Tumour necrosis factor-α is a cytokine released during myocardial infarction. According to the literature, the effect of TNFα on myocardial infarction is controversial, especially when administered before the ischemic period. The deleterious effects of TNFα seem to be related to the triggering of apoptosis. This study has been designed to determine if different doses of TNFα, administered before the ischemic period, have the same effect on infarct size and on activation of caspase-3 and-8, two enzymes involved in apoptosis. Four groups, using a porcine model of myocardial infarction, have been used: placebo and TNFα (0.1 μg/kg;1 μg/kg and 3 μg/kg). All administered 15 minutes before a 50 minutes occlusion of the left anterior descending artery. Myocardial infarct size has been determined at 3 hours of reperfusion. In a subgroup of animals, reperfusion period has been limited to 15 min to determine the activity of caspase-3 and-8 by spectrofluorometry. Results indicated that infarct size is significantly smaller in groups 0.1 μg/kg and 1 μg/ kg as compared to the placebo group. In contrast, the 3 μg/kg group presented an infarct size similar to the placebo group. Activity of caspase-3 and-8 is reduced in the ischemic region in groups 0.1 and 1 μg/ kg as compared to the placebo group whereas activity in the 3 μg/kg group was similar to the placebo. The results obtained indicated that a low dose of TNFα administered before the ischemic period reduces infarct size, whereas the cardioprotection is lost with the high dose.

Effect of undulators on the stored electron beam of Indus-2

Indus-2 is an Indian synchrotron light source, operating at 2.5 Ge V and generating synchrotron radiation from its bending magnets. In order to provide more intense synchrotron radiation to the synchrotron users, there is a plan to install five insertion devices in the Indus-2 storage ring. In the first phase of installation of insertion devices, there is a proposal to install two out- vacuum pure permanent magnet linearly polarized undulators in long straight sections of the Indus-2 storage ring. The presence of the insertion devices in the ring has inevitable effects on beam parameters like betatron tune, betatron amplitude function, closed orbit, emittance, energy spread and dynamic aperture etc. In this paper, the effect of two undulators on the above mentioned parameters of the Indus-2stored electron beam at 2.5 Ge V is presented. Moreover a correction scheme for the restoration of the betatron tune and amplitude function is also presented.

背景:氟伐他汀(来适可)干预预防研究(LIPS)发现,稳定或不稳定型心绞痛或无症状性心肌缺血患者在成功进行首次经皮冠脉介入术(PCI)后,每天使用80mg氟伐他汀可使主要不良心脏事件(MACE)的发生率降低22%,与安慰剂相比有显著差异。然而该治疗方案的成本-效果比不明。目的:评估在成功进行首次PCI后使用氟伐他汀的成本-效果比。方法:以Markov模型评估稳定或不稳定型心绞痛或无症状性心肌缺血患者在成功完成首次PCI后可采用的两种治疗方案的预期结果和成本:①饮食/生活方式指导及立即使用氟伐他汀(80mg/d);②仅进行饮食/生活方式指导,在发生非致死性MACE后开始使用氟伐他汀(80mg/d)。该模型的评估基于LIPS和其他相关报道的数据。成本-效果比以预计医疗成本差值与生命年(life-year,LY)或质量调整生命年(QALY)预计差值之间的比值计算。结果:成功进行首次PCI后使用氟伐他汀可使平均生存期增加0.78年(QALY0.68年)。该治疗方案的成本-效果比为每LY节省13505美元(每QALY节省15454美元);这一比值在糖尿病和多血管病变患者中相对较低。结论:与其他常用的医疗干预手段相比,PCI后使用氟伐他汀具有较高的成本-效果比。

Analysis and correction of linear optics errors,and operational improvements in the Indus-2 storage ring

Estimation and correction of the optics errors in an operational storage ring is always vital to achieve the design performance. To achieve this task, the most suitable and widely used technique, called linear optics from closed orbit（LOCO） is used in almost all storage ring based synchrotron radiation sources. In this technique, based on the response matrix fit, errors in the quadrupole strengths, beam position monitor（BPM） gains, orbit corrector calibration factors etc. can be obtained. For correction of the optics, suitable changes in the quadrupole strengths can be applied through the driving currents of the quadrupole power supplies to achieve the desired optics. The LOCO code has been used at the Indus-2 storage ring for the first time. The estimation of linear beam optics errors and their correction to minimize the distortion of linear beam dynamical parameters by using the installed number of quadrupole power supplies is discussed. After the optics correction, the performance of the storage ring is improved in terms of better beam injection/accumulation, reduced beam loss during energy ramping, and improvement in beam lifetime. It is also useful in controlling the leakage in the orbit bump required for machine studies or for commissioning of new beamlines.

Plantlets Regeneration via Somatic Embryogenesis from the Nucellus Tissues of Kinnow Mandarin (Citrus reticulata L.)

Studies were initiated to explore the role of nucellus tissues and growth regulators in plantlets regeneration via somatic embryogenesis of Kinnow mandarin [Citrus reticulata L. (Blanco)]. Nucellus tissues were cultured on MS medium supplemented with different concentrations and combinations of auxins, cytokinins and malt extract for primary callus induction. The best response for primary callus induction (90%) was obtained when MS medium was supplemented with 5 mg/l 2,4-d and 500 mg/l malt extract. Best results for embryogenic callus induction (80%) were obtained in C8 medium. The induction of somatic embryos was highest when MS medium was supplemented with 1 mg/l BAP and maturation of somatic embryos occurred when MS medium was supplemented with 5 mg/l 2,4-d and 1 mg/l BAP. Maximum plantlets were regenerated (92%) from the somatic embryos on half strength MS medium with no hormones. The plantlets were successfully acclimatized in different potting mixtures and highest survival rate (100%) was achieved in potting mixture containing sand and peat moss (2:1).

Optimal placement of magnets in Indus-2 storage ring

In Indus-2, by optimizing the position of the magnetic elements, using the simulated annealing algorithm, at different locations in the ring with their field errors, the effects on beam parameters have been minimized. Closed orbit distortion and beta beat are considerably reduced by optimizing the dipole and quadrupole magnets positions in the ring. For the Indus-2 storage ring, sextupole optimization gives insignificant improvement in dynamic aperture with chromaticity-correcting sextupoles. The magnets have been placed in the ring with the optimized sequence and storage of the beam has been achieved at injection energy without energizing any corrector magnets. Magnet sorting has led to the easy beam current accumulation and the measurement of parameters such as closed orbit distortion, beta function, dispersion, dynamic aperture etc.

目的:评估梅毒性葡萄膜炎患者的临床表现和视力。方法:梅毒性葡萄膜炎患者3例在USM医院接受治疗。结果:患者3例被诊断为继发性梅毒性葡萄膜炎,这3例患者患病之前均不知道患有梅毒,但他们有明确的乱交史。并且每个月都伴有逐步的视力下降。其中两人伴有发热、眼痛、眼前悬浮物。视力从6/12到手动。所有患者均出现前葡萄膜炎,玻璃体炎和视神经炎。第一例患者出现了多灶性脉络膜视网膜炎,并伴有渗出性视网膜脱离。第二例患者出现渗出性视网膜脱离,而第三例患者仅出现了脉络膜视网膜炎。所有患者每周注射苄青霉素2.4MU,共4wk,其中2例患者口服多西霉素200mg2次/d,共3mo。治疗效果良好,其中2例患者有显著的视力上升,分别从6/120到6/21和6/12到6/6。其中较严重的1例患者出现逆转录酶为阳性。结论:眼梅毒作为非肉芽肿性的炎症与渗出性视网膜脱离有关。治疗后虽然视力恢复比较缓慢,但普遍有良好的效果。

Comparison of semilunar coronally repositioned flap with gingival massaging using an Ayurvedic product(irimedadi taila) in the treatment of class-Ⅰ gingival recession: A clinical study

AIM: To study the comparison in terms of root coverage the effect of gingival massaging using an ayurvedic product and semilunar coronally repositioned flap(SCRF) to assess the treatment outcomes in the management of Miller's class Ⅰ gingival recessions over a-6mo period. METHOdS: The present study comprised of total of 90 sites of Miller's class-Ⅰ gingival recessions in the maxillary anteriors, the sites were divided into three groups each comprising 30 sites, Group Ⅰ-were treated by massaging using a Placebo(Ghee) Group Ⅱ-were treated by massaging using an ayurvedic product(irimedadi taila). Group Ⅲ-were treated by SCRF. Clinical parameters assessed included recession height, recession width, probing pocket depth, width of attached gingiva, clinical attachment level and thickness of keratinized tissue. Clinical recordings were performed at baseline and 6 mo later. The results were analyzed to determine improvements in the clinical parameters. The comparison was done using Wilcoxon signed rank test. The overall differences in the clinical improvements between the three groups was done using Kruskal-Wallis test. The probability value(P-value) of less than 0.01 was considered as statistically ***: Non-surgical periodontal therapy and gingival massaging improves facial gingival recessions and prevents further progression of mucogingival defects. Root coverage was achieved in both the experimental groups. The SCRF group proved to be superior in terms of all the clinical ***: Root coverage is significantly better with semilunar coronally repositioned flap compared with the gingival massaging technique in the treatment of shallow maxillary Miller class Ⅰ gingival recession defects.

In Vitro Study of Callogenesis and Regeneration Potential of Elite Wheat (Triticum aestivum L.) Cultivars

The potential for biotechnological applications in crop improvement programs requires identifying genotypes that allow cell/tissue culture with predictable plant regeneration. In the past, many genotypes of wheat (Triticum aestivum L.) have been examined for potential use in tissue culture studies. The present research work has also been designed to study in vitro callogenesis expression and regeneration potential of wheat cultivars under controlled laboratory conditions. Seeds of four elite commercial high yielding cultivars of wheat namely: NARC-2011, AAS-2011, PAK-2013 and GAL-2013, were collected from the Crop Science Institute National Agricultural Research Center (CSI-NARC) Islamabad, as the source of plant material for in vitro studies. The seeds were surface sterilized in 10% sodium hypochlorite solutions for 10 minutes with continuous shaking under laminar air flow hood. After that seeds were placed on MS (Murashige & Skoog, 1962) based callus induction and regeneration medium with various concentrations of 2, 4-d and BAP in separate test tubes. Maximum callus induction frequency of 90% for Pak-13 and AAS-11, followed by 87% and 83% for Gla-13 and NARC-11, respectively, was recorded at 4 mg/l and 6 mg/l of 2, 4-d. Similarly, maximum regeneration of 90% for AAS-11 and Pak-13, followed by 80% and 87% for NARC-11 and Gla-13 respectively, was recorded on MS basal medium containing 1.5 mg/l of BAP. An increasing trend in regeneration from 0.5 to 1.5 mg/l of BAP was observed but it gradually decreased with increasing concentration of BAP from 1.5 mg/l for all wheat cultivars. The callus formed under light was golden brown, dry nodule and smooth compact and less embryogenic while under dark conditions, it was white to yellowish white, dry nodule and compact and more embryogenic. Best results for callus induction and regeneration were obtained at temperature (24°C ± 1°C) for all wheat cultivars.

从1974年以来,海洋洄游性的波罗的海鲑的种群受到一种被称为M7综合症引起的早期生活阶段死亡的影响。M74综合症已经证明和硫胺素(Vitamin B_1)的缺乏有关,维生素B_1的缺乏导致和坏死的脑细胞有关的神经紊乱。用维生素B_1处理能抑制M74综合症的发展。在这项研究中,通过微量注射的方法,将维生素B_1的对抗剂羟基硫胺素和商品化的多氯联苯的混合物ClophenA50注射到海洋洄游性波罗海鲑鱼的睛点期胚胎卵的卵黄囊内。其目的是研究由实验引起的维生素B_1缺乏效应和通过7-乙氧基异吩噁唑酮-O-脱乙基酶(EROd)检测的方法研究它如何影响生物转化系统CYP1A。鲑鱼胚胎孵化以后,我们试图改变每个暴露组一半鲑鱼的维生素B_1缺乏状况,方法是将各组孵化出来的50%鲑鱼放入维生素B_1的溶液中,并且研究其对鲑鱼存活和EROd活性的影响。注射过羟基硫胺素的各组胚胎孵出的具卵黄囊鱼苗无论是否注射过Clophen A50,它们都明显表现出了协调性的丧失,不活泼,眼睛突出,变白的肝脏,接着全部死亡(100%)。基于这些现象和死亡的时间(孵化以后的124～193度日(d℃》,羟基硫胺素的作用可以和M74综合症对鲑鱼的作用相比拟,但是也观察到了不相似的地方。注射过羟基硫胺素的各组鲑鱼苗通过维生素B_1处理后延缓了死亡,而且死亡率下降到64.8%至91.8%。在注射Clophen A50的各组记录到其诱导EROd活性的剂量及时间依赖关系,而这种引导的剂量和时间依赖关系被注射过羟基硫胺素强烈地抑制。在第103和182d℃对注射过羟基硫胺素的鲑鱼进行组织病理学检查发现:肝糖水平下降,肝细胞退化而且坏死的脑细胞更普遍的出现,这些都是在受M74综合症影响的具卵黄囊鱼苗中发现的病理特征。注射Clophen A50的各组鱼无此组织病理学变化。
