Deep brain stimulation (DBS) has an increasing role in the treatment of idiopathic Parkinson’s disease. Although, the subthalamic nucleus (STn) is the commonly chosen target, a number of groups have reported that the most effective contact lies dorsal/dorsomedial to the STn (region of the pallidofugal fibres and the rostral zona incerta) or at the junction between the dorsal border of the STn and the latter. We analysed our outcome data from Parkinson’s disease patients treated with DBS between April 2002 and June 2004. During this period we moved our target from the STn to the region dorsomedial/medial to it and subsequently targeted the caudal part of the zona incerta nucleus (cZI). We present a comparison of the motor outcomes between these three groups of patients with optimal contacts within the STn (group 1), dorsomedial/medial to the STn (group 2) and in the cZI nucleus (group 3). Thirty five patients with Parkinson’s disease underwent MRI directed implantation of 64 DBS leads into the STn (17), dorsomedial/medial to STn (20) and cZI (27). The primary outcome measure was the contralateral Unified Parkinson’s Disease Rating Scale (UPDRS) motor score (off medication/off stimulation versus off medication/on stimulation) measured at follow-up (median time 6 months). The secondary outcome measures were the UPDRS III subscores of tremor, bradykinesia and rigidity. Dyskinesia score, L-dopa medication reduction and stimulation parameters were also recorded. The mean adjusted contralateral UPDRS III score with cZI stimulation was 3.1 (76%reduction) compared to 4.9 (61%reduction) in group 2 and 5.7 (55%reduction) in the STn (P-value for trend nt in tremor with cZI stimulation versus 86%in group 2 versus 61%in group 1 (P-value = 0.01). A novel 5-（（4-（6,7-dihydrothieno[3,2-c]pyridin-5（4H）-ylsulfonyl）phenylamino）methyl）quinolin-8-ol（HTPSMQol） was synthesized by optimized reaction of 4-（6,7-dihydrothieno[3,2-c]pyridin-5（4H）-ylsulfonyl）aniline with 5-chloromethyl-8-hydroxy-quinoline hydrochloride（CMHQ）.Also complexes of HTPSMQol were prepared by using various M（Ⅱ） metal *** compounds were analyzed by spectroscopic techniques and screened against various strains of *** results showed higher antimicrobial activity of HTPSMQol compared to its metal complexes and comparable with Ciprofloxacin.

Determination of asenapine in presence of its inactive metabolites in human plasma by LC-MS/MS

A highly selective and sensitive liquid chromatography-tandem mass spectrometry （LC-MS/MS） assay has been described for the determination of asenapine （ASE） in presence of its inactive metabolites n-desmethyl asenapine （DMA） and asenapine-n-glucuronide （ASG）. ASE, and ASE 13C-d3, used as internal standard （IS）, were extracted from 300 μL human plasma by a simple and precise liquid-liquid extraction procedure using methyl tert-butyl ether. Baseline separation of ASE from its inactive metabolites was achieved on Chromolith Performance RPse （100 mm ×4.6 mm） column using acetonitrile- 5.0 mM ammonium acetate-10% formic acid （90：10：0.1, v/v/v） within 4.5 min. Quantitation of ASE was done on a triple quadrupole mass spectrometer equipped with electrospray ionization in the positive mode. The protonated precursor to product ion transitions monitored for ASE and ASE 13C-d3 were m/z 286.1 → 166.0 and m/z 290.0 → 166.1, respectively. The limit of detection （LOD） and limit of quantitation （LOQ） of the method were 0.0025 ng/mL and 0.050 ng/mL respectively in a linear concentration range of 0.050-20.0 ng/mL for ASE. The intra-batch and inter-batch precision （% CV） and mean relative recovery across quality control levels were 〈 5.8% and 87.3%, respectively. Matrix effect, evaluated as IS-normalized matrix factor, ranged from 1.03 to 1.05. The stability of ASE under different storage conditions was ascertained in presence of the metabolites. The developed method is much simpler, matrix free, rapid and economical compared to the existing methods. The method was suc- cessfully used for a bioequivalence study of asenapine in healthy Indian subjects for the first time.

Highly sensitive LC–MS/MS method to estimate doxepin and its metabolite nordoxepin in human plasma for a bioequivalence study

A selective, sensitive and rugged liquid chromatography–tandem mass spectrometry(LC–MS/MS) assay has been developed for the simultaneous determination of doxepin(Dox) and its pharmacologically active metabolite, nordoxepin(nDox) in human plasma. The analytes and their internal standards(IS)were extracted from 500 m L of human plasma by liquid-liquid extraction using methyl tert-butyl *** separation was achieved on Hypurity C8 column(100 mm ? 4.6 mm, 5 mm) using a mixture of acetonitrile-methanol(95:5, v/v) and 2.0 mM ammonium formate in 93:7(v/v) ratio. Detection was accomplished by tandem mass spectrometry in the positive ionization and multiple reaction monitoring acquisition mode. The protonated precursor to product ion transitions studied for Dox, nDox,and their corresponding ISs, propranolol and desipramine, were m/z 280.1-107.0, 266.0-107.0,260.1-116.1 and 267.1-72.1, respectively. A linear dynamic range of 15.0–3900 pg/mL for Dox and 5.00–1300 pg/mL for nDox was established with mean correlation coefficient(r2) of 0.9991 and 0.9993, respectively. The extraction recovery ranged from 86.6%–90.4% and 88.0%–99.1% for Dox and nDox, respectively. The intra-batch and inter-batch precision(% CV) across quality control levels was r 8.3% for both the analytes. Stability evaluated under different storage conditions showed no evidence of degradation and the % change in stability samples compared to nominal concentration ranged from 4.7% to12.3%. The method was successfully applied to a bioequivalence study of 6 mg doxepin hydrochloride orally disintegrating tablet in 41 healthy Indian subjects under fasting and fed conditions.

The cellulosic bast fibers are recognized as a justifiable and biodegradable substitute for producing moderate strength polymer composite materials because of their characteristics of renewability,ecofriendliness,and higher specific *** the aim of this research work is to fabricate Himalayan bast fibers(nettle fiber(nF)/bauhinia vahlii fiber(BF)) based mono/hybrid epoxy composites at varying weight percentage of 2-6 wt% and evaluate the physical(void fraction and water absorption),mechanical(tensile strength,flexural strength,hardness) and sliding wear properties of as-fabricated *** 6 wt% nBF reinforced composites exhibited higher mechanical properties as compared to nF and BF composites with tensile strength of 34.04 MPa,flexural strength of 42.45 MPa,and hardness of 37.01 Hv *** influence of various control factors(sliding velocity,nF/BF/nBF contents,normal load and sliding distance) on specific sliding wear rate of composites was evaluated by Taguchi(three factors at three levels) experimental design and the percentage contribution of these selected parameters on sliding wear performance was examined by Analysis of variance(AnOVA).The sliding wear property of as-developed composites was found to be greatly influenced by sliding velocity and the wear resistance was observed to be improved with the nF/BF/nBF *** wear mechanism of the as-fabricated composites has been elucidated by scanning electron microscopy *** research outcomes demonstrated that the hybridization of Bauhinia vahlii fiber with nettle fiber led to improve the mechanical and wear properties of epoxy composites.

Surgical management in pediatric neuroblastoma diagnosis and treatment:a 20-year,single-center experience

Friction and wear characteristics of robust carbon–carbon composites developed solely from petroleum pitch without reimpregnation

Friction and wear characteristics correlating the fiber reinforcement percentage of carbon–carbon (C/C) composites solely developed from petroleum pitch matrices were investigated. This study exhibits the tribo-characteristics of C/C composites developed in a single-step carbonization process for varying loads for the first time without a reimpregnation process. A pin-on-disc tribometer with a sliding speed of 0.5 m/s and loads of 5, 10, and 20 n with a flat tool grade stainless steel pin as a static partner was employed. Further, polarized light optical and scanning electron microscopes (SEM) were utilized for a morphological analysis. Elastic modulus and strength were determined by a compression test. A result analysis is conducted to analyze sliding wear accompanied with minor abrasion. The composites with a high percentage of reinforcement exhibit credible wear resistance and mechanical robustness.
