleukin-1^was the upstream factor of miR-337-3p that bridges the mechanical loading with its ***,the target genes of miR-337-3p,NADPH ox.dase 4,and insulin receptor substrate 1,activated chondro-osteogenic differentiation of TDSCs through JNK and ERK signaling,***,these findings not only provide novel insight into the molecular mechanisms underlying ectopic ossification in tendinopathy but also highlight the significance of miR-337-3p as a putative therapeutic target for clinic treatment of tendinopathy.

Discharge plasmas, recognized as unique platforms for investigating the origins of chemical life, have garnered ex.ensive interest for their potential to simulate prebiotic conditions. This paper embarks on a comprehensive overview of recent advancements in the plasma-enabled synthesis of life’s building blocks, charting the complex.environmental parameters believed to have surrounded life’s inception. This discussion elaborates on the fundamental mechanisms of discharge plasmas and their likely role in fostering conditions necessary for the origin of life on early Earth. We consider a variety of chemical reactions facilitated by plasma, specifically the synthesis of vital organic molecules - amino acids, nucleobases, sugars, and lipids. Further, we delve into the impact of plasmas on prebiotic chemical evolution. We ex.ect this review to open new horizons for future investigations in plasma-related prebiotic chemistry that could offer valuable insights for unraveling the mysteries of life's origin.

Cellulose macrofibers (MFs) are gaining increasing interest as natural and biodegradable alternatives to fossil-derived polymers for both structural and functional applications. However, simultaneously achieving their ex.eptional mechanical performance and desired functionality is challenging and requires complex.processing. Here, we reported a one-step approach using a tension-assisted twisting (TAT) technique for MF fabrication from bacterial cellulose (BC). The TAT stretches and aligns BC nanofibers pre-arranged in hydrogel tubes to form MFs with compactly assembled structures and enhanced hydrogen bonding among neighboring nanofibers. The as-prepared BC MFs ex.ibited a very high tensile strength of 1 057 MPa and ex.eptional lifting capacity (over 340 000 when normalized by their own weight). Moreover, due to the volume ex.ansion of BC nanofibers upon water ex.osure, BC MFs quickly harvested energy from environmental moisture to untwist the bundled networks, thus generating a torsional spinning with a peak rotation speed of 884 r/(min·m). The demonstrated rapid and intense actuation response makes the MFs ideal candidates for diverse humidity-response-based applications beyond advanced actuators, remote rain indicators, intelligent switches, and smart curtains.

In this work,melting of a high-temperature inorganic phase change material(PCM)eutectic(with a melting point of 569℃)within a vertical cylindrical tank has been ex.erimentally *** promote the heat transfer rate,a periodic structure that is constructed by a commercial SS-304 mesh screen has been considered and immersed into the PCM *** characteristics of the PCM-periodic structure tank under different initial temperatures(450,490 and 546℃)and wall temperatures(620,640,660,680 and 700℃),are then investigated and *** presented ex.erimental data can facilitate practical engineers to find the best operating condition of similar PCM tanks;meanwhile,it can also be employed for the investigation of thermal response of transient heat conduction before melting starts.

Herein,we report a mild and practical protocol for the copper-catalyzed chlorofluoromethylthiolation of(hetero)aryl boronic acids with the novel reagent *** resulting products are amenable to halogen-ex.hange^(18)F-fluorination with cyclotronproduced[^(18)F]fluoride affording[^(18)F]*** process highlights the combined value of reagent development and(hetero)aryl boron precursors for radiochemistry by adding the[^(18)F]SCF2H group to the list of^(18)F-motifs within reach for positron emission tomography studies.

A genome-wide association study(GWAS)identifies trait-associated loci,but identifying the causal genes can be a bottleneck,due in part to slow decay of linkage disequilibrium(LD).A transcriptome-wide association study(TWAS)addresses this issue by identifying gene ex.ression-phenotype associations or integrating gene ex.ression quantitative trait loci with GWAS ***,we used self-pollinated soybean(Glycine max.L.]Merr.)as a model to evaluate the application of TWAS to the genetic dissection of traits in plant species with slow LD *** generated RNA sequencing data for a soybean diversity panel and identified the genetic ex.ression regulation of 29286 soybean *** TWAS solutions were less affected by LD and were robust to the source of ex.ression,identifing known genes related to traits from different tissues and developmental *** novel pod-color gene L2 was identified via TWAS and functionally validated by genome *** introducing a new ex.n proportion feature,we significantly improved the detection of ex.ression variations that resulted from structural variations and alternative *** a result,the genes identified through our TWAS approach ex.ibited a diverse range of causal variations,including SNPs,insertions or deletions,gene fusion,copy number variations,and alternative *** this approach,we identified genes associated with flowering time,including both previously known genes and novel genes that had not previously been linked to this trait,providing insights complementary to those from *** summary,this study supports the application of TWAS for candidate gene identification in species with low rates of LD decay.

A straight, non-sporulating, Gram-variable bacillus (HKU24T) was recovered from the blood culture of a patient with metastatic breast carcinoma. After repeated subculturing in BACTEC Plus Anaerobic/F blood culture broth, HKU24T grew on brucella agar as non-hemolytic, pinpoint colonies after 96 h of incubation at 37 °C in an anaerobic environment and aerobic environment with 5% CO2. Growth was enhanced with a streak of Staphylococcus aureus. HKU24T was non-motile and catalase-negative, but positive for alkaline phosphatase, β-glucosidase, and α-glucosidase. It hydrolyzed phenylphosphonate and reduced resazurin. 16S rRNA, groEL, gyrB, recA, and rpoB sequencing showed that HKU24T occupies a distinct phylogenetic position among the Leptotrichia species, being most closely related to Leptotrichia trevisanii. Using HKU24T groEL, gyrB, recA, and rpoB gene-specific primers, fragments of these genes were amplified from one of 20 oral specimens. Based on phenotypic and genotypic characteristics, we propose a new species, Leptotrichia hongkongensis sp. nov., to describe this bacterium.
