Transcriptome‑wide mapping of milk somatic cells upon subclinical mastitis infection in dairy cattle

Background Subclinical intramammary infection(IMI)represents a significant problem in maintaining dairy cows’health.Disease severity and extent depend on the interaction between the causative agent,environment,and host.To investigate the molecular mechanisms behind the host immune response,we used RNA-Seq for the milk somatic cells(SC)transcriptome profiling in healthy cows(n=9),and cows naturally affected by subclinical IMI from Proto-theca spp.(n=11)and Streptococcus agalactiae(S.agalactiae;n=11).Data Integration Analysis for Biomarker discov-ery using Latent Components(DIABLO)was used to integrate transcriptomic data and host phenotypic traits related to milk composition,SC composition,and udder health to identify hub variables for subclinical IMI detection.Results A total of 1,682 and 2,427 differentially expressed genes(DEGs)were identified when comparing Prototheca spp.and S.agalactiae to healthy animals,respectively.Pathogen-specific pathway analyses evidenced that Proto-theca’s infection upregulated antigen processing and lymphocyte proliferation pathways while S.agalactiae induced a reduction of energy-related pathways like the tricarboxylic acid cycle,and carbohydrate and lipid metabolism.The integrative analysis of commonly shared DEGs between the two pathogens(n=681)referred to the core-mastitis response genes,and phenotypic data evidenced a strong covariation between those genes and the flow cytometry immune cells(r2=0.72),followed by the udder health(r2=0.64)and milk quality parameters(r2=0.64).Variables with r≥0.90 were used to build a network in which the top 20 hub variables were identified with the Cytoscape cyto-hubba plug-in.The genes in common between DIABLO and cytohubba(n=10)were submitted to a ROC analysis which showed they had excellent predictive performances in terms of discriminating healthy and mastitis-affected animals(sensitivity>0.89,specificity>0.81,accuracy>0.87,and precision>0.69).Among these genes,CIITA could play a key role in regulating the animals’response to subclinical IMI

Background： Adaptive response includes a variety of physiological modifications to face changes in external or internal conditions and adapt to a new situation. The acute phase proteins（APPs） are reactants synthesized against environmental stimuli like stress, infection, inflammation.Methods： To delineate the differences in molecular constituents of adaptive response to the environment we performed the whole-blood transcriptome analysis in Italian Holstein（IH） and Italian Simmental（IS） breeds. For this, 663 IH and IS cows from six commercial farms were clustered according to the blood level of APPs. Ten extreme individuals（five APP＋ and APP-variants） from each farm were selected for the RNA-seq using the Illumina sequencing technology. Differentially expressed（DE） genes were analyzed using dynamic impact approach（DIA）and DAVID annotation clustering. Milk production data were statistically elaborated to assess the association of APP＋ and APP-gene expression patterns with variations in milk parameters.Results： The overall de novo assembly of cDNA sequence data generated 13,665 genes expressed in bovine blood cells. Comparative genomic analysis revealed 1,152 DE genes in the comparison of all APP＋ vs. all APP-variants; 531 and 217 DE genes specific for IH and IS comparison respectively. In all comparisons overexpressed genes were more represented than underexpressed ones. DAVID analysis revealed 369 DE genes across breeds, 173 and 73 DE genes in IH and IS comparison respectively. Among the most impacted pathways for both breeds were vitamin B6 metabolism, folate biosynthesis, nitrogen metabolism and linoleic acid metabolism.Conclusions： Both DIA and DAVID approaches produced a high number of significantly impacted genes and pathways with a narrow connection to adaptive response in cows with high level of blood APPs. A similar variation in gene expression and impacted pathways between APP＋ and APP-variants was found between two studied breeds. Such similarity was also confirmed by annotation clustering of the DE gene

Experimental evaluation of mechanical compression of lattice trusses made with Ti6Al4V for aerospace use

Lattice structures are three-dimensional structures composed of repeated geometrical shapes with multiple interconnected nodes,providing high strength-to-weight ratios,customizable properties,and efficient use of materials.A smart use of materials leads to reduced fuel consumption and lower operating costs,making them highly desirable for aircraft manufacturers.Furthermore,the customizable properties of lattice structures allow for tailoring to specific design requirements,leading to improved performance and safety for aircraft.These advantages make lattice structures an important focus for research and development in the aviation industry.This paper presents an experimental evaluation of the mechanical compression properties of lattice trusses made with Ti6Al4V,designed for use in an anti-ice system.The truss structures were manufactured using additive manufacturing techniques and tested under compressive loads to determine mechanical properties.Results showed that lattice trusses exhibited high levels of compressive strength,making them suitable for use in applications where mechanical resistance and durability are critical,such as in anti-ice systems.We also highlight the potential of additive manufacturing techniques for the fabrication of lattice trusses with tailored mechanical properties.The study provides valuable insights into the mechanical behavior of Ti6Al4V lattice trusses and their potential applications in anti-ice systems,as well as other areas where high strength-to-weight ratios are required.The results of this research contribute to the development of lightweight,efficient,and durable anti-ice systems for use in aviation and other industries.

Background Tuberous sclerosis complex(TSC)is a genetic disorder caused by inactivating mutations in the TSCl and TSC2 genes,causing overactivation of the mechanistic(previously referred to as mammalian)target of rapamycin(mTOR)signaling pathway in fetal life.The mTOR pathway plays a crucial role in several brain processes leading to TSC-related epilepsy,intellectual disability,and autism spectrum disorder(ASD).Pre-natal or early post-natal diagnosis of TSC is now possible in a growing number of pre-symptomatic infants.Data sources We searched PubMed for peer-reviewed publications published between January 2010 and April 2023 with the terms“tuberous sclerosis",“autism",or“autism spectrum disorder""animal models",“preclinical studies",“neurobiology",and“treatment".Results Prospective studies have highlighted that developmental trajectories in TSC infants who were later diagnosed with ASD already show motor,visual and social communication skills in the first year of life delays.Reliable genetic,cellular,electroencephalography and magnetic resonance imaging biomarkers can identify pre-symptomatic TSC infants at high risk for having autism and epilepsy.Conclusions Preventing epilepsy or improving therapy for seizures associated with prompt and tailored treatment strategies for autism in a sensitive developmental time window could have the potential to mitigate autistic symptoms in infants with TSC.

Photoacoustic dual-comb spectroscopy(DCS),converting spectral information in the optical frequency domain to the audio frequency domain via multi-heterodyne beating,enables background-free spectral measurements with high resolution and broad bandwidth.However,the detection sensitivity remains limited due to the low power of individual comb lines and the lack of broadband acoustic resonators.Here,we develop cavity-enhanced photoacoustic DCS,which overcomes these limitations by using a high-finesse optical cavity for the power amplification of dual-frequency combs and a broadband acoustic resonator with a flat-top frequency response.We demonstrate high-resolution spectroscopic measurements of trace amounts of C2H2,NH3 and CO in the entire telecommunications C-band.The method shows a minimum detection limit of 0.6 ppb C2H2 at the measurement time of 100 s,corresponding to the noise equivalent absorption coefficient of 7×10−10 cm−1.The proposed cavityenhanced photoacoustic DCS may open new avenues for ultrasensitive,high-resolution,and multi-species gas detection with widespread applications.

Aspirin interruption before neurosurgical interventions:A controversial problem

Aspirin is widely used for primary or secondary prevention of ischemic events.At the same time,chronic aspirin consumption can affect blood clot formation during surgical intervention and increase intraoperative blood loss.This is especially important for high-risk surgery,including neurosurgery.Current European Society of Cardiology guidelines recommend aspirin interruption for at least 7 d before neurosurgical intervention,but this suggestion is not supported by clinical evidence.This narrative review presents evidence that challenges the necessity for aspirin interruption in neurosurgical patients,describes options for aspirin effect monitoring and the clinical implication of these methods,and summarizes current clinical data on bleeding risk associated with chronic aspirin therapy in neurosurgical patients,including brain tumor surgery,cerebrovascular procedures,and spinal surgery.

Friction and wear performance of polyether ether ketone(PEEK)polymers in three lubrication regimes

This experimental study investigates the friction and wear of three coatings commonly used in industrial applications,particularly in hydrodynamic bearings.The three materials under investigation were Babbitt,polyether ether ketone(PEEK)reinforced with 15%carbon fibers,and PEEK reinforced with 20%carbon fibers.The first polymer material was extruded,while the other was produced by fused deposition modelling(FDM).The materials were subjected to sliding tests in a pin-on-disc configuration,with a steel ball serving as the counter surface.The tests were conducted at room temperature,with a load of 10 N and under three different lubrication conditions:dry,grease,and oil.The linear speed was set at 0.3 m/s for the dry and semi-solid lubrication tests,while for the oil tests,the speed was set at 0.25 m/s.The greases used had consistency grades of NGLI 000 and NGLI 2.An ISO VG 68 circulation oil was used for the oil lubrication tests.Additionally,thermodynamic analyses were performed under the most severe conditions(i.e.,dry)to investigate the steel-Babbitt and steel-PEEK contact.

Double-nylon purse-string suture technique: Another addition to the endoscopist's toolbox for full-thickness defect closure

Iatrogenic perforation is the most feared adverse event in endoscopy.With the expansion of interventional endoscopy in favor of traditional surgery,it is now more crucial than ever to develop effective defect closure techniques.This has culminated in the dissemination of multiple novel closure technologies,including through-the-scope clips,over-the-scope clips,through-the-scope suturing and over-the-scope suturing devices.In this editorial,we comment on the recent publication by Wang and colleagues discussing the performance of the double-nylon purse-string suture technique in the closure of large(>3 cm)gastric full-thickness defects.This technique offers a promising,practical and cost-effective approach to closure of large full-thickness defects that can be readily imple-mented across diverse healthcare settings.

In astrophysics, the boundary conditions for plasma phenomena are provided by nature and the astronomer faces the problem of understanding them from a variety of observations [Hester J J et al 1996 Astrophys. J. 456 225], on the other hand, in laboratory plasma experiments the electromagnetic boundary conditions become a major problem in the set-up of the machine that produces the plasma, an issue that has to be investigated step by step and to be modified and adapted with great patience, in particular in the case of an innovative plasma confinement experiment. The PROTO-SPHERA machine [Alladio F et al 2006 Nucl. Fusion 46 S613] is a magnetic confinement experiment, that emulates in the laboratory the jet + torus plasma configurations often observed in astrophysics: an inner magnetized jet of plasma centered on the(approximate) axis of symmetry and surrounded by a magnetized plasma torus orthogonal to this jet. The PROTO-SPHERA plasma is simply connected, i.e., no metal current conducting rod is linked to the plasma torus, while instead it is the inner magnetized plasma jet(in the following always called the plasma centerpost) that is linked to the torus. It is mandatory that no spurious plasma current path modifies the optimal shape of the plasma centerpost. Moreover, as the plasma torus is produced and sustained, in absence of any applied inductive electric field, by the inner plasma centerpost through magnetic reconnections [Taylor J B and Turner M F 1989 Nucl.Fusion 29 219], it is required as well that spurious current paths do not surround the torus on its outboard, in order not to lower the efficiency of the magnetic reconnections that maintain the plasma torus at the expense of the plasma centerpost. Boundary conditions have been corrected,up to the point that the first sustainment in steady state has been achieved for the combined plasma.

Hybrid robust fault detection and isolation of satellite reaction wheel actuators

In this paper,a combined robust fault detection and isolation scheme is studied for satellite system subject to actuator faults,external disturbances,and parametric uncertainties.The proposed methodology incorporates a residual generation module,including a bank of filters,into an intelligent residual evaluation module.First,residual filters are designed based on an improved nonlinear differential algebraic approach so that they are not affected by external disturbances.The residual evaluation module is developed based on the suggested series and parallel forms.Further,a new ensemble classification scheme defined as blended learning integrates heterogeneous classifiers to enhance the performance.A wide range of simulations is carried out in a high-fidelity satellite simulator subject to the constant and time-varying actuator faults in the presence of disturbances,manoeuvres,uncertainties,and noises.The obtained results demonstrate the effectiveness of the proposed robust fault detection and isolation method compared to the traditional nonlinear differential algebraic approach.
