Molecular evaluation of glutathione S transferase family genes in patients with sporadic colorectal cancer

AIM To evaluate the association between polymorphismsin glutathione S transferases(GSTs) and the risk of sporadic colorectal cancer(SCRC), tumor progression and the survival of *** A case-control study of 970 individuals from the Brazilian population was conducted(232 individuals from the case group with colorectal cancer and 738 individuals from the control group without a history of cancer). PCR multiplex and PCR-RFLP techniques were used to genotype the GST polymorphisms. The tumors were categorized according to the TNM classification: tumor extension(T), affected lymph nodes(N), and presence of metastasis(M). Logistic regression, multiple logistic regression and survival analysis were used to analyze the data. The results are presented in terms of odds ratio(OR) and 95% confidence interval(CI). The level of significance was set at 5%(P ≤ 0.05).RESULTS Age equal to or over 62 years(OR = 8.79; 95%CI: 5.90-13.09, P ciated with increased risk of SCRC. Analysis of the polymorphisms revealed an association between the GSTM1 polymorphisms and a risk of SCRC(OR = 1.45; 95%CI: 1.06-2.00; P = 0.02), as well as between GSTT1 and a reduced risk of the disease(OR = 0.65; 95%CI: 0.43-0.98; P = 0.04). An interaction between the presence of the wild-type allele of GSTP1 Ile105 Val polymorphism and tobacco consumption on risk of SCRC(OR = 2.33; 95%CI: 1.34-4.05; P = 0.05) was observed. There was an association between the GSTM1 null genotype and the presence of advanced tumors(OR = 2.33; 95%CI: 1.23-4.41; P = 0.009), as well as increased risk of SCRC in the presence of a combination of GSTT1 non-null/GSTM1 null genotypes(OR = 1.50; 95%CI: 1.03-2.19; P = 0.03) and GSTT1 non-null/GSTM1 null/GSTP1 Val~*(OR = 1.85; 95%CI: 1.01-3.36, P = 0.04). AIM To investigate the contribution of polymorphisms in the CYP1A1, CYP2E1 and EPHX1 genes on sporadic colorectal cancer(SCRC) risk. METHODS Six hundred forty-one individuals(227 patients with SCRC and 400 controls) were enrolled in the study. The variables analyzed were age, gender, tobacco and alcohol consumption, and clinical and histopathological tumor parameters. The CYP1A1 *2A, CYP1A1 *2C CYP2E1 *5B and CYP2E1 *6 polymorphisms were analyzed by polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP). The EPHX1 Tyr113 His, EPHX1 His139 Arg and CYP1A1 *2C polymorphisms were detected by real-time PCR. Chisquared test and binary logistic regression were used in the statistical analysis. Among the impacts of climate change,there is the intensification of phenomena such as the El Niño South-ern Oscillation(ENSO)responsible for El Niño and La Niñ***,understanding their effects on the functional pro-cesses of forests is ***,this study evaluated the effects of ENSO on litter stock and water holding capac-ity(WHC)in a successional forest in eastern *** occurred in periods with the most rainfall in El Niño(2019)and least in La Niña(2021)*** permanent plots were used to sample *** effects were evident for WHC,higher during El Niñ***,this influence was not clear for litter,as only in the rainy season effects were *** was a positive correlation of WHC with precipitation and humidity,while litter stocks were negatively correlated with temperature and wind *** the subject of this study requires long-term assessments,preliminary results suggests that,depending on the intensity of ENSO,forest functional processes can be strongly impacted and *** conclusion reinforces warnings by the scientific community about the impacts of climate change on the maintenance of litter stocks,decomposition and,consequently,the biogeochemical cycle and essential ecosystem services for the maintenance of Amazonia *** need to develop long-term research to understand the effects of climatic change on litter stocks and water holding capacity is highlighted,especially in Amazonia.

The objective of this study was to establish a baseline for future studies that aim to determine possible contamination from human, agricultural and industrial activities. As well as the determination of the indices of environmental or geological contamination and enrichment factors of heavy metals Cr, Ni, Cu, Zn, As, Cd, Hg and Pb in agricultural soils of Constanza, Jarabacoa, Rancho Arriba and San José de Ocoa (SJO), municipalities located in the valleys of the Central mountain range of the Dominican Republic. The determination of the concentrations of heavy metals was carried out using the X-ray Fluorescence technique. Just like in Azua, San Juan de la Maguana (SJM) y Barahona in the southwest. Producer municipalities of vegetables, rice, beans, corn, melon, watermelon, tomato, banana, avocado, sugar cane and fodder for cattle. The concentration of 160 mg·kg-1 Probable Effect Level (PEL) of Cr according to the SQuiRTs table (USEPA-NOAA) for agricultural soils, were exceeded in 50% of the samples in SJM, SJO, Jarabacoa and Constanza;in Barahona and Azua by 20%. The PEL of 42.8 mg·kg-1 of the Ni was higher in more than 50% of the samples from SJM, Azua, Barahona and Jarabacoa;in SJO and Constanza at 35%. In the case of Cu with a PEL of 108 mg·kg-1 in SJO and Constanza, 5% of the samples exceeded its, in the other areas the concentrations were lower. Zn, As and Pb did not manage to exceed their respective PEL.

Mitochondrial disorders are phenotypically varied, with serious clinical repercussions. Among them, there is the deficiency of combined oxidative phosphorylation of type 20, which occurs due to a defect in the VARS2 gene. This article presents a case of a 2-year-old female with progressive myoclonic epilepsy and psychomotor regression, with refractoriness to multiple anticonvulsants. The diagnosis was only made after the examination was carried out. Therefore, this article highlights the aspects of this rare disease and the importance of the exome for the diagnosis of rare conditions.

Effect of Turbidity on Semi-Automatic Analysis of Copepod Size and Abundance Distribution in the Water Column

Automated image systems to characterize aquatic organisms improve research and enable fast response to environmental risk situations. In November 2015, a dam in Mariana City-MG (Brazil) collapsed and led to the disposal of mud tailings from the mining process to the Doce River. The accident resulted in several casualties and incalculable damage to surrounding communities and the environment. The mud increased water turbidity, an essential condition to the functioning of the image analysis systems, and directly affected the characterization of the organisms, making it impossible to distinguish copepods in the mud, due to the blurred outline. To get a quick response evaluating environmental situations, this work aimed to develop and test different algorithms characterizing and classifying copepods by their size (length and area) using in situ images acquired by the Lightframe On-Sight Keyspecies Investigation device. Field tests were carried out under different turbidity levels throughout the gradient observed in the coastal zone adjacent to the Doce River. The best algorithm reduced nearly 50% of the noise in some images when compared with manual treatment and led to 96% accuracy in measurement and counting. Semi-automated devices that perform post-processing corrections are suitable for fast environmental evaluation under high turbidity scenarios.
