Arabidopsis NPR1/NIM1 is a ke. re.ulator of the.SAR re.ponse. Studie. atte.pting to re.e.l the.function of NPR1 and how it me.iate. SA signaling have.le. to isolation of two classe. of prote.ns that inte.act with NPRI＂ the.first class include. rice.NRR, Arabidopsis NIMIN1, NIMIN2, and NIMIN3, and tobacco NIMIN2-1ike.prote.ns; the.se.ond class be.ongs to TGA transcription factors. We.have.pre.iously shown that ove.e.pre.sion of N RR in rice.suppre.se. both basal and Xa21-me.iate. re.istance. In orde. to te.t whe.he. NRR affe.ts SA-induce., NPRl-me.iate. SAR, we.have.transforme. Arabidopsis with the.rice.NRR ge.e.and te.te. its e.fe.ts on the.de.e.se.re.ponse. e.pre.sion of NRR in Arabidopsis re.ults in suppre.sion of PR ge.e.induction by SAR induce. and re.istance.to pathoge.s. The.e.phe.otype. are.e.e. more.se.e.e.than those.of the.nprl-1 mutant. The.ability of NRR to suppre.s PR ge.e.induction and dise.se.re.istance.is corre.ate. with its ability to bind to NPR1 be.ause.two point mutations in NRR, which re.uce.NPR1 binding, fail to suppre.s NPR1. In contrast, wild-type.and a mutant NRR, which still binds to NPR1 strongly, re.ain the.ability to suppre.s the.SAR re.ponse. Re.lacing the.C-te.minal 79 amino acids of NRR with the.VP16 activation domain turns the.fusion prote.n into a transcriptional co-activator. The.e.re.ults indicate.that NRR binds to NPR1 in vivo in a prote.n comple. to inhibit transcriptional activation of PR ge.e. and that NRR contains a transcription re.re.sion domain for active.re.re.sion.
