Influence of selenium induced oxidative stress on spermatogenesis and lactate dehydrogenase-X in mice testis

Aim: To evaluate the effect of oxidative stress on the spermatogenesis and lactate dehydrogenase-X (LDH-X) activity in mouse testis. Methods: For creating different levels of oxidative stress in mice, three selenium (se) level diets were fed in separate groups for 8 weeks. Group 1 animals were fed yeast-based se-deficient (0.02 ppm) diet. Group 2 and Group 3 animals were fed with the same diet supplemented with 0.2 ppm and 1 ppm se as sodium selenite, respectively. After 8 weeks, biochemical and histopathological observations of the testis were carried out. LDH-X levels in the testis were analyzed by western immunoblot and ELIsA. Results: A significant decrease in testis se level was observed in Group 1 animals, whereas it was enhanced in Group 3 as compared to Group 2. The glutathione peroxidase (GsH-Px) activity was significantly reduced in both the liver and testis in Group 1, but not in Group 2 and 3. A significant increase in the testis glutathione-s-transferase (GsT) activity was observed in Group 1, whereas no significant change was seen in Groups 2 and 3. Histological analysis of testis revealed a normal structure in Group 2. A significant decrease in the germ cell population in Group 1 was observed as compared to Group 2 with the spermatids and mature sperm affected the most. Decrease in the lumen size was also observed. In the se-excess group (Group 3), displacement of germ cell population was observed. Further, a decrease in the LDH-X level in testis was observed in Group 1. Conclusion: Excessive oxidative stress in the se deficient group, as indicated by changes in the GsH-Px/GsT activity, affects the spermatogenic process with a reduction in mature sperm and in turn the LDH-X level.

We reviewed the literature to examine whether an association exists between the quality of life (QoL) of primary informal Alzheimer's disease (AD) caregivers and the level and quality of care that these caregivers provide to their loved ones with AD. We obtained studies focusing on the care that these caregivers provide for their family members with AD. Our outcome of interest was level or quality of care and the independent variable was caregiver QoL. We extracted data in tabular form and used a narrative synthesis approach to describe our findings. Only one relevant study was included in the review. Overall, the evidence was equivocal regarding the associations between caregiver QoL and the level/quality of care in AD.

Many persons worldwide are deficient in micronutrients, two of the most common being vitamin A and iron. Fortification of staple food is a commonly used and effective method of boosting micronutrient intake. Wheat flour is widely used around the world and makes a good vehicle for fortification, but there is a lack of studies on the storage stability of fortified wheat flour. In this study, we fortified wheat flour with elevated levels of vitamin A and three sources of iron and stored it for up to 16 weeks at refrigerated or room temperature or elevated temperature. The rate of disappearance of vitamin A was monitored by high pressure liquid chromatography, and the rate of disappearance was fotmd to be directly proportional to the duration of storage and storage temperature. Higher temperatures contributed to greater loss of vitamin A. Iron, regardless of source, did not play a major role in contributing to the rate of vitamin A loss. Flour functionality was assessed by baking loaves of bread from the stored flour and assessing physical properties as well as subjecting the loaves to an expert sensory panel. Functionality also changed with time and temperature of storage, but sensory analysis found that these changes were not detrimental to the quality of bread baked from the stored flours. Trained sensory panelists found that breads baked from flours fortified with ferrous sulfate and stored at the three temperatures for times up to eight weeks were markedly different from fortified flours using hydrogen reduced irons.
