Comparison of Sensitivity and Specificity of ZN and Fluorescent Stain Microscopy with Culture as Gold Standard

Introduction: Reports indicate that fluorescent staining of smears increases sensitivity of direct microscopy;so ZN staining is being replaced with fluorescent microscopy in RNTCP in India. Chemical processing and sputum concentration may also improve sensitivity of microscopy. Objective: To compare the sensitivity and specificity of microscopy for AFB using ZN and fluorescent stains in direct and concentrated specimen with culture as gold standard. Methods: Morning sputum specimen of patients, suspected of having pulmonary tuberculosis, over a period of 6 months was subjected to direct microscopy using fluorescent stain;the same slide was over-stained with ZN stain. Same sputum sample was concentrated by Petroff’s method and subjected to fluorescent microscopy followed by ZN microscopy and finally to culture for AFB. Results: Sensitivity of fluorescent stained concentrated sputum samples was maximum and of ZN stained unprocessed sputum samples was minimum. Specificity of three of the methods was equal at 0.96 but of ZN stained concentrated sputum smears was 0.97. Sensitivity of total fluorescent stains was 0.85 (Specificity 0.96) and sensitivity of total ZN stained smears was 0.80 (Specificity 0.96). Discussion: We used same smear for fluorescent and ZN stains, so smear related variability is decreased. Blinding for microscopy was practically complete. Conclusion: The sensitivity of sputum microscopy for AFB can be increased by concentrating the sputum and using fluorescent microscopy. The specificity remains high in all the methods.

Primitive neuroectodermal tumor (PNET) of uterus is the rare of the rarest among the tumors of female genital tract. We herein reported a 32 years old female operated for lower abdominal lump,which was diagnosed,as PNET later confirmed by immunohistochemistry (IHC). She was lost to follow-up and presented 10 months later with recurrence. She was treated with cyclical combination chemotherapy followed by definitive surgery and is in complete remission at present.

Novel strategy to diagnose and grade hepatocellular carcinoma

Hepatocellular carcinoma (HCC) is among the most common malignancies worldwide, particularly in South and South East Asia. Unfortunately due to lack of appropriate facilities and awareness only limited information is available about its early diagnosis. Aim of the present study was to determine the efficacy of p53 by immunohistochemistry and Argyrophilicnucleolar organizer regions (AgNORs) in diagnosis of HCC and cirrhosis of liver. A total of 100 liver biopsies were studied, it included 20 cases of HCC, 60 cases of cirrhosis of the liver and 20 cases of normal liver from autopsy specimens as a control. Out of 20 cases of HCC, 15 were positive for p53 stain and 5 were negative. None of the 60 cases of cirrhosis or 20 with normal histology revealed p53 expression. A statistically significant (p . It helps in differentiation from well differentiated to moderately and to poorly differentiated HCC.

Background: Prospective analysis of 500 cases of vaginal discharge in tertiary health centre of Punjab. Objective: To study the prevalence of Sexually transmitted infections (STIs) in 500 outpatient of 15 - 49 years age group with vaginal discharge and to evaluate the various factors influencing pattern of STIs. Method: All cases were subjected to cervical cytology, HIV ELISA, rapid plasma reagen (RPR) test for syphilis and Gram’s staining, Giemsa staining and wet mount study of vaginal discharge. Results: Prevalence of STIs was found to be 58% and majority of infection was caused by G. vaginalis, Candida and Trichomonas. Pap smear showed koilocytosis in 5.6%, mild dysplasia in 1.8% and carcinoma cervix in 1.2% cases. Conclusion: Vaginal discharge is a common gynaecological complaint and STIs are significant cause of vaginal discharge.

“微生物-肠-脑轴”失常在抑郁症的发病中发挥着重要作用,调节肠道菌群失衡成为最新的治疗方向。海红米米糠可发挥益生元作用调控肠道微生物。为探究海红米米糠改善慢性不可预测温和应激(CUMS)诱导的小鼠抑郁样行为的作用。对照和暴露于CUMS的小鼠接受正常或含5%海红米米糠饮食喂养9周,通过糖水偏好实验和悬尾实验分析小鼠的抑郁样行为,实时荧光定量PCR法检测结肠内容物菌群的数量,HPLC测定前额皮层中单胺类神经递质的含量。应激使小鼠糖水偏好度、乳杆菌属和双歧杆菌属的数量、去甲肾上腺素(NE)与其代谢产物、五羟色胺(5-HT)与其代谢产物、多巴胺(DA)与其代谢产物的含量都显著下降,悬尾不动时间和梭菌属的数量都显著升高;海红米米糠干预后提高了小鼠的糖水偏好度、乳杆菌属和双歧杆菌属的数量、5-HT与其代谢产物和NE的含量,降低了悬尾不动时间。海红米米糠改善CUMS诱导的小鼠抑郁样行为可能与改善肠道菌群失衡及神经递质有关。

One hundred twenty-five endophytic microorganisms were isolated from the roots,stems,and leaves of four prominent rice cultivars growing in temperate *** potential to combat rice blast disease and promote plant growth was *** dual culture tests highlighted the strong antagonistic activity of five fungal(ranging from 89%–70%)and five bacterial(72%–61%)*** examination focused on volatile compounds produced by selected isolates to counter the blast *** these,the highest chitinase(13.76μg mL−1)and siderophore(56.64%),was exhibited by Aspergillus flavus,and the highest HCN production was shown by Stenotrophomonas maltophilia(36.15μM mL−1).In terms of growth promotion traits,Aspergillus flavus and Enterobacter cloacae excelled in activities viz,phosphorous solubilization,ammonia production,auxin and gibberellic acid production,and nitrogen *** Identity of these endophytes was confirmed through molecular analysis as Trichoderma afroharzianum,Trichoderma harzianum,Penicillium rubens,Aspergillus flavus,Stenotrophomonas rhizophila,Stenotrophomonas maltophilia,Bacillus cereus,Enterobacter cloacae,and Bacillus *** greenhouse conditions,the highest disease control was shown by isolate Bacillus licheniformis and *** with an inhibition of 79%,followed by ***(77%)and ***(73%).The overall results of this study showed that Bacillus licheniformis and Stenotrophomonas rhizophila have great potential to be used as bio-stimulant and biocontrol agents to manage rice blast disease.

Effect of Biochar Application on Soil Carbon Fluxes from Sequential Dry and Wet Cultivation Systems

Application of biochar has been highly credited for its potential to sequester carbon and GHG mitigation from tropical agro-ecosystems. However, experiments show inconsistent results depending on soil and biochar type, cultivation system, climatic condition and the type of evolved GHGs. This study emphasized on the effect of biochar on carbon emission trends from a sequential dry and wet cultivation system of Bangladesh. An incubation study was conducted with two contrasting soils and eight different treatments viz. control, only fertilizer, three different biochars (10 t·ha-1) with and without recommended fertilizer dose. Results revealed the fact that, emission of carbon was substantially higher from Sara soil than Kalma soil. Biochar treatments did not have any easing effect on CO2 emission at field condition;rather, increased in most of the cases. However, emission was significantly (P 2. Biochar application was ineffective to control CH4 and CO release to atmosphere and submergence further intensified their emission significantly. The overall results indicate that applied biochars have negligible effect on carbon emission except for reducing CO2 from submerged soils.

含手性配位体的铜氢化合物在非对称还原反应中是1种重要的催化剂。为了进一步确定该铜催化体系对α,β-不饱和酯化合物适用范围,筛选配体,优化反应条件,进行了环外烯烃的烯烃还原反应,结果表明含手性配位体的铜氢催化体系能令α,β-不饱和酯的环外碳碳双键非对称还原反应得到较高的产率,对映体的产率可提高至95%,对映体过量值可提高至89%。

Synthesis, Structural and Physical Properties of Cu1–xZnxFe2O4Ferrites

Zn substituted Cu-Zn ferrites with a composition Cu1-xZnxFe2O4 have been synthesized by standard double sintering ceramic method and characterized by X-ray diffraction. The single-phase cubic spinel structure of all the samples has been confirmed from X-ray diffraction analyses. The lattice constant is found to increase linearly with the manganese content obeying Vegard’s law. This increase in lattice parameter is explained in terms of the sizes of component ions. It is well known that density plays a key role in controlling the properties of polycrystalline ferrites. The X-ray and bulk densities of the Cu-Zn ferrite is significantly decreased whereas porosity increased with increasing Zn concentration, thereby giving an impression that zinc might be helping in the densification of the materials. SEM micrographs exhibit a decrease in grain size with increasing Zn content. The real part of initial permeability, μ′ increase with increasing Zn contents upto x = 0.5 after that it decreases with higher Zn content.
