Distribution of hepatitis B virus genotypes:Phylogenetic analysis and virological characteristics of Genotype C circulating among HBV carriers in kolkata,Eastern India

AIM: To evaluate the genotype distribution of hepatitis B virus (HBV) in Eastern India and to clarify the phyloge- netic origin and virological characteristics of the recently identifi ed genotype C in this region. METHODS: Genotype determination, T1762/A1764 mutation in the basal core promoter (BCP) and A1896 mutation in the precore region of 230 subjects were de- termined by restriction fragment length polymorphism method (RFLP) and the result was confi rmed by direct sequencing. RESULTS: The predominant genotypes D (HBV/D) and A (HBV/A) were detected in 131/230 (57%) and 57/230 (25%) samples. In addition, genotype C (HBV/C) was detected in 42/230 (18%) isolates. Surface gene region was sequenced from 45 isolates (27 HBV/C, 9 HBV/A and 9 HBV/D). Phylogenetic analysis revealed that all of the HBV/C sequences clustered with South East Asian subgenotype (HBV/Cs). The sequence data showed re- markable similarity with a Thai strain (AF068756) (99.5% ± 0.4% nucleotide identities) in 90% of the genotype C strains analyzed. T1762/A1764 mutation in BCP re- gion, associated with high ALT was signifi cantly higher in HBeAg negative isolates than HBeAg positive isolates. Frequency of A1896 mutation leading to HBeAg negativ- ity was ***: The present study reports the genotypic distribution and the characteristics of partial genome sequences of HBV/C isolates from Eastern India. Low genetic diversity and confi nement of HBV/C in Eastern India possibly indicate a recent, limited, spread in this region. Genotype C with T1762/A1764 mutation has been reported to increase the risk for hepatocellular car- cinoma; therefore genotype C carriers in Eastern India should be carefully monitored.

AIM: To develop a hepatocellular carcinoma (HCC) xenograft model for studying hepatitis C virus (HCV) replication in a mice, and antiviral ***: We developed a stable S3-green fluorescence protein (GFP) cell line that replicated the GFP-tagged HCV sub-genomic RNA derived from a highly efficient JFH1 virus. S3-GFP replicon cell line was injected subcutaneously into γ-irradiated SCID mice. We showed that the S3-GFP replicon cell line formed human HCC xenografts in SCID mice. Cells were isolated from subcutaneous tumors and then serially passaged multiple times in SCID mice by culturing in growth medium supplemented with G-418. The mouse-adapted S3-GFP replicon cells were implanted subcutaneously and also into the liver of SCID mice via intrasplenic infusion to study the replication of HCV in the HCC xenografts. The tumor model was validated for antiviral testing after intraperitoneal injection of interferon-α (IFN-α). RESULTS: A highly tumorigenic S3-GFP replicon cell line was developed that formed subcutaneous tumors within 2 wk and diffuse liver metastasis within 4 wk in SCID mice. Replication of HCV in the subcutaneous and liver tumors was confirmed by cell colony assay, detection of the viral RNA by ribonuclease protection assay and real-time quantitative reverse transcription polymerase chain reaction. High-level replication of HCV sub-genomic RNA in the tumor could be visualized by GFP expression using fluorescence microscopy. IFN-α cleared HCV RNA replication in the subcutaneous tumors within 2 wk and 4 wk in the liver tumor model. CONCLUSION: A non-infectious mouse model allows us to study replication of HCV in subcutaneous and metastatic liver tumors. Clearance of HCV by IFN-α supports use of this model to test other anti-HCV drugs.

Graphene derivatives as efficient hole transport materials for lead-free double perovskite（Cs2SnI6） solar cells: a numerical study

The double perovskite Cs2SnI6has notable optical and electrical characteristics, rendering it a highly prospective candidate for deployment as the absorber layer in perovskite solar cells （PSCs）. We simulated the performance of PSCs using lead-free Cs2SnI6double perovskite absorber layer and graphene derivatives, namely graphene oxide （GO） and reduced graphene oxide （rGO）, as hole transport layers （HTLs）. Our findings show that r GO offers an excellent hole extraction property with minimal interfacial recombination compared to GO.

Nonlinear convection in a non-Darcy porous medium

In this paper, the natural convection in a non-Darcy porous medium is studied using a temperature-concentration-dependent density relation. The effect of the two parameters responsible for the nonlinear convection is analyzed for different values of the inertial parameter, dispersion parameters, Rayleigh number, Lewis number, Soret number, and Dufour number. In the aiding buoyancy, the tangential velocity increases steeply with an increase in the nonlinear temperature parameter and the nonlinear concentration parameter when the inertial effect is zero. However, when the inertial effect is non-zero, the effect of the nonlinear temperature parameter and the nonlinear concentration parameter on the tangential velocity is marginal. The concentration distribution varies appreciably and spreads in different ranges for different values of the double dispersion parameters, the inertial effect parameter, and also for the parameters which control the nonlinear temperature and the nonlinear concentration. Heat and mass transfer varies extensively with an increase in the nonlinear temperature parameter and the nonlinear concentration parameter depending on Dacry and non-Darcy porous media. The variation in heat and mass transfer when all the effects, i.e., the inertial effect, double dispersion ef- fects, and Soret and Dufour effects, are simultaneously zero and non-zero. The combined effects of the nonlinear temperature parameter, the nonlinear concentration parameter and buoyancy are analyzed. The effect of the nonlinear temperature parameter and the nonlinear concentration parameter and also the cross diffusion effects on heat and mass transfer are observed to be more in Darcy porous media compared with those in non- Darcy porous media. In the opposing buoyancy, the effect of the temperature parameter is to increase the heat and mass transfer rate, whereas that of the concentration parameter is to decrease.

Synthesis and ion transport properties of hot-pressed solid polymer electrolytes （SPEs）, （l-x） PEO： x kI, where x is the content of kI in wt%, are reported. A hot-press technique has been used for the formation of the polymeric membranes in place of the usual solution cast method. The composition （80 PEO：20 kI） was identified as the highest conducting polymer electrolyte on the basis of compositional dependent conductivity studies of PEO：kI films. A conductivity enhancement of more than two orders of magnitude from that of the pure PEO was achieved. Materials characterization and ion transport mechanism were explained by using various experimental techniques.

The perforated stiffened panel is generally found as a sub-component of sophisticated *** fundamental purpose of this panel is to withstand against buckling under complicated loading and environmental ***,an accurate knowledge of critical buckling behaviour of stiffened panels is very much essential for a reliable and lightweight structural *** this paper,the focus is on quasi-laminated panels with different cutout shapes of various sizes and their responses to hygrothermal environments under nonlinearly varying edge loads and is compared with the locally stiffened *** this,the modelling of the panel and stiffener is done by adopting nine-noded heterosis plate elements and three noded beam elements *** stiffener formulation is suitably modified in order to take the torsional effect also into consideration along with the effect of shear ***,the plate and the stiffener elements are treated separately,and then the displacement compatibility is maintained between them by using the transformation *** a given loading and geometric discontinuity,the stress distribution within the perforated panel is highly non-uniform in nature and hence a dynamic approach has been used to calculate buckling loads by adopting two sets of boundary conditions,one set for pre-buckling stress analysis and the second set for buckling *** different quasi-isotropic stacking sequences are deliberated in this work by varying different ply-orientation in each *** study also addresses the effect of various parameters such as nonlinear loads,hygro-thermal loads,cutout size and shapes,position of cutout,stiffener parameters,stacking sequences,thickness of plate and boundary conditions.

Ethno-medicinal plants used by Bengali communities in Tripura, northeast India

Northeastern India has high medicinal plant diversity due to variance in topography and physiognomy. We documented the uses of various medicinal plants by the Bengali people of West district and South district of Tripura state for their own health care as well as for domesti-cated animals. Based on semi structured interviews, group discussions and information from local informants, a total of 93 species of medicinal plants of 52 families and 83 genera were documented. These plants were used to treat more than 55 different human diseases and 6 diseases of livestock. Sixty-eight plant species were used singly and the rest were used in combination with other species for therapeutic formulations of various diseases. Leaves of plants were most often used for most of the ethnobotanical preparations. Maximum consensus value of 96% was recorded for Chromolaena odorata （L.） king ＆amp;H. Rob., and the mini-mum was 15%for Bambusa balcooa Robx. Of the 93 plant species, 75 species showed pharmacological properties. Prospects for augmenting existing knowledge and enhancing the use of traditional medicinal plants are discussed.

Significant increase in HBV, HCV, HIV and syphilis infections among blood donors in West Bengal, Eastern India 2004-2005: Exploratory screening reveals high frequency of occult HBV infection

AIM: To evaluate the prevalence of markers of hepatitis B virus (HBV) and hepatitis C virus (HCV) and human immunodeficiency virus (HIV) among blood donors in kolkata, Eastern India for two consecutive years and to conduct a pilot study to explore the presence of HBV DNA among hepatitis B surface antigen (HBsAg) negative but anti-HBc positive blood donors. METHODS: Seroprevalence of HBsAg, anti-HCV and anti-HIV was studied among 113 051 and 106 695 voluntary blood donors screened in 2004 and 2005, respectively. Moreover, a pilot study on 1027 HBsAg negative donors was carried out for evaluating the presence of HBV DNA by PCR on HBsAg negative/anti- HBc positive donors. RESULTS: A statistically significant increase in the prevalence of HBV (1448 vs 1768, P kolkata West Bengal in 2005 as compared to 2004. Moreover, the exploratory study on 1027 HBsAg negative donors revealed that 188 (18.3%)of them were anti-HBc positive out of which 21% were positive for HBV DNA. CONCLUSION: The findings of this study underscore the significantly increasing endemicity of hepatitis viruses, syphilis and HIV among the voluntary blood donors of our community. The pilot study indicates a high rate of prevalence of HBV DNA among HBsAg negative/anti-HBc positive donors and thus emphasizes the need for a more sensitive and stringent screening algorithm for blood donations.

Structural,Wettability and Optical Investigation of Titanium Oxynitride Coatings:Effect of Various Sputtering Parameters

The objective of the present work is to investigate the effect of various sputtering parameters such as nitrogen flow rate, deposition time and sputtering pressure on structural, wettability and optical properties of titanium oxynitride films deposited on glass substrate by reactive magnetron sputtering. The X-ray diffraction graphs of titanium oxynitride films show evolution of various textures of TiO=N and TiN phases with increasing nitrogen flow rate and deposition time, but an increase in sputtering pressure from 4.0 to 8.0 Pa results in decline of various textures observed for TiO=Ny and TiN phases. The stress and strain calculated by sin2~ method are compressive, which decrease with increasing nitrogen flow rate from 55 to I00 sccm （standard cubic centimeter per minute） and increase with increasing deposition time from 80 to I40 min due to atomic penning effect and increasing thickness of the deposited films. The titanium oxynitride films have contact angle values above 90 deg., indicating that films are hydrophobic. The maximum contact angle of I09.1 deg. is observed at deposition time of 140 min. This water repellent property can add value to potential protective, wear and corrosion resistant application of titanium oxynitride films. The band gap decreases from 1.98 to 1.83 eV as nitrogen flow rate is increased from 55 to 100 sccm; it decreases from 1.93 to 1.79 eV as deposition time is increased from 80 to 140 min as more nitrogen incorporation results in higher negative potential of valence band N2p orbital. But it increases from 2.26 to 2.34 eV for titanium oxynitride films as sputtering pressure increases from 4.0 to 8.0 Pa.
